Effects of chitooligosaccharides (COS) on the activity of peroxidase and its isoenzyme composition were investigated in cocultures of wheat calli (Triticum aestivum L.) with common bunt fungal pathogen (Tilletia caries (DC) Tul.). COS elevated the activity of readily soluble form of peroxidase: at concentrations of 0.01 and 1 mg/l, peroxidases with isoelectric points (pI) of about 3.5 and 9.8 and at concentration of 100 mg/l—those with pI of 4.3–5.2 were activated. It was shown that treatment with COS boosted the resistance of wheat calli to T. caries, inducing the accumulation in the cell wall of pathogen-specific isoperoxidases, especially its anionic isoform with pI ∼ 3.5. The ultimate indicator of defense responses is quick lignification of cultured cell walls under the effect of COS in the contamination area.  相似文献

The dataset on benthic fauna diversity obtained by ecological monitoring of lakes (Mari El Republic) was analyzed for applicability in water quality biological benchmark. The effect of several parameters on biodiversity were tested: (1) calculation errors in species number definition, (2) application of certain evaluation method of rank distribution parameters, (3) adequacy degree of distribution model, (4) evenness: diversity ratio, (5) hydrochemical parameters of lake, and (6) “sampling site effect” (natural reserve area impact). The biodiversity characteristics were suggested, which highly correlated to obtained data. The critical level for each characteristic was proposed to define favorable and adverse conditions of benthic communities. Practical recommendations were given for using studied parameters in the evaluation of the water quality biological benchmark in Mari El Republic.  相似文献

The nitrile hydratase isolated from Rhodococcus ruber strain gt1, displaying a high nitrile hydratase activity, was immobilized on unmodified aluminum oxides and carbon-containing adsorbents, including the carbon support Sibunit. The activity and operational stability of the immobilized nitrile hydratase were studied in the reaction of acrylonitrile transformation into acrylamide. It was demonstrated that an increase in the carbon content in the support led to an increase in the amount of adsorbed enzyme and, concurrently, to a decrease in its activity. The nitrile hydratase immobilized on Sibunit and carbon-containing aluminum α-oxide having a “crust” structure displayed the highest operational stability in acrylonitrile hydration. It was shown that the thermostability of adsorbed nitrile hydratase increased by one order of magnitude.  相似文献

Antibodies hydrolyzing myelin basic protein (MBP) can play an important role in the pathogenesis of multiple sclerosis (MS) and systemic lupus erythematosus (SLE). An immunoglobulin light chain phagemid library derived from peripheral blood lymphocytes of patients with SLE was used. Small pools of phage particles displaying light chains with different affinities for MBP were isolated by affinity chromatography on MBP‐Sepharose, and the fraction eluted with 0.5 M NaCl was used for preparation of individual monoclonal light chains (MLChs, 26–27 kDa). Seventy‐two of 440 individual colonies were randomly chosen, expressed in Escherichia coli in a soluble form, and MLChs were purified by metal chelating chromatography. Twenty‐two of 72 MLChs have high affinity and efficiently hydrolyze only MBP (not other control proteins) demonstrating various pH optima in a 5.7–9.0 range and different substrate specificity in the hydrolysis of four different MBP oligopeptides. Four MLChs demonstrated serine protease‐like and three thiol protease‐like activities, while 11 MLChs were metalloproteases. The activity of three MLChs was inhibited by both phenylmethylsulfonyl fluoride (PMSF) and Ethylenediaminetetraacetic acid (EDTA), two other by EDTA and iodoacetamide, and one by PMSF, EDTA, and iodoacetamide. The ratio of relative activity in the presence of Ca2+, Mg2+, Mn2+, Ni2+, Zn2+, Cu2+, and Co2+ was individual for each of 22 MLCh preparations. It is the first examples of human MLChs, which probably can possess two or even three different proteolytic activities. These observations suggest an extreme diversity of anti‐MBP abzymes in SLE patients. The immune systems of individual SLE patients can generate a variety of anti‐MBP abzymes, which can attack MBP of myelin‐proteolipid sheath of axons and play an important role in MS and SLE pathogenesis. It has been shown for the first time that polyclonal IgG abzymes (Abzs) with DNase activity from the sera of autoimmune‐prone MRL/MpJ‐lpr mice can be separated by isoelectric focusing into many subfractions having the isoelectric points (pI) from 4.5 to 9, with the maximal activity for Abzs with pI = 6.5–9.0. Affinity chromatography on DNA‐cellulose separated DNase IgGs into many subfractions demonstrating a range of affinities for DNA and different levels of the relative DNase activities (RDA) due to intrinsically bound metals and after addition of external Mg2+, Mn2+, Ca2+, and Mg2++Ca2+. Some fractions significantly increase RDAs in the presence of external ions (Mg2++Ca2+ > Mg2+ > Mn2+ > Ca2+), while each of this cofactor can also inhibit or have no influence on the RDAs of another fractions. It is known that complexes of DNA with histones and other proteins of apoptotic cells are the primary immunogens in systemic lupus erythematosus (SLE). Bovine serum albumin (BSA) and methylated BSA (mBSA) increase the RDAs of only some fractions, while have no effect or inhibit other IgG fractions. The ratio of the RDAs in the presence of all metal ions, BSA, and mBSA was individual for every abzyme fraction. Mn2+ and Ca2+ stimulated accumulation of only relaxed form of supercoiled DNA (scDNA) in the case of all subfractions, while in the presence of Mg2+ antibodies (Abs) of some subfractions (and in the presence of Mn2+ +Ca2+ all subfractions) produced relaxed DNA (rDNA) and linear DNA (linDNA) in a variable extent. The data obtained show that the polyclonal Abzs of mice may be a cocktail of Abs directly to DNA, RNA, and their complexes with proteins and anti‐idiotypic Abs to active centers of different nucleases. The diversity of the physicochemical and kinetic characteristics of the Abzs seems to be significantly widened when pre‐diseased mice spontaneously develop the disease. Human immunodeficiency virus type 1 integrase (IN) catalyzes integration of a DNA copy of the viral genome into the host genome. It was shown previously that IN preincubation with various oligodeoxynucleotides (ODNs) induces formation of dimers and oligomers of different gyration radii; only specific ODNs stimulate the formation of catalytically active dimers. Here we have shown that preincubation of IN with specific and nonspecific ODNs leads to a significant and comparable decrease in its hydrolysis by chymotrypsin, while nonspecific ODNs protect the enzyme from the hydrolysis by trypsin worse than specific ODNs; all ODNs had little effect on the IN hydrolysis by proteinase K. In contrast to canonical proteweases, IgGs from HIV‐infected patients specifically hydrolyze only IN. While d(pT)n markedly decreased the IgG‐dependent hydrolysis of IN, d(pA)n and d(pA)n?d(pT)n demonstrated no detectable protective effect. The best protection from the hydrolysis by IgGs was observed for specific single‐ and especially double‐stranded ODNs. Although IN was considerably protected by specific ODNs, proteolytic IgGs and IgMs significantly suppressed both 3′‐processing and integration reaction catalyzed by IN. Since anti‐IN IgGs and IgMs can efficiently hydrolyze IN, a positive role of abzymes in counteracting the infection cannot be excluded. Copyright © 2011 John Wiley & Sons, Ltd.  相似文献

Arabidopsis thaliana has three membrane‐located cytokinin receptors (AHK2, AHK3 and CRE1/AHK4), which are sensor histidine kinases containing a ligand‐binding CHASE domain. Despite their structural similarity the role of these receptors differs in planta. Here we have explored which parameters contribute to signal specification. In a bacterial assay, the CHASE domain of AHK2 has a similar ligand binding spectrum as CRE1/AHK4. It shows the highest affinity for isopentenyladenine (iP) and trans‐zeatin (tZ) with an apparent KD of 1.4 and 4.0 nm , respectively. Real‐time PCR analysis of cytokinin primary response genes in double mutants retaining only single receptors revealed that all receptors are activated in planta by cytokinin concentrations in the low nanomolar range. However, there are differences in sensitivity towards the principal cytokinins iP and tZ. The activation of the cytokinin‐sensitive PARR5:GUS reporter gene in three different double mutants shows specific, but also overlapping, spatial domains of activity, which were for all receptors predominantly in the shoot apical meristems and root cap columella. AHK2 and AHK3 signal specifically in leaf parenchyma cells, AHK3 in stomata cells, and CRE1/AHK4 in the root vasculature. Promoter‐swap experiments demonstrate that CRE1/AHK4 can functionally replace AHK2 but not AHK3. However, the cytoplasmic AHK3 histidine kinase (Hk) domain can be replaced by the CRE1/AHK4 Hk domain, which suggests that functionality is mediated in this case by the extracytosolic domain. Together, the data show that both differential gene expression and ligand preference contribute to specify the receptor activity.  相似文献

The effects of salicylic acid (SA) and jasmonic acid (JA) on plant defense responses were studied with aseptic potato (Solanum tuberosum L.) plantlets infected with Phytophthora infestans (Mont.) de Bary. Plant treatment with 10−6 M SA or 10−7 M JA induced plant resistance; the mixture of these acids was most efficient. After treatment with these compounds, phenolic compounds were accumulated and peroxidase was activated in the sites of pathogen localization, and this might be the reason of resistance enhancement. In addition, more H2O2 was accumulated in infected plants treated with JA or its mixture with SA but not in plants treated with SA alone. It might occur because of observed inhibition of catalase and activation of isoperoxidase with the isoelectric point (pI) of ∼9.3, which manifests an affinity for the pathogen cell wall. The data obtained allow us to recommend the application of these compounds for potato plant protection against late blight.  相似文献
