1. A method was developed to examine the turnover of nitrate reductase by the use of tungstate. 2. Evidence is presented which suggests that the disappearance of nitrate reductase activity from Neurospora mycelia exposed to non-inducing conditions is due to the disappearance of the enzyme protein(s) from the mycelia, and not merely due to the disappearance of its (their) catalytic power. 3. The presence of NO(3) (-) in the culture medium slows down the rate of degradation of nitrate reductase in Neurospora in vivo. 相似文献

The ultraviolet absorption, linear dichroism, circular dichroism, and oriented circular dichroism of collagen are reported and the spectra are resolved into a self-consistent set of bands in accord with exciton theory. The parallel band at 200 nm has 40% of the π → π* intensity; the perpendicular band is placed at 189 nm yielding a splitting of 2700 cm?1. The circular dichroism is resolved into two Gaussians at λ∥ and λτ (rotational strengths +14 × 10?40 and ?32 × 10?40 esu2. cm2) plus a large non-Gaussian (“helix”) band with ampplitude ?25,000° at 201 nm. These data appear to be in reasonably good accord with recent calculations. Measurements of the absorption, linear dichroism and circular dichroism of polyproline I and II are also reported and are resolved into their component bands. Seedlings of Datura stramonium L., although not photoperiodically sensitive, are useful for floral transition studies when raised in a growth chamber at a constant temperature of 25 C with a photoperiod of 8 hr of light (1,600-2,000 ft-c) and 16 hr of darkness. A terminal flower is formed after the seventh or eighth leaf primordium is produced. A constant rate of leaf initiation up to the time of flowering enables specific apical stages to be obtained and studied. Changes in the mitotic index, substantiated with calculated rates of cell division (measured by the accumulation of metaphases following treatment with colchicine) were studied in shoot apical zones during transition to flowering. Fluctuations in the mitotic index of each zone in the vegetative and transition apex with respect to apical stage as well as time of day were not statistically significant. The mitotic index of the summit zone of the vegetative apex was significantly lower than in the other zones whose mitotic indices were not significantly different from one another. During floral transition the mitotic index of the summit zone as well as the central zone (just below the summit zone) significantly increased while no significant changes were detected in the flank zones. It was shown that the mitotic index could be considered representative of the rates of cell division in Datura. 相似文献

Field observations of changes in the populations of aquatic weeds and phytoplankton have confirmed that aquatic weeds have antagonistic activity toward phytoplankton. Nutritional studies in the laboratory indicate that cultures of the aquatic weeds, Myriophyllum sp., Ceratophyllum sp., and duckweed (Lemma minor L.); liquid cultures of barley (Hordeum vulgare L., Dickson variety); and cultures of the filamentous green algae, Cladophora sp. and Pithophora oedogonium (Mont.) Withrock, will remain relatively free of epiphytes or competing phytoplankton if the cultures are nitrogen-limited. Field observations of Cladophora sp. have confirmed that the growth of epiphytes on the Cladophora is related to conditions of surplus available nitrogen compounds. It is proposed that this antagonistic activity may be due to a “nitrogen sink” effect in which the aquatic weeds or filamentous green algae prevent the growth of contaminating algae by competition for the limited nitrogen compounds available. However, the presence of bacteria-sized organisms which have selective toxicity to certain algae indicates that perhaps multiple factors exist. Discussed are the ecological implications of associations of certain algae with bacteria that have selective toxicities for other species of algae under certain environmental conditions such as nitrogen-limited growth. 相似文献

Inhibitors of hemagglutination by type A2 influenza virus and a recently isolated strain of type B influenza virus were separated by sucrose density gradient centrifugation and agarose gel filtration from horse serum. Using selected reagents, it was demonstrated that the active substituent on the horse serum inhibitor of A2 influenza virus was 4-O-acetyl-N-acetylneuraminic acid; however, the active substituent on the inhibitor of the influenza B virus was shown to be N-acetylneuraminic acid (NANA). Sodium metaperiodate treatment of a component of horse serum resulted in a 10 to 15-fold enhancement of inhibitory activity against the type B virus, whereas the A2 inhibitor was completely destroyed. Since this enhancement did not occur with influenza B viruses isolated prior to 1965, it was considered that this sensitivity to an oxidized NANA glycoside may have been a reflection of an antigenic change which occurred at that time. The use of different virus strains and selected chemical reagents to define the important sialic acid prosthetic groups active in inhibition was described. 相似文献

Summary Negative staining showed the presence of microcrystals in various polysaccharides. Cellulose microcrystals from Valoniopsis, Vaucheria, and an unidentified tunicate had widths of 20, 27, and 30 Å, respectively. Mannan microcrystals from Acetabularia were 10x25 Å and were oriented in linear arrays with their long axis perpendicular to the array axis. dichotomosiphon and Caulerpa xylans had respective microcrystal widths of 22 and 24 Å. Summary In negatively stained preparations the cellulose of Dictyostelium discoideum appears in the form of 35 Å wide fibrils of undetermined length. Upon mild acid hydrolysis a periodic pattern may be observed along the fibrils, in the form of fine, electron-dense bands across the full diameter of the fibril spaced apart from each other by electron-transparent segments approximately 100 Å long.
