Thioglycine and l-thiovaline are stable under acidic and basic conditions but in the presence of bicarbonate they liberate the gasotransmitter H(2)S. In cells both thioamino acids were proven to enhance cGMP formation and promote vasorelaxation in mouse aortic rings. Given that H(2)S is known to lower arterial hypertension, reduce oxidative stress and exhibit cardioprotective effects in preclinical models, H(2)S donors hold promise as novel treatments for cardiovascular diseases. 相似文献

The tandem concept involves stacking two or more cells with complementary absorption spectra in series or parallel connection, harvesting photons at the highest possible potential. It is strongly suggested that the roll‐to‐roll production of organic solar cells will employ the tandem concept to enhance the power conversion efficiency (PCE). However, due to the undeveloped deposition techniques, the challenges in ink formulation as well as the lack of commercially available high performance active materials, roll‐to‐roll fabrication of highly efficient organic tandem solar cells currently presents a major challenge. The reported high PCE values from lab‐scale spin‐coated devices are, of course, representative, but not helpful for commercialization. Here, organic tandem solar cells with exceptionally high fill factors and PCE values of 7.66% (on glass) and 5.56% (on flexible substrate), which are the highest values for the solution‐processed tandem solar cells fabricated by a mass‐production compatible coating technique under ambient conditions, are demonstrated. To predict the highest possible performance of tandem solar cells, optical simulation based on experimentally feasible values is performed. A maximum PCE of 21% is theoretically achievable for an organic tandem solar cell based on the optimized bandgaps and achieved fill factors. 相似文献

During the search for inhibitors of N-acetylneuraminic acid biosynthesis, it was shown that 3-O-methyl-N-acetylglucosamine competitively inhibits the N-acetylglucosamine kinase of rat liver in vitro with a Ki value of 17 microM. N-Acetylmannosamine kinase is inhibited non-competitively with a Ki value of 80 microM. In a human hepatoma cell line (HepG2), 3-O-methyl-N-acetyl-D-glucosamine (1 mM) inhibits the incorporation of 14C-N-acetylglucosamine and 14C-N-acetylmannosamine into cellular glycoproteins by 88% and 70%, respectively. 相似文献

Synaptonemal complex (SC) karyotyping is used to analyse the relationship of two Palearctic hamsters. Male Phodopus sungorus have 13 SCs plus an X-Y pair while P. roborovskii has 16 SCs and an X-Y pair. P. sungorus has four nucleolar organizers as opposed to two in P. roborovskii. Centromeres of the X and Y are juxtaposed in the pairing region of the latter but not in the former. SC lengths and relative arm ratios are also discussed. Comparisons with karyotypes of other hamster genera indicate large intergeneric karyotypic differences and expectedly much smaller variation within each genus. Data on the two Phodopus species suggest that their presently accepted congeneric status warrants further examination. 相似文献

Summary The volumetric elastic modulus of the sheath and the osmotic swelling pressure of the axoplasmic polymer network of the giant axon ofLoligo vulgaris were measured. Evidence was obtained that (1) the elastic modulus of the sheath, (2) the swelling pressure of axoplasm, and (3) theeffective osmotic pressure difference due to mobile solutes determine axonal volume. The contributions of the sheath and the axoplasm were significant because theeffective osmotic pressure due to mobile solutes was a small fraction of thetheoretical bulk osmotic pressure due to these solutes. The giant axon was converted from an imperfect to a near perfect osmometer by minimizing the contribution of the sheath and the axoplasmic gel.The early experiments were undertaken in the Stazione Zoologica, Naples, and the Democritus Nuclear Research Center, Athens, Greece; the more recent experiments were undertaken in facilities provided by Prof. N. We describe here the cloning and characterization of a novel member of the thioredoxin family, thioredoxin-like protein 2 (Txl-2). The Txl-2 open reading frame codes for a protein of 330 amino acids consisting of two distinct domains: an N-terminal domain typical of thioredoxins and a C-terminal domain belonging to the nucleoside-diphosphate kinase family, separated by a small interface domain. The Txl-2 gene spans approximately 28 kb, is organized into 11 exons, and maps at locus 3q22.3-q23. A splicing variant lacking exon 5 (Delta 5Txl-2) has also been isolated. By quantitative real time PCR we demonstrate that Txl-2 mRNA is ubiquitously expressed, with testis and lung having the highest levels of expression. Unexpectedly, light and electron microscopy analyses show that the protein is associated with microtubular structures such as lung airway epithelium cilia and the manchette and axoneme of spermatids. Using in vitro translated proteins, we demonstrate that full-length Txl-2 weakly associates with microtubules. In contrast, Delta 5Txl-2 specifically binds with very high affinity brain microtubule preparations containing microtubule-binding proteins. Importantly, Delta 5Txl-2 also binds to pure microtubules, proving that it possesses intrinsic microtubule binding capability. Taken together, Delta 5Txl-2 is the first thioredoxin reported to bind microtubules and might therefore be a novel regulator of microtubule physiology. 相似文献

gpDB is a publicly accessible, relational database, containing information about G-proteins, G-protein coupled receptors (GPCRs) and effectors, as well as information concerning known interactions between these molecules. The sequences are classified according to a hierarchy of different classes, families and subfamilies based on literature search. The main innovation besides the classification of G-proteins, GPCRs and effectors is the relational model of the database, describing the known coupling specificity of GPCRs to their respective alpha subunits of G-proteins, and also the specific interaction between G-proteins and their effectors, a unique feature not available in any other database. Hypoxia-inducible factor-1 (HIF-1), consisting of two subunits, HIF-1alpha and HIF-1beta, is a key regulator for adaptation to low oxygen availability, i.e., hypoxia. Compared to the constitutively expressed HIF-1beta, HIF-1alpha is regulated by hypoxia but also under normoxia (21% O(2)) by several stimuli, including nitric oxide (NO). In this study, we present evidence that overexpression of mitochondrial-located thioredoxin 2 (Trx2) or thioredoxin reductase 2 (TrxR2) attenuated NO-evoked HIF-1alpha accumulation and transactivation of HIF-1 in HEK293 cells. In contrast, cytosolic-located thioredoxin 1 (Trx1) enhanced HIF-1alpha protein amount and activity under NO treatments. Taking into consideration that thioredoxins affect the synthesis of HIF-1alpha by altering Akt/mTOR signaling, we herein show that p42/44 mitogen-activated protein kinase and p70S6 kinase are involved. Moreover, intracellular ATP was increased in Trx1-overexpressing cells but reduced in cells overexpressing Trx2 or TrxR2, providing thus an understanding of how protein synthesis is regulated by thioredoxins. 相似文献

Circulating polypeptides and proteins have been implicated in reversing or accelerating aging phenotypes, including growth/differentiation factor 8 (GDF8), GDF11, eotaxin, and oxytocin. These proteoforms, which are defined as the protein products arising from a single gene due to alternative splicing and PTMs, have been challenging to study. Both GDF8 and GDF11 have known antagonists such as follistatin (FST), and WAP, Kazal, immunoglobulin, Kunitz, and NTR domain‐containing proteins 1 and 2 (WFIKKN1, WFIKKN2). We developed a novel multiplexed SRM assay using LC‐MS/MS to measure five proteins related to GDF8 and GDF11 signaling, and in addition, eotaxin, and oxytocin. Eighteen peptides consisting of 54 transitions were monitored and validated in pooled human plasma. In 24 adults, the mean (SD) concentrations (ng/mL) were as follows: GDF8 propeptide, 11.0 (2.4); GDF8 mature protein, 25.7 (8.0); GDF11 propeptide, 21.3 (10.9); GDF11 mature protein, 16.5 (12.4); FST, 29.8 (7.1); FST cleavage form FST303, 96.4 (69.2); WFIKKN1, 38.3 (8.3); WFIKKN2, 32.2 (10.5); oxytocin, 1.9 (0.9); and eotaxin, 2.3 (0.5). Actin and myosin are commonly involved in cell motility systems. This study attempts to identify these two proteins in the synaptonemal complex (SC), a nuclear organelle associated with chromosome pairing during meiotic prophase. The several experimental approaches reported here give no evidence of SC staining with either anti-myosin or anti-actin antibodies. These findings differ from those of other reportings. 相似文献
