This paper reports a study of oxygen and redox conditions, trophic status, and phytoplankton community in the meromictic Lake Idro (Italy) from 2010 to 2014. The sequence of causes and effects of meromixis are also evaluated by comparing recent research with studies conducted from the late 1960s to the mid-1990s. In the last half century, Lake Idro was steadily meromictic due to solutes which accumulated in its deep waters, along with both dissolved nutrients and chemically reduced substances produced by the anaerobic microbial metabolism. These substances were retained in bottom waters and made unavailable to upper layers until stratification broke. Mixing episodes occurred in 2005–2006 altering stratification, and oxygen and nutrient distribution within the lake. The potential full overturn effects were also evaluated as potential oxygen consumption due to the oxidation of reduced substances to forecast possible oxygen exhaustion and collapse of biological communities. Finally, meromixis is discussed as a potential threat for deep perialpine lakes using Lake Idro as a reference to comparatively evaluate the present status and possible future trends.  相似文献

Extreme climatic events, such as heatwaves and droughts, are occurring more frequently in many regions of the world. Lakes may be especially vulnerable to climatic perturbations, which can trigger sudden ecosystem changes through alterations in the hydrologic regime. However, the nature of lake response to climatic extremes, and associated long-term ecosystem-level implications are difficult to predict, due to the paucity of time series allowing exploration of ecosystem behavior before, during, and after extreme events. We investigated the impacts of the 2003 European heatwave on a small, stratifying lake by analyzing available limnological data between 1986 and 2012. In summer 2003, a shift from an unvegetated to a macrophyte-dominated regime occurred, due to the rapid spread of a benthic charophyte. We explored candidate mechanisms driving the shift by comparing empirical observations with the outcome of a model on lake alternative states parameterized for our study lake. Our results support the hypothesis that enhanced light availability due to a heatwave-induced decrease in water level drove the switch in dominant primary producers. The spread of the charophyte was associated with strong depletion of inorganic nutrients and suppression of the typical summer phytoplankton peak. These bottom-up interactions triggered cascading effects at higher trophic levels, inducing a decline in herbivorous zooplankters with high food requirements and in predatory taxa. Some of the changes in the lake food web persist through the available time series. If incidence of heatwaves increases, as projected across temperate regions, our findings suggest that abrupt and long-lasting ecosystem-level reorganizations may occur in small, stratifying lakes.  相似文献

Idiopathic pleuroparenchymal fibroelastosis (IPPFE) is a recently described clinical-pathologic entity characterized by pleural and subpleural parenchymal fibrosis, mainly in the upper lobes. As this disease is extremely rare (only 7 cases have been described in the literature to date) poorly defined cases of IPPFE can go unrecognized. The clinical course of disease is progressive and prognosis is poor, with no therapeutic options other than lung transplantation currently available, yet. Environmental stress imposes strong natural selection on clonal populations, promoting evolutionary change in clonal structure. Environmental stress may also lead to reduction in population size, which together with clonal selection may reduce genotypic diversity of the local populations. We examined how clonal structure in wild-collected samples of two parthenogenetic populations of the freshwater ostracod Eucypris virens responded to hypersalinity and starvation, and the combination of the two stressors. We applied the stress treatments in a factorial design for one generation. When 60% of the individuals per experimental unit had died, post-experimental clonal structure was compared to that of the start of the experiment, which reflected the field conditions. We used five polymorphic allozyme loci as genotype markers. All stress treatments reduced survival compared to the control treatment. In the population “Rivalazzetto”, we observed a reduction of clonal richness in the control treatment, with the initially dominant clone maintaining dominance. This may have resulted from interclonal competition and clone-specific survival under the different laboratory conditions. Clonal richness remained high in the salinity treatment while it was reduced in the combined stress and starvation treatments. In the population “Fornovo”, clonal richness reduced in all treatments including control, while the salinity and combined stress treatment reduced clonal evenness. The clone dominating at the start of the experiment increased in frequency in all treatments, but the change in clonal structure during the experiment was more pronounced in this population. These results suggest that in some conditions an intermediate level of environmental stress may lessen the decline in genetic diversity by strong inter-clonal competition. Moreover, the variation in clonal structure among the stress treatments and distinct genetic backgrounds indicates that more general predictions of stress effects on clonal structure may be difficult.  相似文献

Binding sites recognition is generally based on geometry often combined with physico-chemical properties of the site since the conformation, size and chemical composition of the protein surface are all relevant for the interaction with a specific ligand. Recent studies have suggested that Rap1 and Rap2 small GTP-binding proteins are both expressed in human red blood cells (RBCs). In this work, we carefully examined the expression of Rap proteins in leukocytes- and platelets-depleted RBCs, whose purity was established on the basis of the selective expression of the beta2 subunit of the Na+/K+ -ATPase, as verified according to the recently proposed "beta-profiling test" [J.F. Hoffman, A. Wickrema, O. Potapova, M. Milanick, D.R. Yingst, Na pump isoforms in human erythroid progenitor cells and mature erythrocytes, Proc. Natl. Acad. Sci. U. S. A. 99 (2002) 14572-14577]. In pure RBCs preparations, Rap2, but not Rap1 was detected immunologically. RT-PCR analysis of mRNA extracted from highly purified reticulocytes confirmed the expression of Rap2b, but not Rap2a, Rap2c, Rap1a or Rap1b. In RBCs, Rap2 was membrane-associated and was rapidly activated upon treatment with Ca2+/Ca2+ -ionophore. In addition, Rap2 segregated and was selectively enriched into microvesicles released by Ca2+ -activated RBCs, suggesting a possible role for this GTPase in membrane shedding.  相似文献

Valproic acid (VPA), a histone deacetylase inhibitor, causes differentiation in different cell lines and in a cell-specific manner; yet, its effect on megakaryocytic (MK) differentiation has not been studied. We evaluated whether VPA induces MK differentiation in a UT-7 cell line through histone acetylation in the GpIIIa gene region and activation of the ERK pathway. UT-7 cells, derived from megakaryoblastic leukemia, were treated with VPA at various concentrations, and the expression of differentiation markers as well as the gene expression profile was assessed. Flow cytometry, immunoblot analysis, and RT-PCR demonstrated that VPA induced the expression of the early MK markers GpIIIa (CD61) and GpIIb/IIIa (CD41) in a dose-dependent manner. The VPA-treated cells showed hyperacetylation of the histones H3 and H4; in particular, histone acetylation was found to have been associated with CD61 expression, in that the GpIIIa promoter showed H4 hyperacetylation, as demonstrated by the chromatin immunoprecipitation assay. Furthermore, activation of the ERK pathway was involved in VPA-mediated CD61/CD41 expression and in cell adhesion, as demonstrated by using the MEK/ERK inhibitor U0126. In conclusion, the capacity of VPA to commit UT-7 cells to MK differentiation is mediated by its inhibitory action on HDAC and the long-lived activation of ERK1/2.  相似文献

Over the past fifteen years of research in the alpine area, the Dalmeri Rockshelter, located on the northeastern edge of the Sette Comuni Plateau (Trento, Italy), has become a key site for reconstructing the exploitation of the mountain territories by the human groups of the end of the Upper Palaeolithic. Excavations, carried out between 1991 and 2005, brought to light a sequence of Recent Epigravettian anthropic levels, which were radiocarbon dated to the Allerød interstadial. The well-preserved paleosurfaces provided the opportunity to carry out a thorough analysis on the faunal and lithic remains, enabling us to reconstruct the palaeoeconomy, the function and the internal organisation of the site. In the past five years, the discovery of a notable quantity of red ochre painted stones, which are linked to the earliest settlement phase, revealed different representations, both naturalistic paintings as well as signs. Following this discovery, the interpretation of the site began to touch the artistic-religious sphere, providing new insights into Epigravettian art.  相似文献

Structural aberrations, their frequency and distribution as well as distribution of the tandem repetitive minisatellite DNA clusters of Alu and Hinf elements and two retroelements, the LINE NLRCth1 and the SINE CTRT1, were analyzed in the genome of the chironomid C. piger Strenzke larvae from a Bulgarian population. A consistent somatic variability in the structure of the polytene chromosomes was detected, showing that the C. piger genome is more actively rearranging than supposed before. Breakpoints were concentrated in proximal parts of chromosomes significantly more often than in distal parts. By FISH analysis we could detect only one locus containing Alu elements and 38 Hinf cluster loci which appear to be dispersed equally all over the chromosomes. The retrotransposons NLRCth1 and CTRT1 are present only in a few loci, but highly variant among different individuals. The mean number of NLRCth1 sites per individual was 18.4 ± 2.09 and of CTRT1 was 54.8 ± 8.42. A third of breakpoint locations were close to or coincide with a locus occupied by a retroelement (either NLRCth1 or CTRT1). Nineteen percent of breakpoints coincided with Hinf repetitive DNA elements. Some breakpoints were identical in the two sibling species C. piger and C. riparius Meigen (syn.: C. thummi thummi) and are considered as conserved hot spots of chromosome breakage.  相似文献
