Based on our original pyrazine hit, CP-0809101, novel conformationally-restricted 5HT2c receptor agonists with 2-piperazin-azaindane scaffold were designed. Synthesis and structure–activity relationship (SAR) studies are described with emphasis on optimization of the selectivity against 5HT2a and 5HT2b receptors with excellent 2c potency. Bartonellae were detected in a total of 152 (23.7%) of 642 tissues from 108 (48.4%) of 223 small mammals trapped in several urban areas of Nepal. Hybridization experiments with variants of an oligomeric protein often provide important information regarding subunit structure, function, and interactions. In some systems, however, the variants are so similar electrophoretically and chromatographically that purification of individual hybrids is not feasible. Therefore a method was developed for preparing hybrids by using 3,4,5,6-tetrahydrophthalic anhydride as a reversible acylating agent for protein amino groups. The technique involved acylating about 30% of the amino groups at pH 8 to give a derivative with a markedly altered net charge, formation of the hybrid set with unmodified and modified species, separation of the individual components by ion-exchange chromatography, and finally removal of the tetrahydrophthaloyl groups from the desired hybrid by incubation for about 1 day at pH 6 and room temperature. Experiments with model compounds and two enzymes showed that the anhydride was sepcific for amino groups. The extent of modification of proteins was measured by the spectral change at 250 nm, the loss of free amino groups, and the change in electrophoretic mobility of the polypeptide chains in polyacrylamide gels containing 8 M urea. Deacylation of modified, inactive aldolase and the catalytic subunit of aspartate transcarbamylase led to the restoration of the enzyme activity and electrophoretic mobility of the unmodified proteins. Both intra- and inter-subunit hybrids of aspartate transcarbamylase were prepared and isolated by using the tetrahydrophthaloyl groups as a reversible "chromatographic handle". Prior to deacylation the inter-subunit hybrid containing one acylated and one native catalytic subunit (and negative regulatory sub-units) exhibited no homotropic cooperativity and after deacylation the characteristic allosteric properties of the enzyme were regained. Similarly the ligand-promoted conformational changes associated with the allosteric transition were resotred upon deacylation of the intra-subunit hybrid containing one acylated and two native chains in each catalytic subunit. Criteria are described which must be satisfied if a reversible "chromatographic handle" is to be effective in hybridization experiments and it is shown that, despite some heterogeneity in its reaction with protein amino groups, 3,4,5,6-tetrahydrophthalic anhydride shows considerable promise for studies of oligomeric proteins. 相似文献

Summary Isolated trees and small patches of trees – paddock trees – are a prominent feature of agricultural landscapes in Australia, but are declining in many areas due to natural senescence, clearing, dieback and the general absence of recruitment. We assessed the importance of paddock trees for woodland conservation in a 30 000 ha sample of the New South Wales (NSW) South‐west Slopes using Satellite Pour l’Observation de la Terre (SPOT) panchromatic satellite imagery combined with models predicting the original distribution of vegetation communities. Tree‐cover occurred over 12% of the study area. The patch‐size distribution of vegetation in the study area varied between woodland types. For woodland communities that were confined to hills and ridges, most tree‐cover occurred as few, large remnants. For woodland communities of the foothills and plains (Blakely's Red Gum, Eucalyptus blakelyi and Yellow Box, Eucalyptus melliodora, or White Box, Eucalyptus albens and Red Stringybark), 54% of remnant tree‐cover occurred as patches < 1 ha. The loss of paddock trees will cause substantial reductions to some woodland communities. For example, the loss of patches < 1 ha in woodlands dominated by Blakely's Red Gum and Yellow Box would reduce this association from 7.4% to 3.4% of its predicted pre‐1750 distribution. Mean distance to tree‐cover across the study area increased almost fourfold if patches < 1 ha were removed from the landscape, which may have consequences for movements of some flora and fauna. Failure to protect and perpetuate paddock trees will diminish the likelihood of achieving the conservation objectives of comprehensiveness, adequacy and representativeness in agricultural landscapes. 相似文献

Human-associated bacteria dominate the built environment (BE). Following decontamination of floors, toilet seats, and soap dispensers in four public restrooms, in situ bacterial communities were characterized hourly, daily, and weekly to determine their successional ecology. The viability of cultivable bacteria, following the removal of dispersal agents (humans), was also assessed hourly. A late-successional community developed within 5 to 8 h on restroom floors and showed remarkable stability over weeks to months. Despite late-successional dominance by skin- and outdoor-associated bacteria, the most ubiquitous organisms were predominantly gut-associated taxa, which persisted following exclusion of humans. Staphylococcus represented the majority of the cultivable community, even after several hours of human exclusion. Methicillin-resistant Staphylococcus aureus (MRSA)-associated virulence genes were found on floors but were not present in assembled Staphylococcus pan-genomes. Viral abundances, which were predominantly enterophages, human papilloma virus, and herpesviruses, were significantly correlated with bacterial abundances and showed an unexpectedly low virus-to-bacterium ratio in surface-associated samples, suggesting that bacterial hosts are mostly dormant on BE surfaces. 相似文献
