Bread‐in‐beer and bread‐in‐water are prevalent home recipe trap baits for attracting German cockroaches (GCRs), Blattella germanica (L.) (Dictyoptera: Blattellidae), which are significant urban pests. Our objectives were to (1) test the attractiveness of these baits, (2) study the underlying factors of GCR attraction, and (3) determine whether a blend of synthetic bread odorants could replace bread in a trap lure. In large‐arena laboratory experiments with laboratory‐reared GCR males, traps baited with rye bread not only captured eightfold more males than unbaited control traps but also most males released into bioassay arenas. Neither beer nor water enhanced the attractiveness of bread. Bread crust as a bait was more effective than bread crumbs. As Porapak Q headspace volatile extracts of rye bread attracted GCRs, all rye bread odorants in extracts were identified by gas chromatography‐mass spectrometry. Synthetic rye bread odorants and other known bread odorants were then assembled into a master blend. This master blend, and even partial blends lacking certain groups of organic volatiles such as aldehydes and ketones, proved very attractive to GCRs. We conclude that rye bread could be used as an effective bait in retainer traps or, laced with insecticide, as a food source in bait stations. A lure of synthetic bread odorants may eventually replace bread as bait, but the minimum number of essential odorants for that lure has yet to be determined.  相似文献

Background

Cerebral malaria (CM) is a syndrome characterized by neurological signs, seizures and coma. Despite the fact that CM presents similarities with cerebral stroke, few studies have focused on new supportive therapies for the disease. Hyperbaric oxygen (HBO) therapy has been successfully used in patients with numerous brain disorders such as stroke, migraine and atherosclerosis.

Methodology/Principal Findings

C57BL/6 mice infected with Plasmodium berghei ANKA (PbA) were exposed to daily doses of HBO (100% O2, 3.0 ATA, 1–2 h per day) in conditions well-tolerated by humans and animals, before or after parasite establishment. Cumulative survival analyses demonstrated that HBO therapy protected 50% of PbA-infected mice and delayed CM-specific neurological signs when administrated after patent parasitemia. Pressurized oxygen therapy reduced peripheral parasitemia, expression of TNF-α, IFN-γ and IL-10 mRNA levels and percentage of γδ and αβ CD4+ and CD8+ T lymphocytes sequestered in mice brains, thus resulting in a reduction of blood-brain barrier (BBB) dysfunction and hypothermia.

Conclusions/Significance

The data presented here is the first indication that HBO treatment could be used as supportive therapy, perhaps in association with neuroprotective drugs, to prevent CM clinical outcomes, including death.  相似文献

Two distantly related classes of cylindrical chaperonin complexes assist in the folding of newly synthesized and stress-denatured proteins in an ATP-dependent manner. Group I chaperonins are thought to be restricted to the cytosol of bacteria and to mitochondria and chloroplasts, whereas the group II chaperonins are found in the archaeal and eukaryotic cytosol. Here we show that members of the archaeal genus Methanosarcina co-express both the complete group I (GroEL/GroES) and group II (thermosome/prefoldin) chaperonin systems in their cytosol. These mesophilic archaea have acquired between 20 and 35% of their genes by lateral gene transfer from bacteria. In Methanosarcina mazei G?1, both chaperonins are similarly abundant and are moderately induced under heat stress. The M. mazei GroEL/GroES proteins have the structural features of their bacterial counterparts. The thermosome contains three paralogous subunits, alpha, beta, and gamma, which assemble preferentially at a molar ratio of 2:1:1. As shown in vitro, the assembly reaction is dependent on ATP/Mg2+ or ADP/Mg2+ and the regulatory role of the beta subunit. The co-existence of both chaperonin systems in the same cellular compartment suggests the Methanosarcina species as useful model systems in studying the differential substrate specificity of the group I and II chaperonins and in elucidating how newly synthesized proteins are sorted from the ribosome to the proper chaperonin for folding.  相似文献

Zusammenfassung Die indirekten und direkten Flugmuskelfasern der Wanderheuschrecke enthalten parallel und voneinander getrennt verlaufende Myofibrillen. In den interfibrillären Räumen liegen zahlreiche, säulenförmig aneinandergereihte Mitochondrion, ein dichtes endoplasmatisches Retikulum und Tracheolen. In Höhe der A-I-Band-Grenzen befindet sich ein charakteristisches retikuläres System, dessen Queranordnung als Diade zu bezeichnen ist. Die chromatinreichen Kerne liegen peripher dicht unter dem Sarkolemm.Sowohl bei den indirekten als auch bei den direkten Flugmuskelgruppen kommt es im Verlaufe des Dauerfluges zu Transformationen der Mitochondrien, die sich in einer erheblichen Schwellung und in einer Umwandlung der Cristae zu Tubuli äußern. Diese Umwandlung wird als intramitochondriale Kompensation aufgefaßt, um das Verhältnis von Oberfläche der inneren membranösen Phase zum Volumen der inneren wäßrigen Phase — d. h. die spezifische Oberfläche — möglichst konstant zu halten. Eine Vesikulation der Tubuli könnte der morphologische Ausdruck für eine Dekompensation sein, da dann der Zusammenhang der äußeren wäßrigen Phase verlorengeht.Die Mitochondrien verschiedener Muskeln reagieren nicht synchron auf die Dauerbelastung, sondern ausgesprochen belastungs- und funktionsspezifisch: Zunächst transformieren sich die Mitochondrien der indirekten, dann erst die der direkten Flugmuskelgruppen, und der anfängliche Schwirrflug geht während fortschreitenden Dauerfluges in einen Flatterflug über. Die unterschiedlichen Erschöpfungszeitpunkte der indirekten Flugmuskeln werden durch die jeweils spezifische Funktion und die ontogenetische Entwicklung erklärt.  相似文献

Human prominin-1/CD133 has been reported to be expressed in neural and hematopoietic stem/progenitor cells and in embryonic, but not adult, epithelia. This lack of detection of human prominin-1, as defined by its glycosylation-dependent AC133 epitope, is surprising given the expression of the murine ortholog in adult epithelia. Here, we demonstrate, by using a novel prominin-1 antiserum (hE2), that the decrease of AC133 immunoreactivity observed during differentiation of the colonic adenocarcinoma-derived Caco-2 cells is not paralleled by a down-regulation of prominin-1. We have also shown that hE2 immunoreactivity, but not AC133 immunoreactivity, is present in several adult human tissues, such as kidney proximal tubules and the parietal layer of Bowmans capsule of juxtamedullary nephrons, and in lactiferous ducts of the mammary gland. These observations suggest that only the AC133 epitope is down-regulated upon cell differentiation. Furthermore, hE2 immunoreactivity has been detected in several kidney carcinomas derived from proximal tubules, independent of their grading. Interestingly, in one particular case, the AC133 epitope, which is restricted to stem cells in normal adult tissue, was up-regulated in the vicinity of the tumor. Our data thus show that (1) in adults, the expression of human prominin-1 is not limited to stem and progenitor cells, and (2) the epitopes of prominin-1 might be useful for investigating solid cancers. It is increasingly recognized that psychological stress influences inflammatory responses and mood. Here, we investigated whether psychological stress (combined acoustic and restraint stress) activates the tryptophan (Trp) catabolizing enzyme indoleamine 2,3-dioxygenase 1(IDO1) and thereby alters the immune homeostasis and behavior in mice. We measured IDO1 mRNA expression and plasma levels of Trp catabolites after a single 2-h stress session and in repeatedly stressed (4.5-days stress, 2-h twice a day) naïve BALB/c mice. A role of cytokines in acute stress-induced IDO1 activation was studied after IFNγ and TNFα blockade and in IDO1−/− mice. RU486 and 1-Methyl-L-tryptophan (1-MT) were used to study role of glucocorticoids and IDO1 on Trp depletion in altering the immune and behavioral response in repeatedly stressed animals. Clinical relevance was addressed by analyzing IDO1 activity in patients expecting abdominal surgery. Acute stress increased the IDO1 mRNA expression in brain, lung, spleen and Peyer''s patches (max. 14.1±4.9-fold in brain 6-h after stress) and resulted in a transient depletion of Trp (−25.2±6.6%) and serotonin (−27.3±4.6%) from the plasma measured 6-h after stress while kynurenine levels increased 6-h later (11.2±9.3%). IDO1 mRNA up-regulation was blocked by anti-TNFα and anti-IFNγ treatment. Continuous IDO1 blockade by 1-MT but not RU486 treatment normalized the anti-bacterial defense and attenuated increased IL-10 inducibility in splenocytes after repeated stress as it reduced the loss of body weight and behavioral alterations. Moreover, kynurenic acid which remained increased in 1-MT treated repeatedly stressed mice was identified to reduce the TNFα inducibility of splenocytes in vitro and in vivo. Thus, psychological stress stimulates cytokine-driven IDO1 activation and Trp depletion which seems to have a central role for developing stress-induced immunosuppression and behavioral alteration. Since patients showed Trp catabolism already prior to surgery, IDO is also a possible target enzyme for humans modulating immune homeostasis and mood.  相似文献

Comprehensive field investigations have been conducted four times on Nam Co, central Tibet, from September 2005 to September 2008. Here, we present the preliminary results focusing on the bathymetric survey and water quality measurements. The isobathic map shows that Nam Co is a high-altitude, deep lake where a flat and large basin lies in the central part with a water depth of more than 90 m. Water depth data from the northwestern bank areas of Nam Co provide unquestionable evidence of rising water levels in the last 3 decades because of the formation of two small islands that were still peninsulas in the 1970s. Water quality measurements taken at 19 stations during three summer field campaigns (2006, 2007 and 2008) covering almost all of the lake areas showed that the temperature, pH, dissolved oxygen and electric conductivity of surface water are on average 11.43°C, 9.21, 8.90 mg l−1 and 1,851 μS cm−1, respectively. The surface water shows no obvious spatial variability among all the stations. Vertical fluctuations of profiles, however, display some differences in thermocline and related parameters, such as pH and dissolved oxygen. According to the vertical variations of water quality parameters, the water column in relatively deep lake areas of Nam Co could be divided into three layers with distinctly various features: the epilimnion is from the surface to about 18–20 m depth in which the parameters are homogeneous with higher temperature and abundant sunlight; the metalimnion ranged from 20–60 m where a thermocline develops; the deepest layer forms a cold and dark hypolimnion.  相似文献

The mechanism underlying perinuclear calcium spiking induced during legume root endosymbioses is largely unknown. Lotus japonicus symbiosis-defective castor and pollux mutants are impaired in perinuclear calcium spiking. Homology modeling suggested that the related proteins CASTOR and POLLUX might be ion channels. Here, we show that CASTOR and POLLUX form two independent homocomplexes in planta. CASTOR reconstituted in planar lipid bilayers exhibited ion channel activity, and the channel characteristics were altered in a symbiosis-defective mutant carrying an amino acid replacement close to the selectivity filter. Permeability ratio determination and competition experiments reveled a weak preference of CASTOR for cations such as potassium over anions. POLLUX has an identical selectivity filter region and complemented a potassium transport–deficient yeast mutant, suggesting that POLLUX is also a potassium-permeable channel. Immunogold labeling localized the endogenous CASTOR protein to the nuclear envelope of Lotus root cells. Our data are consistent with a role of CASTOR and POLLUX in modulating the nuclear envelope membrane potential. They could either trigger the opening of calcium release channels or compensate the charge release during the calcium efflux as counter ion channels.  相似文献

Little is known about the use of heterothermy by wild bats during summer, especially for tree-roosting species. Because thermal conditions within tree roosts can fluctuate widely with ambient temperature, which affects thermoregulatory energy expenditure during diurnal roosting, we measured skin temperatures of free-ranging male Nyctophilus geoffroyi (8 g) to quantify the relation between summer torpor use and roost thermal conditions. Bats roosted under bark on the northern (sunny) side of trees and entered torpor every day, usually near sunrise. Bats exhibited two bouts of torpor on most days: the first occurred in the morning, was terminated by partially passive rewarming, and was followed by a period of normothermy during the warmest part of the day; a second torpor bout occurred in the late afternoon, with arousal near sunset. On the warmest days, bats had only a single, short morning bout. On the coolest days, bats remained torpid throughout the day, and one 2-d bout was observed. Thus, presumably owing to their poorly insulated roosts and the high energetic cost of normothermy at temperatures below 30 degrees C, the extent and timing of heterothermy was closely related to the cycle of diurnal temperatures. Our study indicates that torpor use is important for energy maintenance during summer diurnal roosting of N. geoffroyi and likely of other small, tree-roosting bats.  相似文献
