A system for growing in liquid medium whole plants of Valeriana glechomifolia, endemic to southern Brazil and capable of accumulating bioactive valepotriates, is described. Murashige and Skoog (MS) and Gamborg B5 (B5) media (1.0×, 0.3× and 0.1× strength) without phytohormones were evaluated after four weeks of culture in relation to growth and valepotriate yield. Plants grown in 1.0× MS displayed greatest growth and valepotriate yields and the study of the light condition showed that plants grown under light and dark had similar weight increase and maximum valepotriate yield, 27.2 mg/g DW and 25.0 mg/g DW, respectively. Valtrate was the most abundant valepotriate, followed by acevaltrate and didrovaltrate.  相似文献

With the world currently facing a global amphibian extinction crisis, the development of techniques to help meet the needs of conservation managers and researchers studying the reproductive biology of amphibians is needed. Here, we developed enzyme immunoassays to measure estrone, testosterone, and progesterone hormone metabolites in the urine of Litoria raniformis, the southern bell frog. Concentrations of urinary estrone, testosterone, and progesterone increased during the breeding season for females (P < 0.05). Concentrations of urinary testosterone and progesterone increased for males during the breeding season compared with that for months where no reproductive behaviors were observed (P < 0.05). Furthermore, urinary estrone concentrations proved to be a reliable sexing tool for adult frogs, with no overlap between the sexes in 98% of cases, regardless of season. There was no difference in estrone (P = 0.204) or testosterone (P = 0.485) metabolite concentrations between samples taken immediately upon capture and those taken 12 to 24 h later from the same individual. Progesterone metabolite concentrations were lower on Day 2 than upon collection (P = 0.004). This is the first study to show that urinary hormone analysis can be a useful technique for reproductive monitoring in an amphibian. Additionally, hormone metabolite measures offer promise as sex identification tools for monomorphic species and for those whose secondary sex characteristics are visible only during the breeding season.  相似文献

The effects of collagenase on the immunohistochemical demonstrability of laminin, fibronectin and Factor VIII/RAg in human nervous tissue have been studied. The influence of this, and other proteolytic enzymes such as pepsin and trypsin, has been investigated in relation to different fixatives. Collagenase gave better results with Carnoy fixed material than after formalin fixation; unlike trypsin and pepsin, it did not produce tissue digestion.  相似文献

G0, G1, and mammalian cells and nuclei were shortly digested with either micrococcal nuclease or DNAse I, both before and after mild fixation, either before (G0) or after (G1) partial hepatectomy. Cells were Feulgen stained and examined by high resolution light microscopy. In metabolically active G1 nuclei, intranuclear DNA appears organized at least in two distinct domains, whereby the highly dispersed one is large enough to be detected at the resolution of the light microscope and appears preferentially attacked by limited DNAse I digestion. The action of the enzyme is readily apparent only in the nuclei that are first digested and then fixed. Spectroscopic characterization of the same nuclei reveals that the fixation causes a sizeable removal of proteins, mostly in the soluble chromatin subfraction. Results are discussed in terms of two control levels for gene expression and for higher order DNA structure.  相似文献

In this study, 40 individuals ofCallithrix jacchus were analyzed in order to investigate the frequency of chimerism and to determine whether the fertility of the colony was in any way affected by chimerism. In the individuals considered the degree of chimerism ranged from 2% to 97% and no correlation was established between the degree of chimerism and fertility of the individuals. Many “good breeders” were chimeric and the best female breeders displayed 84% and 97% chimerism respectively.  相似文献

  1. Pissodes castaneus represents an emerging pest species for Pinus spp. production in Latin America. This species attacks all the cultivated pine species in Patagonia Argentina causing wood damage and tree death. The objectives of this work were to describe the host preference behaviour of P. castaneus and the influence of wood damage by conspecifics on its host selection.
  2. In two-choice bioassays, P. castaneus showed a feeding preference for Pinus contorta over P. ponderosa. However, percent weight gain when feeding on either species was similar.
  3. In other bioassays, P. castaneus spent more time in areas with twigs on which other conspecifics had recently fed, although they were able to successfully feed on twigs without previous damage.
  4. These results show that while P. castaneus can successfully colonize different pine species, P. contorta may be more susceptible to attack than P. ponderosa. Silvicultural management should be prioritized considering the weevil's preference for attacking damaged hosts.

The Malvaceae family has many species that are the subject of systematic controversy. Its broad spectrum of fruit and pollen morphology has challenged taxonomists because often morphological studies lead to different conclusions. Therefore, this study of selected stages of pollen wall development in Malvastrum corchorifolium is offered as another approach to verify its current taxonomic assignment and assist in the clarification of malvacean ancestry. Aperture number, endexine thickness and the presence of basal cushions are features that concur with its present placement in the Abutileae tribe, Sidinae subtribe and genus Malvastrum. Its advanced spine morphology, long spines with pointed apices and prominent basal cushions, suggest that Malvastrum corchorifolium is highly specialized and evolutionarily advanced.  相似文献
