The relation between intracellular pH and the mitotic cycle of Physarum polycephalum was studied by two-independent techniques. Both techniques revealed a long term cycling of intracellular pH which has the same period as the mitotic cycle, Qualitative detection of the changes in intracellular pH was made by measuring the changes in fluorescence of 4-methylesculetin which had been absorbed by the plasmodium. Quantitative measurements of intracellular pH were made throughout the mitotic cycle with antimony micro pH electrodes. The cycle of intracellular pH is sinusoidal in appearance. The maximum intracellular pH (pH 6.6) occurred at, or very near to, mitosis, and was approximately 0.6 pH units higher than the minimum pH, which occurred near the middle of the mitotic cycle.  相似文献

90° stopped-flow light scattering experiments have been used to determine the forward and reverse rate constants of the reversible hexamer-dodecamer reaction of lobster hemocyanin in glycine buffer at pH 9.6. Results were obtained at 20.5° and 25° at two different levels of calcium ion, and equilibrium constants derived from the data are in good agreement with those measured earlier with the ultracentrifuge.  相似文献

Adult neurogenesis is often correlated with learning new tasks, suggesting that a function of incorporating new neurons is to permit new memory formation. However, in the zebra finch, neurons are added to the song motor pathway throughout life, long after the initial song motor pattern is acquired by about 3 months of age. To explore this paradox, we examined the relationship between adult song structure and neuron addition using sensitive measures of song acoustic structure. We report that between 4 and 15 months of age there was an increase in the stereotypy of fine-grained spectral and temporal features of syllable acoustic structure. These results indicate that the zebra finch continues to refine motor output, perhaps by practice, over a protracted period beyond the time when song is first learned. Over the same age range, there was a decrease in the addition of new neurons to HVC, a region necessary for song production, but not to Area X or the hippocampus, regions not essential for singing. We propose that age-related changes in the stereotypy of syllable acoustic structure and HVC neuron addition are functionally related.  相似文献

AIMS: To completely eliminate animal and dairy products from the lyophilization menstrum and the seed medium used to produce tetanus toxin with Clostridium tetani. METHODS AND RESULTS: Tetanus toxin production in a recently developed fermentation medium lacking animal and dairy products was studied with different seed media. It was found that soy peptone could completely replace the beef heart infusion plus animal peptone previously used as seed medium. In addition, we found that cells lyophilized in soy milk could replace the usual type of cells lyophilized in cow's milk. CONCLUSIONS: We have now developed a complete tetanus toxin production process containing no animal and dairy products. SIGNIFICANCE AND IMPACT OF THE STUDY: Toxoid preparations made from toxin produced with animal and dairy products can contain undesirable contaminants such as the prion causing bovine spongiform encephalopathy (Mad Cow's Disease) or antigenic peptides that stimulate anaphylactic reactions and other undesirable immune reactions in immunized hosts. The new vegetable-based process described here avoids such unfortunate possibilities.  相似文献

Tetranychus turkestani Ugarov and Nikolskii and Tetranychus urticae Koch RF (red form) (Acari: Tetranychidae) are closely related species. Previously, the two species were found in separate agricultural habitats in Israel. Here, additional collections were undertaken and mixed populations of the two species were found. Manipulation experiments were conducted in order to test whether sexual interactions occur when T. turkestani and T. urticae RF share the same host. Interspecific crosses showed that the two species are capable of producing viable F1 females, but that these females are sterile as their F2 eggs failed to hatch. This indicates a post-zygotic reproductive barrier, supporting the current placement of T. turkestani as a separate taxon. Mating behavior parameters revealed that males of both species courted virgin conspecific and heterospecific females at the same rate and readily tried to copulate with them. Female mate recognition seemed to be more reliable in T. turkestani than in T. urticae RF as the number of copulations was significantly higher and their duration significantly shorter in the T. turkestani interspecific (T. turkestani ♀ × T. urticae RF ♂) as compared to the intraspecific crosses, a phenomenon not observed in T. urticae RF. In mixed cultures, a significant reduction in female production was observed for T. urticae RF but not for T. turkestani, suggesting an asymmetric reproductive interference effect in favor of T. turkestani. The long term outcome of this effect is yet to be determined since additional reproductive factors such as oviposition rate and progeny survival to adulthood may reduce the probability of demographic displacement of one species by the other in overlapping niches.  相似文献
