Available tools to study rhizosphere characteristics at a sub-mm spatial resolution suffer from a number of shortfalls, including geometrically and physiologically ill-defined root layers containing soil or other growth medium. Such designs may result in over- or underestimation of root-induced changes in the rhizosphere. We present a novel rhizobox design that overcomes these shortfalls. Plants are pre-grown in a soil–root compartment with an opening slit at the bottom. As plants reach the targeted physiological stage, this compartment is transferred on top of a rhizosphere soil compartment attached to a vertical root-only compartment. The latter is made up of a membrane (pore size 7 m to restrict root hair growth into the rhizosphere compartment or 30 m to restrict only root growth) and a transparent acrylic window which is gently pressed against the membrane and rhizosphere soil compartment using an adjustable screw. This design allows roots to penetrate from the upper soil–root compartment through the slit into the root-only compartment. Root growth and distribution can be monitored through the acrylic window using digital camera equipment. Upon termination of the experiment, the rhizosphere compartment is removed and frozen prior to separation of sub-mm soil layers using microtome techniques. In a test experiment, canola (Brassica napus L. cv. Sprinter) developed a fairly dense root monolayer within 8 days. Using measurement of soil characteristics at 0.5–1-mm increments across the rhizosphere we demonstrate that the proposed rhizobox design is yielding reproducible data. Due to exudation of LMWOC, we found a statistically significant increase of DOC towards the root plane, whereas more stable soil characteristics were not affected by root activity. Limitations and further extensions of this rhizobox design, including the use of micro suction cups and microsensors for pH and redox potential to measure spatial and temporal changes in a non-destructive manner are discussed along with potential applications such as validation of rhizosphere models.  相似文献

The Saccharomyces cerevisiae SAC1 gene encodes an integral membrane protein of the endoplasmic reticulum (ER) and the Golgi apparatus. Yeast SAC1 mutants display a wide array of phenotypes including inositol auxotrophy, cold sensitivity, secretory defects, disturbed ATP transport into the ER, or suppression of actin gene mutations. At present, it is not clear how these phenotypes relate to the finding that SAC1 displays polyphosphoinositide phosphatase activity. Moreover, it is still an open question whether SAC1 functions similarly in mammalian cells, since some phenotypes are yeast-specific. Potential protein interaction partners and, connected to that, possible regulatory circuits have not been described. Therefore, we have cloned human SAC1 (hSAC1), show that it behaves similar to ySac1p in terms of substrate specificity, demonstrate that the endogenous protein localizes to the ER and Golgi, and identify for the first time members of the coatomer I (COPI) complex as interaction partners of hSAC1. Mutation of a putative COPI interaction motif (KXKXX) at its C terminus abolishes interaction with COPI and causes accumulation of hSAC1 in the Golgi. In addition, we generated a catalytically inactive mutant, demonstrate that its lipid binding capacity is unaltered, and show that it accumulates in the Golgi, incapable of interacting with the COPI complex despite the presence of the KXKXX motif. These results open the possibility that the enzymatic function of hSAC1 provides a switch for accessibility of the COPI interaction motif.  相似文献

The central protein of the sulfur-oxidizing enzyme system of Paracoccus pantotrophus, SoxYZ, formed complexes with subunits associated and covalently bound. In denaturing SDS-polyacrylamide gel electrophoresis (PAGE) SoxY migrated at 12 and SoxZ at 16kDa. SDS-PAGE of homogeneous SoxYZ without reductant separated dimeric complexes of 25, 29, and 32kDa identified by the N-terminal amino acid sequences as SoxY-Y, SoxY-Z, and SoxZ-Z, and subunit cleavage by reduction suggested their linkage via protein disulfide bonds. SoxYZ was reversibly redox active between -0.25 and 0.2V, as monitored by a combined electrochemical and FTIR spectroscopic approach. The dimanganese SoxB protein (58.611Da) converted the covalently linked heterodimer SoxY-Z to SoxYZ with associated subunits which in turn aggregated to the heterotetramer Sox(YZ)(2). This reaction depended on time and the SoxB concentration, and demonstrated the interaction of these two Sox proteins.  相似文献

The restriction endonuclease EcoRII requires the cooperative interaction with two copies of the sequence 5'CCWGG for DNA cleavage. We found by limited proteolysis that EcoRII has a two-domain structure that enables this particular mode of protein-DNA interaction. The C-terminal domain is a new restriction endonuclease, EcoRII-C. In contrast to the wild-type enzyme, EcoRII-C cleaves DNA specifically at single 5'CCWGG sites. Moreover, substrates containing two or more cooperative 5'CCWGG sites are cleaved much more efficiently by EcoRII-C than by EcoRII. The N-terminal domain binds DNA specifically and attenuates the activity of EcoRII by making the enzyme dependent on a second 5'CCWGG site. Therefore, we suggest that a precursor EcoRII endonuclease acquired an additional DNA-binding domain to enable the interaction with two 5'CCWGG sites. The current EcoRII molecule could be an evolutionary intermediate between a site-specific endonuclease and a protein that functions specifically with two DNA sites such as recombinases and transposases. The combination of these functions may enable EcoRII to accomplish its own propagation similarly to transposons.  相似文献

Experimental data on rhizosphere characteristics at high spatial resolution are required to improve our knowledge on phytoavailability of nutrients and pollutants. In numerous studies, sectioning using refrigerated microtomes has been employed to obtain thin soil layers at defined distances from the root surface. In this study, we assessed the effect of thin slicing and freezing on soil chemical characteristics. Two experimental soils were frozen at –20°C and sliced using a refrigerated microtome. In general, chemical changes relative to the non-sliced control were more pronounced as the trim thickness (thickness of a single slice) decreased. Maximum increases in pH and electrical conductivity (EC) for the smallest trim thickness used (20 m) were 0.9 units and 50%, respectively. Extractable fractions of P (0.5 M NaHCO3) K, Mg, Mn, Na and Si (1 M NH4NO3) increased up to 40, 91, 19, 621, 50 and 100%, respectively. Based on these results, we suggest to use a trim thickness of 200 m. Apart from slicing, freezing (a prerequisite for the microtome technique) was found to bias soil chemical parameters. To circumvent microtome-related artifacts we present a home-made slicing device as a cost-effective alternative, which allows sectioning of non-frozen rhizosphere soil employing one single slice.  相似文献

A new phenotypic test, called the Carbapenem Inactivation Method (CIM), was developed to detect carbapenemase activity in Gram-negative rods within eight hours. This method showed high concordance with results obtained by PCR to detect genes coding for the carbapenemases KPC, NDM, OXA-48, VIM, IMP and OXA-23. It allows reliable detection of carbapenemase activity encoded by various genes in species of Enterobacteriaceae (e.g., Klebsiella pneumoniae, Escherichia coli and Enterobacter cloacae), but also in non-fermenters Pseudomonas aeruginosa and Acinetobacter baumannii. The CIM was shown to be a cost-effective and highly robust phenotypic screening method that can reliably detect carbapenemase activity.  相似文献

Granulocyte-colony-stimulating-factor (G-CSF) induces mobilization of progenitor cells but may also exert pro-inflammatory and pro-thrombotic effects. Treatment with recombinant G-CSF after acute myocardial infarction is currently under examination and has been associated with in-stent restenosis. However, it is not known whether plasma levels of endogenous G-CSF are also associated with an increased cardiovascular risk. Therefore we included 280 patients with angiographically proven stable coronary artery disease. G-CSF was measured by specific ELISA and patients were followed for a median of 30 months for the occurrence of major adverse cardiovascular events (MACE: death, myocardial infarction, re-hospitalization). Those with cardiac events during follow-up showed significant higher G-CSF levels (32.3 pg/mL IQR 21.4–40.5 pg/mL vs. 24.6 pg/mL IQR 16.4–34.9 pg/mL; p<0.05) at baseline. Patients with G-CSF plasma levels above the median had a 2-fold increased risk for MACE (p<0.05). This was independent from established cardiovascular risk factors. In addition, G-CSF above the median was a predictor of clinical in-stent restenosis after implantation of bare-metal stents (6.6% vs. 19.4%; p<0.05) but not of drug-eluting stents (7.7% vs. 7.6%; p = 0.98). This data suggests that endogenous plasma levels of G-CSF predict cardiovascular events independently from established cardiac risk factors and are associated with increased in-stent restenosis rates after implantation of bare metal stents.  相似文献

Sexual selection takes place in complex environments where females evaluating male mating signals are confronted with stimuli from multiple sources and modalities. The pattern of expression of female preferences may be influenced by interactions between modalities, changing the shape of female preference functions, and thus ultimately altering the selective landscape acting on male signal evolution. We tested the hypothesis that the responses of female gray treefrogs, Hyla versicolor, to acoustic male advertisement calls are affected by interactions with visual stimuli. We measured preference functions for several call traits under two experimental conditions: unimodal (only acoustic signals presented), and multimodal (acoustic signals presented along with a video‐animated calling male). We found that females were more responsive to multimodal stimulus presentations and, compared to unimodal playbacks, had weaker preferences for temporal call characteristics. We compared the preference functions obtained in these two treatments to the distribution of male call characteristics to make inferences on the strength and direction of selection expected to act on male calls. Modality interactions have the potential to influence the course of signal evolution and thus are an important consideration in sexual selection studies.  相似文献
