CheY, a small cytoplasmic response regulator, plays an essential role in the chemotaxis pathway. The concentration of phospho-CheY is thought to determine the swimming behaviour of the cell: high levels of phospho-CheY cause bacteria to rotate their flagella clockwise and tumble, whereas low levels of the phos-phorylated form of the protein allow counter-ciockwise rotation of the flagella and smooth swimming. The phosphorylation state of CheY in vivo is determined by the activity of the phosphoryl donor CheA, and by the antagonistic effect of dephosphorylation of phospho-CheY. The dephosphorylation rate is controlled by the intrinsic autohydrolytic activity of phospho-CheY and by the CheZ protein, which accelerates dephosphorylation. We have analysed the effect of CheZ on the dephosphorylation rates of several mutant CheY proteins. Two point mutations were identified which were 50-fold and 5-fold less sensitive to the activity of CheZ than was the wild-type protein. Nonetheless, the phosphorylation and autodephos-phorylation rates of these mutants, CheY23ND and CheY26KE, were observed to be identical to those of wild-type CheY in the absence of CheZ. These are the first examples of CheY mutations that reduce sensitivity to the phosphatase activity of CheZ without being altered in terms of their intrinsic phosphorylation and autodephospborylation rates, interestingly, the residues Asn-23 and Lys-26 are located on a face of CheY far from the phosphorylation site (Asp-57), distinct from the previously described site of inter-action with the histidine kinase CheA, and partially overlapping with a region implicated in interaction with the flagellar switch. 相似文献

The A. A. studied the distribution of nematodes of the genus Ancylostoma (Dubini, 1843) along the digestive tube of 45 dogs. For the collection of the worms they used the method developed by Mello and Campos. More than 60 institutions and 100 researchers were involved in Rhizobium research in 1978 in Latin America. Half of these researchers were located in Argentina and Brazil. Research activity and the application of research findings vary widely among countries. Problems that plague research include 1) inadequate training of research personnel and insufficient attention paid to the Rhizobium/Legume symbiosis at agriculture schools; 2) poorly-established research priorities that do not sufficiently weigh the immediate needs for the farmers such as the identification of limiting environmental factors (e.g. nutritional deficiencies), techniques for smallscale inoculant production, and quality control of available inoculants; 3) isolation of the researchers and a lack of adequate library support; 4) poorly integrated research teams (e.g. in many institutes researchers are either microbiologists with no agricultural background or agronomists lacking microbiological training); and 5) insufficient dissemination of research findings. Problems with inoculant production and control include 1) a local dependence on national or imported inoculants rather than on locally-selected strains, 2) poor inoculant quality control which results in low inoculation success rates and subsequent discredit to the inoculation practice, and 3) high prices for inoculants. Extension problems include 1) lacking or deficient legume-promotion programs by government agencies, 2) poor contact between research and extension workers, and 3) administrators, leaders, extension workers and agronomists working in the field that lack adequate knowledge of the Rhizobium/Legume symbiosis. Immediate measures to foster extension and legume promotion programs and informal and/or official quality control are needed in Argentina, Uruguay, Brazil, Mexico, and probably Colombia. Countries where combined efforts should primarily be directed toward stimulating research and extension include Peru, Venezuela, Costa Rica, and Chile. In Ecuador, Paraguay, Bolivia, Nicaragua, Honduras, Guatemala, the Dominican Republic and Panama, priority should be given to research. Colombia should also be included in this group as national research institutions need to be strengthened. Table 2 lists these priorities more fully. 相似文献

Prothymosin alpha (ProT alpha) is a 12.5 kDa acidic polypeptide that is considered to have a nuclear function related to cell proliferation. Inspection of its amino acid sequence revealed the presence of sequences that may serve as targets for phosphorylation by casein kinase-2 (CK-2). ProT alpha isolated from calf thymocytes was phosphorylated in vitro by CK-2. The phosphorylation sites are Ser and Thr residues located among the first 14 amino acid residues in the ProT alpha sequence. Another site that is theoretically suitable for phosphorylation by CK-2, at the C-terminus of the polypeptide, is not, in fact, phosphorylated. Thymosin alpha 1 (T alpha 1), a peptide whose sequence corresponds to the first 28 amino acids of ProT alpha, is also phosphorylated by CK-2 at the same phosphorylation sites as ProT alpha. In cultured splenic lymphocytes ProT alpha was phosphorylated at Thr residues located at positions 7, 12 and/or 13. Based on these observations we conclude that CK-2, or another cellular kinase with similar sequence specificity, is responsible for phosphorylation of ProT alpha in vivo. 相似文献

Thermodynamic identification of stable folding intermediates in the B-subunit of cholera toxin 总被引:1,自引:0,他引:1
The structural stability and domain structure of the pentameric B-subunit of cholera toxin have been measured as a function of different perturbants in order to assess the magnitude of the interactions within the B-subunits. For these studies, temperature, guanidine hydrochloride (GuHCl), and pH were used as perturbants, and the effects were measured by high-sensitivity differential scanning calorimetry, isothermal reaction calorimetry, fluorescence spectroscopy, and partial protease digestion. At pH 7.5 and in the absence of any additional perturbants, the thermal unfolding of the B-subunit pentamer is characterized by a single peak in the heat capacity function centered at 77 degrees C and characterized by a delta Hcal of 328 kcal/mol of B-subunit pentamer and delta Hvh/delta Hcal of 0.3. Lowering the pH down to 4 or adding GuHCl up to 2 M results in a decrease of the calorimetric enthalpy with no significant effect on the van't Hoff enthalpy. The transition enthalpy decreases in a sigmoidal fashion with pH, with an inflection point centered at pH 5.3. Isothermal titration calorimetric studies as a function of pH also report a transition centered at pH 5.3 and characterized by an enthalpy change of 27 kcal/mol of B-subunit pentamer at 27 degrees C. Below this pH, the enthalpy change for the unfolding transition is reduced to approximately 100 kcal/mol of B-subunit pentamer. Similar behavior is obtained with GuHCl. We examined the effect of ovule position within the ovary on the probability of seed maturation, on seed weight, and on progeny performance in the outbreeding legume Phaseolus coccineus. Ovaries of P. coccineus possess six linearly arranged ovules (ovule position one = stylar end). We found that in both 1987 and 1988, ovule position had a significant effect on the probability of seed maturation under field conditions. In 1987, ovule positions one. two, and three had a higher probability of maturing seeds than the three most basal ovule positions. In 1988, the probability of producing a mature seed in ovule position one was more similar to the three most basal ovule positions than to ovule positions two and three. The position of the ovule in the ovary had no significant effect on seed weight in 1987, but it had a significant effect in 1988. Overall, seeds from ovule positions one, two, and three tended to produce heavier seeds than the three most basal ovule positions. The effects of ovule position on progeny performance were determined in a greenhouse and a field study. In the greenhouse study, we found no significant overall effect of the position of the ovule that produced the seed on progeny performance. In the field study, we did find a significant ovule position effect on several measures of reproductive performance as well as an overall effect on reproductive performance. In addition, we found a significant interaction between ovule position and number of seeds per fruit. Progeny from the stylar end of the fruit outperformed the progeny from the peduncular end in fruits containing many seeds, whereas there were no significant differences between progeny produced in the stylar and peduncular ends of fruits containing few seeds.
