Lake restoration with aluminium (Al) has been widely used in shallow lakes, but it is unknown how ageing of the Al floc affect resuspension behaviour, sediment stability and entrainment of Al and phosphorus (P). High primary production in shallow lakes can lead to high pH in the water column with a potential release of Al and P during resuspension events. A laboratory resuspension experiment at defined hydrodynamic conditions using a calibrated erosion chamber was performed with intact sediment cores (with intact vs. disturbed biofilm on the surface) from previously Al-treated shallow Lake Möllen, NE Germany. Newly applied Al reduced surface sediment stability, but ageing led to the same stability as untreated sediment within 2 months with an intact biofilm and within 4 months with a disturbed biofilm. Dissolved Al increased markedly at resuspension after 2 days and 2 weeks of ageing and with elevated pH (9–11) in the water. The Al floc were redissolved and dissolved Al increased with elevated pH and shear velocity. Dissolved P concentration was constantly low due to excess binding capacity of the Al floc. In conclusion, Al application to shallow lakes prone to resuspension and with a high production must be done in periods with less resuspension risk to allow 2–4 months time for floc stabilization. ABSTRACT: BACKGROUND: Synthetic antisense molecules have an enormous potential for therapeutic applications in humans. The major aim of such strategies is to specifically interfere with gene function, thus modulating cellular pathways according to the therapeutic demands. Among the molecules which can block mRNA function in a sequence specific manner are peptide nucleic acids (PNA). They are highly stable and efficiently and selectively interact with RNA. However, some properties of non-modified aminoethyl glycine PNAs (aegPNA) hamper their in vivo applications. RESULTS: We generated new backbone modifications of PNAs, which exhibit more hydrophilic properties. When we examined the activity and specificity of these novel phosphonic ester PNAs (pePNA) molecules in medaka (Oryzias latipes) embryos, high solubility and selective binding to mRNA was observed. In particular, mixing of the novel components with aegPNA components resulted in mixed PNAs with superior properties. Injection of mixed PNAs directed against the medaka six3 gene, which is important for eye and brain development, resulted in specific six3 phenotypes. CONCLUSIONS: PNAs are well established as powerful antisense molecules. Modification of the backbone with phosphonic ester side chains further improves their properties and allows the efficient knock down of a single gene in fish embryos. 相似文献

A series of 31 patients with various degrees of chronic obstructive pulmonary disease underwent right heart catheterization using flow-directed thermodilution catheters. Both rest and supine exercise values were obtained. The patients were divided into two groups on the basis of their reduction in forced expiratory volume in 1 s (FEV1). In patients with FEV1 values of greater than or equal to 1,300 ml (group 1), the arterial oxygen partial pressure (PaO2) did not significantly change with exercise, while in patients with FEV1 of less than or equal to 1,200 ml (group 2) PaO2 significantly (p less than 0.05) fell in response to exercise. In group 2, a significant increase of total pulmonary resistance (TPR) with exercise was found (p less than 0.01). Pulmonary vascular resistance (PVR) remained unchanged in both subgroups. It is suggested that the value of PVR for subgroup 2 is artificially low because an important variable, namely pulmonary artery wedge pressure, is influenced by alveolar pressure in patients with an uneven distribution of perfusion and ventilation at pulmonary venous pressures lower than alveolar pressure. The steeper slope of the pressure-flow relationship in these patients is probably due to an increased vascular tone caused by chronic hypoxia at rest and further fall of PaO2 and rise of arterial CO2 partial pressure in response to exercise. 相似文献
