Irradiation of [Ce(hfac)3(diglyme)] (hfac = 1,1,1,5,5,5-hexafluoro-2,4-pentanedionato and diglyme (DG) = 2,5,8,11,14-pentaoxapentadecane) in chlorinated solvents (CH2Cl2, CCl4) with UV light led to luminescent colloidal CeCl3 that was characterized by transmission electron microscopy (TEM) analysis. When a substrate, quartz or silicon was present in the reaction cell, photoluminescent films were obtained, containing either pure CeCl3 or mixtures of CeCl3, CeF3 and CeOx in function of the experimental parameters of irradiation. Nanostructured and luminescent pure CeCl3 films were obtained by irradiation of the cerium complex in CCl4 at high intensity light for a few minutes. The films were characterized by X-ray diffraction (XRD), Energy dispersive X-ray (EDX), X-ray photoelectron spectroscopy (XPS), TEM, scanning electron microscopy (SEM) and atomic force microscopy (AFM). The kinetics of the [Ce(hfac)3(diglyme)] solution photodegradation, followed by UV spectrophotometry and spectrofluorimetry, pointed to CeCl3 formation by a solvent-initiated reaction, whereas the other inorganic compounds were the products of side reactions. 相似文献

Paramagnetic relaxation enhancement (PRE) measurements constitute a powerful approach for detecting both permanent and transient protein–protein interactions. Typical PRE experiments require an intrinsic or engineered paramagnetic site on one of the two interacting partners; while a second, diamagnetic binding partner is labeled with stable isotopes (15N or 13C). Multiple paramagnetic labeled centers or reversed labeling schemes are often necessary to obtain sufficient distance restraints to model protein–protein complexes, making this approach time consuming and expensive. Here, we show a new strategy that combines a modified pulse sequence (1HN-Γ2-CCLS) with an asymmetric labeling scheme to enable the detection of both intra- and inter-molecular PREs simultaneously using only one sample preparation. We applied this strategy to the non-covalent dimer of ubiquitin. Our method confirmed the previously identified binding interface for the transient di-ubiquitin complex, and at the same time, unveiled the internal structural dynamics rearrangements of ubiquitin upon interaction. In addition to reducing the cost of sample preparation and speed up PRE measurements, by detecting the intra-molecular PRE this new strategy will make it possible to measure and calibrate inter-molecular distances more accurately for both symmetric and asymmetric protein–protein complexes. 相似文献

We investigated the possible role of calmodulin (CaM) in the control of histamine release from human basophil leukocytes using several CaM antagonists. Trifluoperazine (TFP) (10(-6)-2 X 10(-5) M), pimozide (10(-6)-1.5 X 10(-5) M), chlorpromazine (CPZ) (10(-5)-10(-4) M) and promethazine (PMZ) (2 X 10(-5)-10(-4) M) inhibited in vitro histamine secretion from human basophils induced by several immunological (antigen, anti-IgE, and formyl-L-methionyl-L-leucyl-L-phenylalanine: f-met peptide) and nonimmunological (Ca2+ ionophore A23187 and the tumor promoter 12-0-tetradecanoyl-phorbol-13-acetate: TPA) stimuli. Trifluoperazine sulfoxide (TFP-S) and chlorpromazine sulfoxide (CPZ-S), which have very low affinity to CaM, had practically no inhibitory effect on histamine release from human basophils. The inhibitory effect of TFP could be made irreversible by irradiating the cells with UV light. A sulfonamide derivative, the compound N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide hydrochloride (W-7) (2.5 X 10(-5)-2 X 10(-4) M), which selectively binds to CaM, inhibited the release of histamine from basophils. In contrast, the chloride deficient analogue, W-5, which interacts only weakly with CaM, had practically no inhibiting effect. The IC50 for enzyme release by a series of eight CaM antagonists was closely correlated (r = 0.91; p less than 0.001) with the CaM specific binding, supporting the concept that these agents act by binding to CaM and thereby inhibiting histamine release. TFP and W-7 inhibited histamine release in the absence and in the presence of increasing concentrations of extracellular Ca2+.
