Mosquito‐borne infectious diseases are emerging in many regions of the world. Consequently, surveillance of mosquitoes and concomitant infectious agents is of great importance for prediction and prevention of mosquito‐borne infectious diseases. Currently, morphological identification of mosquitoes is the traditional procedure. However, sequencing of specified genes or standard genomic regions, DNA barcoding, has recently been suggested as a global standard for identification and classification of many different species. Our aim was to develop a genetic method to identify mosquitoes and to study their relationship. Mosquitoes were captured at collection sites in northern Sweden and identified morphologically before the cytochrome c oxidase subunit I (COI) gene sequences of 14 of the most common mosquito species were determined. The sequences obtained were then used for phylogenetic placement, for validation and benchmarking of phenetic classifications and finally to develop a hierarchical PCR‐based typing scheme based on single nucleotide polymorphism sites (SNPs) to enable rapid genetic identification, circumventing the need for morphological characterization. The results showed that exact phylogenetic relationships between mosquito taxa were preserved at shorter evolutionary distances, but at deeper levels, they could not be inferred with confidence using COI gene sequence data alone. Fourteen of the most common mosquito species in Sweden were identified by the SNP/PCR‐based typing scheme, demonstrating that genetic typing using SNPs of the COI gene is a useful method for identification of mosquitoes with potential for worldwide application. 相似文献

Bacillus thuringiensis Cry toxins are used in the control of insect pests. They are pore-forming toxins with a complex mechanism that involves the sequential interaction with receptors. They are produced as protoxins, which are activated by midgut proteases. Activated toxin binds to cadherin receptor, inducing an extra cleavage including helix α-1, facilitating the formation of a pre-pore oligomer. The toxin oligomer binds to secondary receptors such as aminopeptidase and inserts into lipid rafts forming pores and causing larval death. The primary threat to efficacy of Bt-toxins is the evolution of insect resistance. Engineered Cry1AMod toxins, devoid of helix α-1, could be used for the control of resistance in lepidopterans by bypassing the altered cadherin receptor, killing resistant insects affected in this receptor. Here we analyzed the mechanism of action of Cry1AbMod. We found that alkaline pH and the presence of membrane lipids facilitates the oligomerization of Cry1AbMod. In addition, tryptophan fluorescence emission spectra, ELISA binding to pure aminopeptidase receptor, calcein release assay and analysis of ionic-conductance in planar lipid bilayers, indicated that the secondary steps in mode of action that take place after interaction with cadherin receptor such as oligomerization, receptor binding and pore formation are similar in the Cry1AbMod and in the wild type Cry1Ab. Finally, the membrane-associated structure of Cry1AbMod oligomer was analyzed by electron crystallography showing that it forms a complex with a trimeric organization. 相似文献

The role of the frontal lobe in control of behavioral and cognitive abilities is explored in a group of 34 patients with brain lesions restricted to the prefrontal cortex. The scores in both structured behavioral questionnaires and standard neuropsychological tests were analyzed using the injured area of the frontal lobe as the independent variable. Our results show that patients with simultaneous lesions in supero- and inferomedial areas of the prefrontal cortex exhibit higher behavioral disturbances. Bilateral lesions also are associated with greater behavioral troubles. On the contrary, cognitive abilities are globally impaired in prefrontal patients. Results are discussed in relation to current models of the organization of the prefrontal cortex and its role on behavior control. 相似文献

A virulent strain of Photobacterium damselae subsp. piscicida (Pdp) was grown without (C form) or with (C+ form) glucose supplementation, the latter to enhance capsule formation. Both forms were resistant to killing by normal serum of seabream, red porgy and seabass. However, the C form was killed by immune serum of all three fish species while the C+ form was killed only by seabream and red porgy sera and to a lesser extent than the C form. Both C and C+ forms consumed complement in normal serum and this consumption was enhanced by precoating the bacteria in specific fish antibody. Complement consumption was greatest in seabass serum, especially with antibody-coated C+ form yet in this case the bacteria were not killed. The killing of the C form in immune serum of all three fish species was completely inhibited by EGTA/Mg(2+), indicating that the mechanism of complement activation leading to killing of the bacteria was by the classical pathway. The results suggest that immune serum killing by the classical complement pathway may provide some degree of protection against pasteurellosis, but enhanced expression of the capsule by Pdp in vivo may restrict complement-mediated killing, especially in immunised seabass. 相似文献

In Chilean evergreen temperate forest, fern species of the genus Blechnum occur in diverse microhabitats in a light gradient. We hypothesized that differences in the habitat preferences of three
co-occurring Blechnum species would be associated with differences in the magnitude of responses of light capture [chlorophyll (Chl) content] and
use (photosynthetic capacity and performance) to light availability. We measured the abundance, chlorophyll content, photosynthetic
capacity (A), and photosynthetic performance (chlorophyll fluorescence of photosystems I and II) of juvenile individuals of each species
growing in different light levels in the field. While Blechnum magellanicum covers a broad light environments range, B. mochaenum is restricted to shade, and B. penna-marina occupies full sun sites. Despite significant interspecific differences in average total chlorophyll content, this trait did
not differ among species along the light gradient. There was significant interspecific variation in both the mean value and
the plasticity of Chl a:Chl b ratio and A to light availability. While B. penna-marina showed a flatter reaction norm (lower response) of Chl a:Chl b ratio to light availability than its two congeners, B. mochaenum showed a lower response of A to light availability. B. penna-marina and B. magellanicum individuals from open sites had higher light saturation points of the electron transport rate (ETR) of both photosystems
(ETRLSP I and II) and photochemical quenching (qL and NA) than the shade restricted B. mochaenum. Additionally, non-photochemical quenching values for both photosystems (NPQ and ND) were higher in ferns species occurring
in shaded sites. The adjustment of the photosynthetic capacity and performance to light availability appears to be an important

Aggresomes are dynamic structures formed when the ubiquitin–proteasome system is overwhelmed with aggregation-prone proteins. In this process, small protein aggregates are actively transported towards the microtubule-organizing center. A functional role for autophagy in the clearance of aggresomes has also been proposed. In the present work we investigated the molecular mechanisms involved on aggresome formation in cultured rat cardiac myocytes exposed to glucose deprivation. Confocal microscopy showed that small aggregates of polyubiquitinated proteins were formed in cells exposed to glucose deprivation for 6 h. However, at longer times (18 h), aggregates formed large perinuclear inclusions (aggresomes) which colocalized with γ-tubulin (a microtubule-organizing center marker) and Hsp70. The microtubule disrupting agent vinblastine prevented the formation of these inclusions. Both small aggregates and aggresomes colocalized with autophagy markers such as GFP-LC3 and Rab24. Glucose deprivation stimulates reactive oxygen species (ROS) production and decreases intracellular glutathione levels. ROS inhibition by N-acetylcysteine or by the adenoviral overexpression of catalase or superoxide dismutase disrupted aggresome formation and autophagy induced by glucose deprivation. In conclusion, glucose deprivation induces oxidative stress which is associated with aggresome formation and activation of autophagy in cultured cardiac myocytes. 相似文献

Principal component analysis (PCA) is a method of simplifying complex datasets to generate a lower number of parameters, while retaining the essential differences and allowing objective comparison of large numbers of datasets. Glycosaminoglycans (GAGs) are a class of linear sulfated carbohydrates with diverse sequences and consequent complex conformation and structure. Here, PCA is applied to three problems in GAG research: (i) distinguishing origins of heparin preparations, (ii) structural analysis of heparin derivatives, and (iii) classification of chondroitin sulfates (CS). The results revealed the following. (i) PCA of heparin (13)C NMR spectra allowed their origins to be distinguished and structural differences were identified. (ii) Analysis of the information-rich (1)H and (13)C NMR spectra of a series of systematically modified heparin derivatives uncovered underlying properties. These included the presence of interactions between residues, providing evidence that a degree of degeneracy exists in linkage geometry and that a different degree of variability exists for the two types of glycosidic linkage. The relative sensitivity of each position (C or H nucleus) in the disaccharide repeating unit to changes in O-, N-sulfation and N-acetylation was also revealed. (iii) Analysis of the (1)H NMR and CD spectra of a series of CS samples from different origins allowed their structural classification and highlighted the power of employing complementary spectroscopic methods in concert with PCA. 相似文献

Mammals that build extensive open burrow systems are often classified as ecosystem engineers, since they have the potential
to modulate the availability of resources for themselves and other organisms. Lizards may benefit from the heterogeneity created
by these structures, especially if coupled with an increased offer of sites for refuge and thermoregulation. However, information
about these engineering effects by burrowing animals is scarce. We investigated the influence of European rabbit burrows on
several parameters of a Mediterranean lizard community (abundance, density, diversity and body condition) in three different
habitats (open pastures, holm oak and scrub patches). We found that lizards were positively associated with burrows, and that
burrows determined lizard presence at otherwise unfavourable habitats. Moreover, community parameters such as density and
species richness were higher in sites with burrows. Burrows influenced lizard species in different ways, and were also relevant
for other Mediterranean vertebrates, as revealed by questionnaires to experts. We also explored the possible resources provided
by burrows for lizards. Warrens offer relatively abundant prey and appropriate retreat sites for refuge and thermoregulation.
Warrens may have further implications within the ecosystem, acting as stepping stones, allowing lizards to reach otherwise
inaccessible habitat patches. This study shows that European rabbit warrens have a positive influence on lizard density and
diversity, and confirms the role of rabbits as ecosystem engineers. This reinforces the need for appropriate conservation
measures for rabbits, especially given their threatened status in the Iberian Peninsula. Furthermore, our study highlights
that taking into account the influence of engineering activities increases our awareness of species interactions, and may
translate into more adequate conservation measures for the preservation of biodiversity. 相似文献
