The relationship between the mitogen‐activated protein kinase response, nuclear factor‐κB (NFκB) expression and the apoptosis in human acute promyelocytic leukaemia NB4 cells treated with vinblastine was investigated in this work. Cell viability, subdiploid DNA and cell cycle were analysed by propidium iodide permeability and flow cytometry analyses. Apoptosis was determined by annexin V‐Fluorescein isothiocyanate assays. Western‐blot analysis was used for determination of expression levels of apoptotic factors (p53, Bax and Bcl2), intracellular kinases [serine/threonine‐specific protein kinase, extracellular signal‐regulated kinase and c‐Jun N‐terminal kinase (JNK)], NFκB factor and caspases. Electrophoretic mobility shift assay was usefully applied to study DNA‐NFκB interaction. In NB4 cells, vinblastine produces alteration of p53 and DNA fragmentation. Vinblastine treatment had an antiproliferative effect via the induction of apoptosis producing Bax/Bcl‐2 imbalance. Vinblastine treatment suppressed NFκB expression and depressed NFκB‐DNA binding activity while maintaining JNK activation that subsequently resulted in apoptotic response through caspase‐dependent pathway. Our study provides a possible anti‐cancer mechanism of vinblastine action on NB4 cells by deregulation of the intracellular signalling cascade affecting to JNK activation and NFκB expression. The purpose of this study was to quantify the influence of inevitable ankle joint motion during an isometric contraction on the measured change of the gastrocnemius medialis muscle (GM) architecture in vivo during the loading and the unloading phase. Sitting on a dynamometer subjects performed isometric maximal voluntary contractions as well as contractions induced by electrostimulation. Synchronous joint angular motion, plantarflexion moment, foot’s centre of pressure and real-time ultrasonography of muscle architecture changes of the GM were obtained. During the contraction the ankle joint position altered and significantly affected the change in muscle architecture. At maximal tendon force (1094 ± 323 N), the measured fascicle length overestimated the change in fascicle length due to the tendon force by 1.53 cm, while the measured pennation angle overestimated the change in pennation angle due to the tendon force by 5.5°. At the same tendon force the measured fascicle length and pennation angle were significantly different between loading and unloading conditions. After correcting the values for the change in ankle joint angle no differences between the loading and the unloading phase at the same tendon force were found. Concerning the estimation of GM fascicle length–force and pennation angle–force curves during the loading and unloading phase of an isometric contraction, these findings indicate that not accounting for ankle joint motion will produce unreliable results.  相似文献

 Axial and apical flowers of Cryptantha capituliflora were analyzed with regard to morphology and pollen tube growth to assess the occurrence of cleistogamy. Although intermediate floral forms do occur, cleistogamous flowers were significantly smaller than chasmogamous flowers, had fewer anthers, and showed a distinctive stigmatic surface. Chasmogamous flowers can be cross-pollinated. Nevertheless, the growth of self-pollen tubes in few chasmogamous buds jointly with flower characters suggests that these flowers can probably produce fruits through autonomous selfing. The mean seed number per fruit did not differ between fruits from chasmogamous and cleistogamous flowers. Cleistogamous flowers were only observed in axial inflorescences, which are completely covered by the leaf. Other species of section Cryptantha also show the same trend, with cleistogamous flowers located in the lower half of the stems. This pattern is discussed in relation to dissimilarities in the outcrossing opportunities between flower types within the plant.   1. Urbanisation is one of the main drivers of insect species loss worldwide. However, its impacts on ecological interactions involving insects still deserve further research, especially seed predation and parasitism of seed predators.
  2. Here, we evaluated the seed predation rate by the specialist bruchid beetle Pseudopachymerina spinipes and its parasitism rate in the native tree Vachellia caven (Fabaceae) along an urbanisation gradient in Cordoba (Argentina). Since resource availability can influence these ecological interactions, we also investigated whether seed and prey availability could affect seed predation and parasitism rates, respectively.
  3. We sampled trees in 10 sampling sites along an urbanisation gradient estimated by the Normalised Difference Vegetation Index (NDVI) within a 100 m radius. In our system, sites with low NDVI, beyond representing the low amount of vegetation cover, also indicate high surface temperature and low availability of host trees.
  4. Seed predation in V. caven and the parasitism rate of P. spinipes were significantly reduced with increasing urbanisation. Notably, seed availability at the pod level did not affect seed predation rate, while prey availability was negatively correlated with parasitism rate.
  5. These findings suggest a deleterious effect of urbanisation on the studied antagonistic interactions, giving no support to the idea of resource limitation effects.

The olive is considered one of the most important fruit crops of the Mediterranean basin where it shows a wide range of variability, with about 2,000 cultivars. Italy, with about 500 cultivars, plays a fundamental role. The ability to discriminate olive cultivars and estimate genetic variability are important factors in better management of genetic resources and in helping to understand how genetic diversity is partitioned among cultivars. The two main objectives of the present investigation were to evaluate the identity of cultivars grown in Abruzzo region, central Italy, and to study their genetic structure. We applied amplified fragment length polymorphism (AFLP) methodology on 84 genotypes belonging to the most relevant and oldest varieties cultivated in Abruzzo and on six unknown genotypes. The information content of data was evaluated using the Marker Ratio index and the Polymorphic Index Content. Moreover, STRUCTURE software was used to investigate the genetic population structure. The analysis enabled us to clearly distinguish eight cultivars within seven clusters. Additionally, one cluster was found to have various minor cultivars and showed a relatively high level of diversity. The partitioning of genetic diversity showed that the largest amount of molecular variance was within groups. Our data suggest that both sexual and clonal propagation have played an important role in the evolution of olive cultivars. In our hypothesis, some ancestral population spread in central Italy with a relevant role of seed propagation, followed by a selection of superior clones from which more traditional varieties originated. In a few cases, hybridization should be taken into consideration to explain the diffusion of recently developed cultivars.  相似文献
