This case study sought to examine how temporal variability in hydrological and hydraulic conditions might affect the feeding opportunities of juvenile Atlantic salmon in two hydrologically contrasting years of 2002 and 2003, which were characterised by high and low flows respectively. Firstly, measures of hydraulic influence were calculated to define what might be ecologically meaningful disturbance periods during high flows. Secondly, for identifying such periods, the parameter Critical Displacement Velocity (CDV) was derived from a river discharge time series as a first approximation of the amount of time in two hydrologically extreme years when fish foraging strategies for specific age classes of juvenile Atlantic salmon (Salmo salar) might be disrupted by flows. The CDV estimates the threshold velocity above which juvenile salmon are unable to hold station and it is dependent upon fish size and stream temperature. In the wet year 2002, the CDV was exceeded on 18% and 21% of days for 0+ and 1+ fish respectively. In 2003 these respective numbers fell to 6% and 15%. The data suggest that hydrological conditions during certain times of the year have the potential to affect foraging behaviour. This in turn might have implications for recruitment and growth rates for juvenile salmon in upland streams.  相似文献

To experimentally test the suggestion made in the preceding paper that the microtubules are involved in cell shape development during the formation and differentiation of the primary mesenchyme, we applied to the embryos two types of agents which affect cytoplasmic microtubules: (a) colchicine and hydrostatic pressure, which cause the microtubules to disassemble, and (b) D2O, which tends to stabilize them. When the first type of agent is applied to sea urchin gastrulae, the development of the primary mesenchyme ceases, the microtubules disappear, and the cells tend to spherulate. With D2O development also ceases, but the tubules appear "frozen," and the cell asymmetries persist unaltered. These agents appear to block development by primarily interfering with the sequential disassembly and/or reassembly of microtubules into new patterns. The microtubules, therefore, appear to be influential in the development of cell form. On the other hand through a careful analysis of the action of these agents and others on both intra- and extracellular factors, we concluded that the microtubules do rather little for the maintenance of cell shape in differentiated tissues.  相似文献

A rapid and efficient method for plasmid transformation of Klebsiella pneumoniae M5a1 and Escherichia coli K12 has been developed. The method, which uses a freeze-thaw cycle in the presence of CaCl2 to facilitate DNA uptake, is substantially more efficient for K. pneumoniae M5a1 than the conventional transformation procedure for E. coli. The simplicity and speed of the method makes it very attractive for routine transformation of K. pneumoniae M5a1 and E. coli K12.  相似文献

Steam treatment of peat at 200 degrees C for 3 min, followed by instantaneous decompression (steam explosion), solubilized up to 28% of the dry matter. Seventy-five percent of the solubilized material was carbohydrate, 33% of which was composed of mono- and disaccharides, including galactose, glucose, xylose, mannose, arabinose, and cellobiose, in order of decreasing concentration. The solubilized materials served as the sole source of carbohydrate for growth and solvent production by Clostridium acetobutylicum and C. butylicum which utilized up to 40% of the carbohydrate. Of the saccharides in this mixture, galactose was the least readily utilized. Approximately 30% of the fermentable carbohydrate used was converted to fatty acids and solvents, with the primary fermentation product being butyrate. Clostridium thermohydrosulfuricum was able to utilize ca. 50% of the carbohydrate, and simultaneously produced slightly more than 1 mol ethanol/mol saccharide metabolized. This organism, like other strains tested, used galactose less readily than the other sugars. The residue from the steam explosion process contained 24% cellulose, but it could not serve as a source of carbohydrate for the growth of either Bacteroides succinogenes or Clostridium thermocellum, suggesting that inhibitors were released during the steam treatment.  相似文献

Clostridium acetobutylicum ATCC 824 cells harvested from a phosphate-limited chemostat culture maintained at pH 4.5 had intracellular concentrations of acetate, butyrate, and butanol which were 13-, 7-, and 1.3-fold higher, respectively, than the corresponding extracellular concentrations. Cells from a culture grown at pH 6.5 had intracellular concentrations of acetate and butyrate which were only 2.2-fold higher than the respective external concentrations. The highest intracellular concentrations of these acids were attained at ca. pH 5.5. When cells were suspended in anaerobic citrate-phosphate buffer at pH 4.5, exogenous acetate and butyrate caused a concentration-dependent decrease in the intracellular pH, while butanol had relatively little effect until the external concentration reached 150 mM. Acetone had no effect at concentrations up to 200 mM. These data demonstrate that acetate and butyrate are concentrated within the cell under acidic conditions and thus tend to lower the intracellular pH. The high intracellular butyrate concentration presumably leads to induction of solvent production, thereby circumventing a decrease in the intracellular pH great enough to be deleterious to the cell.  相似文献

Summary The innervation of the major arteries and heart of the toad (Bufo marinus) was examined by use of glyoxylic acid-induced catecholamine fluorescence and peptide immunohistochemistry. All arteries possessed a moderate to dense plexus of adrenergic axons, which also showed neuropeptide Y-like immunoreactivity (NPY-LI). Some adrenergic axons in the intracardiac vagal trunks showed NPY-LI, but the varicose adrenergic axons innervating the cardiac muscle of the atria and ventricle, and the coronary blood vessels did not display NPY-LI. About half of the nerve cell bodies in the anterior sympathetic chain ganglia with dopamine--hydroxylase-LI (DBH-LI) also contained NPY-LI. The nerve cell bodies with DBH-LI alone were generally larger (median diameter 30 m) than those with both DBH-LI and NPY-LI (median diameter 20 m). Some cell bodies showing DBH-LI alone were surrounded by boutons with NPY-LI but not DBH-LI. Axons that displayed simultaneously both substance P-LI (SP-LI) and calcitonin gene-related peptide-LI (CGRP-LI) also formed a plexus around all arteries studied, being particularly dense around the mesenteric and pulmonary arteries. These axons are most likely sensory since SP-LI was reduced by capsaicin treatment, and nerve cell bodies with both SP-LI and CGRP-LI were found in dorsal root ganglia and the vagal ganglion. A dense plexus of axons showing somatostatin-LI was located around the pulmonary artery and its main intrapulmonary branches. A few nerves with vasoactive intestinal polypeptide-LI were found around the dorsal aorta and pulmonary artery. No perivascular nerves with enkephalin-LI were observed. Reversed-phase, high-pressure liquid chromatography of acid extracts of the large arteries showed that the major peaks of NPY-LI and SP-LI coeluted with porcine NPY (1–36) and synthetic SP (1–11), respectively. Thus, the location and structure of these peptides in perivascular nerves has been highly conserved during vertebrate evolution.  相似文献

Summary A primary rabbit kidney epithelial cell culture system has been developed which retains differentiated functions of the renal proximal tubule. In addition, the cells have a distinctive metabolism and spectrum of hormone responses. The primary cell were observed to retain in vitro a Na+-dependent sugar transport system (distinctive of the proximal segment of the nephron) and a Na+-dependent phosphate transport system. Both of these transport processes are localized on the apical membrane of proximal tubule cells in vivo. In addition, probenicid-sensitivep-aminohippurate (PAH) uptake was observed in basolateral membranes of the primary tubule cells, and the PAH uptake by these vesicles occurred at a rate that was very similar to that observed with membranes derived from the original tissue. Several other characteristics of the primary cells were examined, including hormone-sensitive cyclic AMP production and phosphoenolpyruvate carboxykinase (PEPCK) activity. Like the cells in vivo, the primary proximal tubule cells were observed to produce significant cyclic AMP in response to parathyroid hormone, but not in response to arginine vasopressin or salmon calcitonin. Significant PEPCK acivity was observed in the particulate fraction derived from a homogenate of primary rabbit kidney proximal tubule cells.
