为研究玉米(Zeamays L.)19kD醇溶贮藏蛋白(zein)基因启动子种子特异性表达的控制区段,将全长694bp的启动子进行5’端缺失,共得到6个缺失突变体,长度分别为488bp、378bp、302bp、152bp、124bp和85bp。将6个片段分别与报告基因gus连接构建成表达载体pDGB系列,经土壤农杆菌(Agrobacterium)介导转化,引入烟草。GUS活性检测证明,488bp启动子片段能促使gus基因在种子中特异表达。378bp、302bp、152bp和124bp片段启动子引导的gus基因在烟草根、叶柄、种子中均可表达。  相似文献

目的:将gfp基因克隆到pGEM3Z-f( )载体。方法:将PCR扩增得到的gfp基因插入到pGEM3Z-f( )质粒的SmaⅠ位点,构建重组质粒转化大肠杆菌DH5α。结果:含重组质粒的大肠杆菌在自然光下呈现黄绿色,而在涂X-gal的培养基上它不仅发出黄绿色荧光,而且还出现蓝色。结论:gfp基因的插入没有引起载体上lacZ基因的失活,而且形成的gfp-lacZ融合基因在大肠杆菌得到了表达。表达产物不仅具有GFP活性,而且保持了β-半乳糖苷酶的生物学活性。  相似文献

Summary Airborne pollens were sampled in L'Aquila (Italy) throughout 1988 to 1990. Our data indicate that the flowering seasons of pollens mostly accounting for airborne concentrations are somewhat unique as compared to other Mediterranean areas. The prolonged season of Cupressaceae as well as the two different flowering seasons of Oleaceae, namely ofFraxinus, Ligustrum andOlea europaea, respectively, were the most relevant findings of our study. Furthermore, it is also worth to note thatParietaria judaica could be detected during the study period throughout March to June, but not in late summer and early autumn, as expected.Moreover, we evaluated the frequency of different pollen sensitizations in 2813 patients requiring medical care during the study period because of allergic diseases, such as conjunctivitis, oculorhinitis, and bronchial asthma. Our data demonstrate that both Compositae and Urticaceae proved to be the most allergenic pollens despite the low airborne concentrations detected. Finally, a relevant finding was the highOlea sensitization among patients with bronchial asthma.  相似文献

Rat cerebral microvascular smooth muscle cells in culture   总被引:1,自引:0,他引:1  
This report describes the development and establishment of long-term serial cultures of adult rat vascular smooth muscle cells (SMC) derived from cerebrocortical resistance vessels (small arteries and arterioles). Electron microscopic examination of microvessels isolated off a 150 microns nylon mesh sieve clearly demonstrated the predominance of these vessel types. Initial outgrowth from collagenase-elastase-treated microvessel fragments yielded both endothelium and smooth muscle cells. However, at confluency (2-3 weeks) these cultures consisted of a homogeneous population of broad, polygonal cells that grew in a multilayered "hill and valley" pattern typical of SMC in vitro. For comparative morphological and functional studies, SMC cultures were also initiated from rat thoracic aortas utilizing ring segments as explants. The smooth muscle origin of cultures derived from both resistance vessel (RV) and aorta (RA) was further demonstrated by positive immunofluorescent staining by the specific smooth muscle alpha-actin and myosin antibodies. Ultrastructural examination of these SMC cultures revealed similar morphologic features consisting of typical cytoplasmic myofilament bundles with associated dense bodies and numerous pinocytotic vesicles. Cell growth studies on early (less than P 15)- and late (greater than P 15)-passage RV- and RA-SMC populations revealed markedly different cell growth responses. Representative growth curves of early- and late-passage RA-SMC showed a significantly higher growth rate (two- to fourfold) than RV-SMC cultures. Both cultures, however, exhibited a marked increase in growth potential at higher passage levels. Heparin, at a concentration of 100 micrograms/ml inhibited the growth of RV-SMC during the first 3 days after addition in both exponential and growth-arrested culture states, whereas RA-SMC cultures showed no inhibitory response. These studies indicate that long-term RV-SMC cultures can serve as a useful model system to study functional and metabolic properties of this cell type and provide the means to explore further the heterogeneity of SMC derived from different vasculatures in normal as well as various disease states.  相似文献

The aim of this work is to study the binding of nickel ions to hexahistidine (His(6)) combining potentiometric titrations and spectroscopic (UV-Vis and circular dichroism) determinations in order to establish the species distribution as a function of the pH, their stoichiometry, stability and geometry. For comparative purposes, the same procedure was applied to the Ni-histidine (His) system. His behaves as a tridentate ligand, coordinating the carboxyl group, the imidazole and the amino nitrogen atoms to Ni(II) ions in an octahedral coordination and a bis(histidine) complex is formed at pH higher than 5. For the Ni-His(6) system, the complex formation starts at pH 4 and five different species (Ni(His(6))H, Ni(His(6)), Ni(n)(His(6))(n), Ni(n)(His(6))(n)H(-n/2), Ni(n)(His(6))(n)H(-n)) are formed as a function of the pH. Ni(His(6))H involves the coordination of the imidazole nitrogen and a deprotonated amide nitrogen (N(Im), N(-)) resulting in an octahedral geometry. In Ni(His(6)), an imidazole nitrogen is deprotonated and coordinated (2N(Im), N(-)) to the metal ion with a square planar geometry. The aggregated forms result from the extra Ni-N(Im) coordination, resulting in a 4N square planar geometry that is stabilized by inter/intramolecular hydrogen bonds. This coordination mode is not altered during the deprotonation steps from Ni(n)(His(6))(n).  相似文献

香根草(Vetiveria zizanioides)这一优良的水土保持和多用途植物目前在热带和亚热带地区广泛推广应用。中国的香根草栽培种主要是在50年代从国外引进的,但在广东和海南也有这一植物的野生种存在,尤其在广东的吴川有一块面积达6670 hm2左右的野生香根草群落。然而,由于人类活动的影响和过度开发利用,40年间,这片热带草原已遭到严重破坏,面积和蕴藏量都大幅度减少。为了不使这片珍贵的湿地消失殆尽,必须尽快采取措施加以保护。另外,香根草属的系统分类还很不完善,上述在中国分布的野生居群究竟是香根草还是野香根草(V. nigritana),迄今还没有定论,经对标本的研究鉴定,初步确定为香根草(Vetiveria zizanioides)。  相似文献

土壤微生物释放的胞外酶是决定碳(C)、氮(N)、磷(P)生物地球化学循环的关键因素,为了阐明青藏高原典型小流域土壤微生物生物量和胞外酶活性沿海拔和土层的分布特征并揭示影响该分布格局的主要养分限制状况,于2021年8月采集了青藏高原廓琼岗日冰川小流域5个海拔梯度(4900 m; 5000 m; 5100 m; 5200 m; 5300 m)中4个土壤发生层(A层:腐殖质层、E层:淋溶层、B层:淀积层和C层:母质层)的土壤样品,定量分析了土壤基本理化性质、微生物生物量、胞外酶活性等指标。结果表明:1)微生物生物量碳氮磷的海拔差异变化规律不同,随着土层加深微生物生物量碳氮磷随海拔变化越小。同时,各海拔之间微生物生物量均有随土层加深而显著降低的趋势(P<0.05);2)四种酶活性的海拔间变化规律各异,但整体呈现随海拔升高而升高的趋势且在表层(A和E层)增长趋势更明显,而且随土层加深显著降低(P<0.05);3)该区域土壤微生物受到碳和磷共同限制,土层越深限制越高,而且海拔越高C限制越强,但P限制降低;4)青藏高原典型冰川小流域土壤微生物生物量和胞外酶活性在海拔和土层之间存在较明显的...  相似文献

肠道菌群是一个数量庞大且种类复杂的微生物群落, 是人类的“第二基因组”, 同时又是一个被遗忘的“器官”, 参与调控宿主生理及病理过程。随着检测技术的不断提高, 肠道菌群的研究已经成为了热点, 其与各个系统疾病关系的研究也不断深入。生命早期(包括胎儿期和婴幼儿期)是婴幼儿尤其是早产儿肠道菌群定植及演替的主要阶段, 具备动态变化的特征, 且易受多种因素的影响, 主要包括剖宫产、母乳喂养减少和抗生素使用。这些因素可能会扰乱生命早期肠道菌群分布, 进而影响人类宿主近期或远期健康状况。本文就早产儿生命早期肠道菌群的建立、影响因素及其与疾病关系的研究进展作一综述。
