Pachycondyla marginata (P.m.), a migratory and termitophageous ant, hunting only the termite species Neocapritermes opacus, migrates significantly oriented 13 degrees with respect to the magnetic North-South axis. Results of hysteresis curves at room temperature of four Pachycondyla marginata heads, thorax, pairs of antennae and abdomens, oriented parallel to the magnetic field, indicate that the antennae give the strongest saturation magnetization, suggesting this sensory organ as being also a magnetic sensory organ. The total saturation magnetization in a whole P.m. is composed by 42 +/- 3%, 24 +/- 3%, 19 +/- 3% and 15 +/- 3% of antennae, head, thorax and abdomen contributions, respectively. The abdomen hysteresis curve presents a wasp-waisted loop with Hcr/Hc of 4.75, characteristic of mixed magnetic systems.  相似文献

Plant genetic engineering has become an invaluable tool in plant research. Although plant transformation is a well-established technique, transgene expression is still unpredictable. Silencing may involve epigenetic modifications or nuclear and chromosomal localization of transgenes. In this way, understanding nuclear structure and organization is important not only for increasing our knowledge of fundamental aspects of the genome but also for taking the greatest advantage of inserting foreign genes and controlling their expression in biotechnological applications. Integrated approaches are clearly required in order to elucidate such complex processes. By combining the analysis of the physical position of transgenes with markers for epigenetic modifications in the plant genome we can better understand the factors affecting transgene expression levels and analyze the genomic environments of differentially expressed transgenes. Medicago truncatula Gaertn. has become a well-known model for the legume family and is used in studies ranging from nodulation to environmental stresses. More recently its use in biotechnology has been explored. In this report we describe the application of fluorescence in situ hybridization (FISH) to detect foreign DNA sequences and to determine the organization of the nucleolar organizer regions (NORs) genes in both metaphase chromosomes and interphase nuclei. We also studied chromatin distribution by immunodetection of epigenetic marks in M. truncatula interphase nuclei from tissue sections. We present evidence that M. truncatula is amenable to this kind of studies, which will in turn contribute to a better exploitation of biotechnology applications for this important plant family.  相似文献

The replication error status analysis of DNA, through microsatellite instability detection, has become an indispensable tool for hereditary non-polyposis colorectal cancer screening. This study investigated the microsatellite instability in Brazilian individuals presenting colorectal cancer. In this study, 66 patients were clinically analyzed according to Amsterdam II and Bethesda guidelines. Normal and tumour tissues were collected and analyzed for MSI degree according to molecular markers BAT25, BAT26, BAT40, APC–D5S346, D2S123, and D17S250. Eight patients (12.1%) fulfilled the Amsterdam II guidelines, and 15 (22.7%) met the Bethesda guidelines. BAT25 was the most sensitive marker (86.7%), while BAT26 was the least sensitive (66.7%). The specificity of both markers was 100%, but all of the markers must be used since the contribution of each marker to the sensitivity and specificity of the test is complementary. Proximal tumours were significantly predominant among RER+ patients. Conclusions: Patients with a family history of colorectal cancer with the tumour in the proximal colon must be screened to replication error status as early as possible in order to avoid the progression of the disease.  相似文献

Herbivory has significant impacts on individual plants and plant communities, both at ecological and evolutionary time scales. In this context, this study aims to evaluate herbivore damage and its relationship with leaf chemical and structural traits, nutritional status, and forest structural complexity along a successional gradient. We predicted that trees in early successional stages support conservative traits related to drought tolerance (high specific leaf mass and phenolics), whereas trees in light-limited, late successional stages tend to enhance light acquisition strategies (high nitrogen content). We sampled 261 trees from 26 species in 15 plots (50 × 20 m; five per successional stage). From each tree, twenty leaves were collected for leaf trait measures. Phenolic content increased whereas specific leaf mass and nitrogen content decreased from early to late stages. However, leaf damage did not differ among successional stages. Our results partially corroborate the hypothesis that early successional plants in tropical dry forests exhibit leaf traits involved in the conservative use of water. The unexpected decrease in nitrogen content along the chronosequence is likely related to the fact that thinner leaves with low specific leaf mass could have less nitrogen-containing mesophyll per unit area. Mechanisms affecting herbivory intensity varied across scales: at the species level, leaf damage was negatively correlated with tannin concentration and specific leaf mass; at the plot level, leaf damage was positively affected by forest structural complexity. Herbivory patterns in tropical forests are difficult to detect because abiotic factors and multiple top-down and bottom-up forces directly and indirectly affect herbivores.  相似文献

Physical training (PT) has been considered as a treatment in metabolic syndrome (MS), since it induces thermogenic activity in brown (BAT) and white (WAT) adipose tissues. We evaluated the therapeutic effect of PT on activity of WAT and BAT in rats with MS induced by high-fat diet (30% lard) for 13 weeks and submitted, for the last 6 weeks, to swimming or kept sedentary (SED) rats. MS-SED rats compared to control diet (CT-SED) rats showed low physical fitness and high levels of glucose, insulin, homeostasis evaluation of insulin resistance (HOMA-IR), homeostasis evaluation of the functional capacity of β-cells (HOMA-β), and blood pressure. The gastrocnemius muscle decreased in peroxisome proliferator-activated receptor gamma coactivator 1-alpha and beta (PGC-1α, PGC-1β), and uncoupled protein 2 and 3 (UCP2 and UCP3) expressions. Both WAT and BAT increased in the adipocyte area and decreased in blood vessels and fibroblast numbers. WAT increased in expression of pro-inflammatory adipokines and decreased in anti-inflammatory adipokine and adiponectin. WAT and gastrocnemius showed impairment in the insulin signaling pathway. In response to PT, MS rats showed increased physical fitness and restoration of certain biometric and biochemical parameters and blood pressure. PT also induced thermogenic modulations in skeletal muscle, WAT and BAT, and also improved the insulin signaling pathway. Collectively, PT was effective in treating MS by inducing improvement in physical fitness and interchangeable effects between skeletal muscle, WAT and BAT, suggesting a development of brown-like adipocyte cells.  相似文献

Since the early studies on the genetic effects of chemical and physical agents, species and clones of Tradescantia have been used as experimental subjects, by virtue of a series of favorable genetic characteristics. Bearing just six pairs (2n = 12) of large, easily observable chromosomes, cells from almost every part of the plant, from the root tips to the developing pollen tube, yield excellent material for cytogenetic studies.

As a consequence of the intensive use of Tradescantia in genetic studies, a series of genetic characteristics have been found that offer opportunities for the detection of agents affecting the stability of the genome. At least five such characteristics have been selected as endpoints for the establishment of assays to evaluate mutagenesis. Three of these, root-tip mitosis, pollen-tube, and microspore mitosis are essentially chromosome aberration assays, wherein one observes and evaluates the visible damage in the chromosomes. A fourth, the stamen-hair mutation assay (Trad-SHM), is a point mutation mitotic assay based on the expression of a recessive gene for flower color in heterozygous plants. The fifth assay is a cytogenetic test based on the formation of micronuclei (Trad-MCN) that result from chromosome breakage in the meiotic pollen mother cells.

This article examines the characteristics and fundamentals of the Trad-MCN and the Trad-SHM assays and reviews the results obtained to date with these systems in the assessment of environmental mutagenesis.  相似文献

Herbivores can be associated with distinct ontogenetic stages of their host in a nonseasonal, directional, and continuous pattern of colonization and extinction of species populations called ontogenetic succession, but the processes behind this pattern are still largely unknown. We used plants of Cryptocarya aschersoniana Mez (Lauraceae) belonging to different ontogenetic stages, to examine how the density of different gall‐inducing insects varies along the ontogeny of the host, and how gall density is influenced by mechanisms associated with host quality (plant height, plant shape, leaf area, specific leaf area, and hypersensitivity), and by mechanisms associated with their natural enemies (parasitoids, pathogens, and predators). In a remnant of Araucaria Forest, located in the São Francisco de Paula National Forest (Brazil), gall density (ind./100 g of leaf ) was obtained for 42 plants of C. aschersoniana divided into three height classes. Two galling species were recorded, showing quite distinct density patterns among height classes of C. aschersoniana. While Hymenoptera gall density decreased almost 50 times from small plants to canopy trees, Hemiptera gall density increased almost 10 times. Path analyses showed that Hymenoptera density was higher in smaller plants, independent of other host traits, while Hemiptera density was higher in plants exhibiting smaller leaves. Natural enemies were not detected in the Hemiptera population, and mortality rates due to predators, parasitoids, and pathogens did not affect Hymenoptera density. Processes associated with plant quality play the main role in generating the observed ontogenetic succession pattern.  相似文献
