In this study, a statistical classifier for Mediterranean taxa of Juniperus, based on 98 seed morphometric parameters, was tested at interspecific, specific and intraspecific levels. Ripe cones of 10 taxa were collected in different regions of the Mediterranean Basin to compare and discuss their taxonomic treatments according to two different sources. High percentages of correct identification were reached for both taxonomic treatments at the specific and intraspecific level and from the comparison among taxa of the J. oxycedrus, J. communis and J. phoenicea complexes. Moreover, ripe cones of J. macrocarpa were collected from four Sardinian populations, in two seasons, and from plant and soil, in order to analyse inter-population, seasonal and source variability in seed morphology. This statistical classifier discriminated J. macrocarpa seeds collected in spring more accurately than those collected in autumn, but it failed to distinguish between the seeds collected from plants and soil, or between those collected from different populations of the same geographic region.  相似文献

In a 5-period study (2007–2011) we examined the effects of human trampling on the last remaining population of Anchusa littorea on its sandy coastal habitat in Sardinia. This species, considered extinct in the wild for several years, was casually rediscovered in a small population at Is Arenas (SW Sardinia). We monthly monitored six trampled and six un-trampled permanent plots, mapping individuals and recording their size and reproductive variables. Trampled and un-trampled plots showed significant differences with respect to plant density, plant size and reproductive performance (flowers and fruits production) of A. littorea. This study demonstrated that human trampling represents a severe threat to the conservation of this species that can be appreciated as a keystone species with concern to human trampling effects on coastal dune plants. In case of A. littorea urgent measures should be undertaken to protect this unique remnant population in the dune system of Is Arenas. In particular, tourist paths should be redirected and confined to others areas in order to promote the natural expansion of A. littorea in its original habitat. A possible integrated strategy for the conservation and management of the species consists in a combination of in situ and ex situ measures.  相似文献

Thirty-four seed morphometric and colorimetric features of the five taxa belonging to the Lavatera triloba aggregate were measured using an image analysis system. Data were statistically analysed using a Linear Discriminant Analysis (LDA), which provided evidence to reassess the taxonomic position of some neglected or doubtful taxa. The results confirm the differentiation of the insular endemics L. agrigentina, L. triloba ssp. pallescens and L. triloba ssp. minoricensis. Moreover, the Sardinian populations of L. triloba ssp. triloba are clearly differentiated from their Iberian counterparts, but on the other hand, the seed morpho-colorimetric variation between L. triloba ssp. triloba and L. flava is not easy to interpret.  相似文献

The prognosis of locally advanced colorectal cancer (CRC) is currently unsatisfactory. This is mainly due to drug resistance, recurrence, and subsequent metastatic dissemination, which are sustained by the cancer stem cell (CSC) population. The main driver of the CSC gene expression program is Wnt signaling, and previous reports indicate that Wnt3a can activate p38 MAPK. Besides, p38 was shown to feed into the canonical Wnt/β-catenin pathway. Here we show that patient-derived locally advanced CRC stem cells (CRC-SCs) are characterized by increased expression of p38α and are “addicted” to its kinase activity. Of note, we found that stage III CRC patients with high p38α levels display reduced disease-free and progression-free survival. Extensive molecular analysis in patient-derived CRC-SC tumorspheres and APCMin/+ mice intestinal organoids revealed that p38α acts as a β-catenin chromatin-associated kinase required for the regulation of a signaling platform involved in tumor proliferation, metastatic dissemination, and chemoresistance in these CRC model systems. In particular, the p38α kinase inhibitor ralimetinib, which has already entered clinical trials, promoted sensitization of patient-derived CRC-SCs to chemotherapeutic agents commonly used for CRC treatment and showed a synthetic lethality effect when used in combination with the MEK1 inhibitor trametinib. Novel coronavirus disease 2019 (COVID-19) is associated with a hypercoagulable state, characterized by abnormal coagulation parameters and by increased incidence of cardiovascular complications. With this study, we aimed to investigate the activation state and the expression of transmembrane proteins in platelets of hospitalized COVID-19 patients. We investigated transmembrane proteins expression with a customized mass cytometry panel of 21 antibodies. Platelets of 8 hospitalized COVID-19 patients not requiring intensive care support and without pre-existing conditions were compared to platelets of healthy controls (11 donors) with and without in vitro stimulation with thrombin receptor-activating peptide (TRAP). Mass cytometry of non-stimulated platelets detected an increased surface expression of activation markers P-Selectin (0.67 vs. 1.87 median signal intensity for controls vs. patients, p = 0.0015) and LAMP-3 (CD63, 0.37 vs. 0.81, p = 0.0004), the GPIIb/IIIa complex (4.58 vs. 5.03, p < 0.0001) and other adhesion molecules involved in platelet activation and platelet–leukocyte interactions. Upon TRAP stimulation, mass cytometry detected a higher expression of P-selectin in COVID-19 samples compared to controls (p < 0.0001). However, we observed a significantly reduced capacity of COVID-19 platelets to increase the expression of activation markers LAMP-3 and P-Selectin upon stimulation with TRAP. We detected a hyperactivated phenotype in platelets during SARS-CoV-2 infection, consisting of highly expressed platelet activation markers, which might contribute to the hypercoagulopathy observed in COVID-19. In addition, several transmembrane proteins were more highly expressed compared to healthy controls. These findings support research projects investigating antithrombotic and antiplatelet treatment regimes in COVID-19 patients, and provide new insights on the phenotypical platelet expression during SARS-CoV-2 infection.Subject terms: Mechanisms of disease, Viral infection  相似文献

The multiplicity of systems affected in Alzheimer’s disease (AD) brains calls for multi-target therapies. Although mesenchymal stem cells (MSC) are promising candidates, their clinical application is limited because of risks related to their direct implantation in the host. This could be overcome by exploiting their paracrine action. We herein demonstrate that in vivo systemic administration of secretome collected from MSC exposed in vitro to AD mouse brain homogenates (MSC-CS), fully replicates the cell-mediated neuroreparative effects in APP/PS1 AD mice. We found a complete but transient memory recovery by 7 days, which vanished by 14 days, after a single MSC-CS intravenous administration in 12-month or 22–24-month-old mice. Treatment significantly reduced plaque load, microglia activation, and expression of cytokines in astrocytes in younger, but not aged, mice at 7 days. To optimize efficacy, we established a sustained treatment protocol in aged mice through intranasal route. Once-weekly intranasal administration of MSC-CS induced persistent memory recovery, with dramatic reduction of plaques surrounded by a lower density of β-amyloid oligomers. Gliosis and the phagocytic marker CD68 were decreased. We found a higher neuronal density in cortex and hippocampus, associated with a reduction in hippocampal shrinkage and a longer lifespan indicating healthier conditions of MSC-CS-treated compared to vehicle-treated APP/PS1 mice. Our data prove that MSC-CS displays a great multi-level therapeutic potential, and lay the foundation for identifying the therapeutic secretome bioreactors leading to the development of an efficacious multi-reparative cocktail drug, towards abrogating the need for MSC implantation and risks related to their direct use.Subject terms: Neural ageing, Neurological disorders, Inflammation  相似文献
