Feeding experiments were conducted in situ in flow through boxes to determine the vulnerability of different prey types (the mayfly larvae Baetis, Ephemerella and Ecdyonurus spp., the caddisfly larva Hydropsyche spp. and the crustacean Gammarus spp.) to the predaceous water bug Aphelocheirus aestivalis and to estimate the predation impact of Aphelocheirus on prey populations. Experiments with equal densities of mixed prey and experiments where each prey was tested individually revealed that Baetis and Ephemerella were most vulnerable to Aphelocheirus predation: Hydropsyche, Ecdyonurus and Gammarus were little/not preyed upon. The present results suggest that vulnerability of prey depends mainly upon prey mobility and microhabitat overlap between predator and prey and that feeding behaviour of Aphelocheirus resembles more that of megalopterans than of stoneflies. Experiments with different prey densities (120–720 prey m−2) showed that the predation rate of Aphelocheirus increased with increasing prey density. Experiments with different substrates documented that mortality rates of prey decreased with increasing substrate complexity. When substrate conditions were complex mortality rates in the control and in the experimental boxes were the same which suggest little importance of Aphelocheirus predation on mayflies in the study site.  相似文献

A Gram-positive spore-forming thermophilic strict anaerobic bacterium, designated FH1, was isolated from enrichments at 65 degrees C with dextran as sole carbon and energy source. A sequence analysis of the 16S rRNA gene revealed 99.2% identity of FH1 to Thermoanaerobacterium thermosaccharolyticum. Furthermore, the substrate spectra of both organisms were similar. It was therefore concluded that FH1 represents a new strain within the species T. thermosaccharolyticum. The optimal growth temperature of strain FH1 was 68 degrees C. The isolated organism produced a thermostable and thermoactive dextranase with a native molecular mass of approximately 200,000 Da. The enzyme was concentrated from the cell-free culture supernatant by ammonium sulfate precipitation. The resulting crude dextranase exhibited optimal activity from 65 to 70 degrees C and a pH optimum of 5.5.  相似文献

Zusammenfassung Die Vorgeschichte und der Verlauf von 8 Bruten eines Hohltauben-Männchens mit einem Ringeltauben-Weibchen (Columba oenas × Columba palumbus), die 16 Eier und daraus 4 lebensfähige Jungvögel (2 , 2 ) ergaben, werden geschildert.Die Bastarde sind nach Größe, Gewicht und Färbung intermediär. Sie haben den grünen Halsschild der Hohltaube, doch tritt der Einfluß der Ringeltaube insgesamt stärker hervor, z. B. in der Färbung der Brust und des Flügels, sowie in der Flügellänge, die etwa die Länge des Ringeltaubenflügels erreicht.Der Balzruf des einzigen männlichen Bastards, der das Jugendalter überlebte (HR3), klang wie kurrrú oder rruuhg; er hatte keine Ähnlichkeit mit dem der beiden Elternarten. Auch sein Verhalten bei der Balz (Haltung beim Ruf, Beteiligung beim Nest-bau) wich von dem Verhalten männlicher Hohl- und Ringeltauben ab. Er verpaarte sich mit seiner Schwester HR2. HR2 trug auf einer flachen hölzernen Unterlage wenige Zweige zusammen und legte ein unbefruchtetes Ei, das es gleich im Stich ließ. Herrn Prof. Dr. Erwin Stresemann zum 70. Seven commercial 3- to 7-ring (R) polycyclic aromatic hydrocarbons (PAH) as well as PAH derived from lignite tar were spiked into 3 soils (0.8 to 9.7% of organic carbon). The disappearance of the original PAH was determined for the freshly spiked soils, for soils incubated for up to 287 d with their indigenous microflora, and for autoclaved, unsterile and pasteurized soils inoculated with basidiomycetous and ascomycetous fungi. Three to 12 d after spiking, 22 to 38% of the PAH could no longer be recovered from the soils. At 287 d, 88.5 to 92.7%, 83.4 to 87.4%, and 22.0 to 42.1% of the 3-, 4-, and 5- to 7-R PAH, respectively, had disappeared from the unsterile, uninoculated soils. In 2 organic-rich sterile soils, the groups of wood- and straw-degrading, terricolous, and ectomycorrhizal fungi reduced the concentration of 5 PAH by 12.6, 37.9, and 9.4% in 287 d. Five- to 7-R PAH were degraded as efficiently as most of the 3- to 4-R PAH. In organic-rich unsterile soils inoculated with wood- and straw-degrading fungi, the degradation of 3- to 4-R PAH was not accelerated by the presence of fungi.The 5- to 7-R PAH, which were not attacked by bacteria, were degraded by fungi to 29 to 42% in optimum combinations of fungal species and soil type. In organic-poor unsterile soil, these same fungi delayed the net degradation of PAH possibly for 2 reasons. Mycelia of Pleurotus killed most of the indigenous soil bacteria expected to take part in the degradation of PAH, whereas those of Hypholoma and Stropharia promoted the development of opportunistic bacteria in the soil, which must not necessarily be PAH degraders. Contemporarily, the contribution of the fungi themselves to PAH degradation may be negligible in the absence of soil organic matter due to the lower production of ligninolytic enzymes. It is concluded that fungi degrade PAH irrespective of their molecular size in organic-rich and wood chip-amended soils which promote fungal oxidative enzyme production.  相似文献

In determining the structure of large proteins by NMR, it would be desirable to obtain complete backbone, side-chain, and NOE assignments efficiently, with a minimum number of experiments and samples. Although new strategies have made backbone assignment highly efficient, side-chain assignment has remained more difficult. Faced with the task of assigning side-chains in a protein with poor relaxation properties, the Tetrahymena histone acetyltransferase tGCN5, we have developed an assignment strategy that would provide complete side-chain assignments in cases where fast 13C transverse relaxation causes HCCH-TOCSY experiments to fail. Using the strategy presented here, the majority of aliphatic side-chain proton and carbon resonances can be efficiently obtained using optimized H(CC-CO)NH-TOCSY and (H)C(C-CO)NH-TOCSY experiments on a partially deuterated protein sample. Assignments can be completed readily using additional information from a 13 C-dispersed NOESY-HSQC spectrum. Combination of these experiments with H(CC)NH-TOCSY and (H)C(C)NH-TOCSY may provide complete backbone and side-chain assignments for large proteins using only one or two samples.  相似文献

Metabolic engineering of the early non-mevalonate terpenoid pathway of Escherichia coli was carried out to increase the supply of prenyl pyrophosphates as precursor for carotenoid production. Transformation with the genes dxs for over-expression of 1-deoxy-d-xylulose 5-phosphate synthase, dxr for 1-deoxy-d-xylulose 5-phosphate reductoisomerase and idi encoding an isopentenyl pyrophosphate stimulated carotenogenesis up to 3.5-fold. Co-transformation of idi with either dxs or dxr had an additive effect on ß-carotene and zeaxanthin production which reached 1.6 mg g–1 dry wt.  相似文献

We have employed a novel in vivo approach to study the structure and function of the eukaryotic kinetochore multiprotein complex. RNA interference (RNAi) was used to block the synthesis of centromere protein A (CENP-A) and Clip-170 in human cells. By coexpression, homologous kinetochore proteins from Saccharomyces cerevisiae were then tested for the ability to complement the RNAi-induced phenotypes. Cse4p, the budding yeast CENP-A homolog, was specifically incorporated into kinetochore nucleosomes and was able to complement RNAi-induced cell cycle arrest in CENP-A-depleted human cells. Thus, Cse4p can structurally and functionally substitute for CENP-A, strongly suggesting that the basic features of centromeric chromatin are conserved between yeast and mammals. Bik1p, the budding yeast homolog of human CLIP-170, also specifically localized to kinetochores during mitosis, but Bik1p did not rescue CLIP-170 depletion-induced cell cycle arrest. Generally, the newly developed in vivo complementation assay provides a powerful new tool for studying the function and evolutionary conservation of multiprotein complexes from yeast to humans.  相似文献

BACKGROUND AND AIMS: The increase of molecular data and the resulting insights into legume systematics make the search for new morphological characters and a careful re-investigation of already stated characters necessary. Bracteoles are small, reduced leaves borne close to the base of lateral branches. Although they seem unimportant in older buds, they have an ecological function in protecting the sepal primordia. Furthermore, a morphogenetic function in mediating the onset of sepal initiation is suspected in the literature. The occurrence of bracteoles varies within Papilionoideae, and their distribution is used in legume systematics. But this is open to criticism, because there is a tendency to use 'absent' for 'caducous'. Thus attention here was paid to the initiation of bracteoles as well as to the sequence of sepal initiation. METHODS: The floral development of 30 taxa out of 15 tribes of Papilionoideae was investigated using scanning electron microscopy (SEM). KEY RESULTS: In five taxa the bracteoles initiated, but suppressed early. Furthermore, a broad variability of sepal initiation was found. Besides the widely stated unidirectional pattern, modified unidirectionality, tendencies towards whorled, fully whorled, bidirectional and successive initiation of sepals were all found. CONCLUSION: Initiated but suppressed bracteoles are presented as a 'new' character in Papilionoideae. Considering the presence of bracteoles as a plesiomorphy, their suppression can be seen as a step towards completely reduced bracteoles. The remarkable variability of the sequence of sepal initiation questions the widely stated unidirectionality of organ initiation in Papilionoideae. The different modes of sepal initiation are deducible from the helical pattern of some caesalpinioids, which is seen as a developmental link of the flowers of Papilionoideae and Caesalpinioideae. The bidirectional sepal initiation is possibly a consequence of the morphogenetic function of bracteoles, although bidirectionality is not found in all taxa with reduced bracteoles.  相似文献
