Phytophthora cinnamomi occurs worldwide and has a host range in excess of 1,000 plant species. Avocados (Persea americana Mill) have been described as highly susceptible to this soil-borne pathogen. Here, the regulation of defence responses in avocado root seedlings inoculated with P. cinnamomi mycelia is described. A burst of reactive oxygen species (ROS) was observed 4 days after inoculation. The higher physiological concentration of H2O2 induced by P. cinnamomi on avocado roots had no effect on in vitro growth of the oomycete. Total phenols and epicathecin content showed a significant decrease, but lignin and pyocianidins exhibited no changes after inoculation. Also, increased nitric oxide (NO) production was observed 72 h after treatment. We studied the effects of one NO donor [sodium nitroprusside (SNP)], and one NO scavenger [2- to 4-carboxyphenyl-4,4,5,5-tetramethylimidazole-1-oxyl-3-oxide (CPTIO)] to determine the role of NO during root colonisation by P. cinnamomi mycelia. Pretreatment of the roots with CPTIO, but not with SNP, inhibited root colonisation suggesting an important role for NO production during the avocado–P. cinnamomi interaction. Our data suggest that although defence responses are activated in avocado roots in response to P. cinnamomi infection, these are not sufficient to avoid pathogen invasion.  相似文献

Galectin-3 (Gal-3) is a multifunctional protein that plays different roles in cancer biology. To better understand the role of Gal-3 and its ligands during colon carcinogenesis, we studied its expression in tumors induced in rats treated with 1,2-dimethylhydrazine (DMH) and in human tissues. Normal colon from untreated rats showed no staining using two specific monoclonal antibodies. In contrast, morphologically normal colon from DMH-treated rats and dysplastic aberrant crypt foci were strongly stained, indicating that increased Gal-3 expression is an early event during the neoplastic transformation in colon cells. Gal-3 was weakly expressed in adenocarcinomas. Overall, the Gal-3 expression pattern observed in the DMH rat model closely resembles that displayed by human colon stained with the same antibodies. We also found that Gal-3 phosphorylation diminishes in serines while increasing in tyrosines during rat colon carcinogenesis. Finally, we showed that Gal-3–ligands expression is strikingly similar in rat and human malignant colon and in non-malignant tissues. In conclusion, the DMH-induced rat colon cancer model displays expression patterns of Gal-3 and its ligands very similar to those observed in human samples. This animal model should contribute to clarifying the role of Gal-3 in colon carcinogenesis and also to finding effective preventive cancer agents based on Gal-3 targeting. In contrast to the classical view of development as a preprogrammed and deterministic process, recent studies have demonstrated that stochastic perturbations of highly non-linear systems may underlie the emergence and stability of biological patterns. Herein, we address the question of whether noise contributes to the generation of the stereotypical temporal pattern in gene expression during flower development. We modeled the regulatory network of organ identity genes in the Arabidopsis thaliana flower as a stochastic system. This network has previously been shown to converge to ten fixed-point attractors, each with gene expression arrays that characterize inflorescence cells and primordial cells of sepals, petals, stamens, and carpels. The network used is binary, and the logical rules that govern its dynamics are grounded in experimental evidence. We introduced different levels of uncertainty in the updating rules of the network. Interestingly, for a level of noise of around 0.5-10%, the system exhibited a sequence of transitions among attractors that mimics the sequence of gene activation configurations observed in real flowers. We also implemented the gene regulatory network as a continuous system using the Glass model of differential equations, that can be considered as a first approximation of kinetic-reaction equations, but which are not necessarily equivalent to the Boolean model. Interestingly, the Glass dynamics recover a temporal sequence of attractors, that is qualitatively similar, although not identical, to that obtained using the Boolean model. Thus, time ordering in the emergence of cell-fate patterns is not an artifact of synchronous updating in the Boolean model. Therefore, our model provides a novel explanation for the emergence and robustness of the ubiquitous temporal pattern of floral organ specification. It also constitutes a new approach to understanding morphogenesis, providing predictions on the population dynamics of cells with different genetic configurations during development.  相似文献

We report on room temperature electron transfer in the reaction center (RC) complex purified from Rhodobacter sphaeroides. The protein was embedded in trehalose-water systems of different trehalose/water ratios. This enabled us to get new insights on the relationship between RC conformational dynamics and long-range electron transfer. In particular, we measured the kinetics of electron transfer from the primary reduced quinone acceptor (Q(A)(-)) to the primary photo oxidized donor (P(+)), by time-resolved absorption spectroscopy, as a function of the matrix composition. The composition was evaluated either by weighing (liquid samples) or by near infrared spectroscopy (highly viscous or solid glasses). Deconvolution of the observed, nonexponential kinetics required a continuous spectrum of rate constants. The average rate constant ( = 8.7 s(-1) in a 28% (w/w) trehalose solution) increases smoothly by increasing the trehalose/water ratio. In solid glasses, at trehalose/water ratios > or = 97%, an abrupt increase is observed ( = 26.6 s(-1) in the driest solid sample). A dramatic broadening of the rate distribution function parallels the above sudden increase. Both effects fully revert upon rehydration of the glass. We compared the kinetics observed at room temperature in extensively dried water-trehalose matrices with the ones measured in glycerol-water mixtures at cryogenic temperatures and conclude that, in solid trehalose-water glasses, the thermal fluctuations among conformational substates are inhibited. This was inferred from the large broadening of the rate constant distribution for electron transfer obtained in solid glasses, which was due to the free energy distribution barriers having become quasi static. Accordingly, the RC relaxation from dark-adapted to light-adapted conformation, which follows primary charge separation at room temperature, is progressively hindered over the time scale of P(+)Q(A)(-) charge recombination, upon decreasing the water content. In solid trehalose-water glasses the electron transfer process resulted much more affected than in RC dried in the absence of sugar. This indicated a larger hindering of the internal dynamics in trehalose-coated RC, notwithstanding the larger amount of residual water present in comparison with samples dried in the absence of sugar.  相似文献

Abstract. Patterns of β‐diversity in a highly diverse tropical dry forest tree community are described; the contribution of environmental heterogeneity and distance to β‐diversity was assessed. Significant differences in elevation, insolation, slope and soil water holding capacity (p < 0.01), variables related to water availability, were found among 830 m × 100 m transects laid along contrasting slopes of a system of three parallel microbasins. A gradient in elevation and insolation was found within north‐facing transects, among 10 m × 10 m sites; south‐facing transects showed an elevation gradient while crest transects showed a gradient in water holding capacity. In total 119 species were registered, with 27 to 64 species per transect, and 4 to 16 species per site. A large β‐diversity was found among and within transects; two indices of β‐diversity consistently showed a higher β‐diversity within transects than among them. Among transects, 64% of the variance in species composition could be attributed to the environmental variables; an additional 22% to the spatial distribution of sites. Within transects, 42% of the deviance in β‐diversity values was explained by insolation, and 19% by distance. β‐diversity increased with distance and with difference in insolation among sites; north‐facing transects, those with most contrasting insolation conditions, had the steepest increase in β‐diversity with distance. Such increase was clearly associated with changes in species composition, not with changes in species richness.  相似文献

Bouteloua gracilis (blue grama grass) native populations have been shown to be highly variable, however the genetic basis of this variability has not been well established. Determining the extent of genetic variability within and among plant populations have important repercussions for the management and conservation of species, and in particular for those subjected to intensive use such as forage plants. Using RAPD, this study was undertaken to investigate the genetic variability of four B. gracilis native populations developed in three grasslands and one shrubland at the southernmost part of the North American Graminetum in México. Significant differences in grass aboveground production were found among the study sites, while considerable genetic variation within each of the four blue grama populations evaluated was detected. The molecular analysis, based on 55 individuals, revealed a total of 108 scorable repeatable bands, with 99 of them being polymorphic (overall polymorphism= 91.7%). Within every population each individual was genetically distinct and no population-specific bands (fixed marker differences) were identified. Pair-wise Φ ST comparisons indicated that the four blue grama populations examined were significantly different in their genetic constitution (P<0.001). AMOVA revealed that most of the genetic variation detected in Bouteloua gracilis was explained by intra- (88.53%), rather than by inter-population (11.47%) differences. UPGMA based on the Φ ST values indicated that the blue grama population collected from the shrubland displayed the RAPD profiles that most differed among the study sites. Possible causes of these results could reside on intensive grazing reducing, and proper management conserving, the forage production and genetic diversity of blue grama native populations. Our results are consistent with previous studies analyzing population genetic variation in outcrossing grasses and, in particular, with ecological and cytological evidence for a high genetic variability in native populations of B. gracilis. The implications of our findings and prospective studies to be undertaken using molecular tools in the study of blue grama biology and ecology are discussed.
