The results presented in this paper show that lysis of human erythrocytes by linoleic acid is not caused by peroxidation of the fatty acid. Peroxidase, superoxide dismutase and scavengers of O 2 and OH had no effect on the lysis while catalase showed only marginal inhibition suggesting that O 2 , OH, O 2 and H2O2 do not play any direct role in hemolysis by linoleic acid. Generators of H2O2 inhibited the lysis completely and methemoglobin cells were more resistant to hemolysis by linoleic acid. The fatty acid did neither bind to nor fomed complex with red cell ghosts. Membrane oxidation of sulphydryl groups was also not involved in the lysis. Β-Carotene, retinol and bile salts enhanced the lysis, while, cholesterol but not cholesterol acetate, inhibited it. Taurocholate-pretreated cells were more susceptible to linoleic acid lysis. These observations suggested-that lysis by linoleic acid may be due to its detergent property.  相似文献

Terminal labeling of embryonic feather keratin mRNA with [3H]KBH4 followed by digestion with ribonuclease T1 and T2, alkaline phosphatase, snake venom phosphodiesterase, and nucleotide pyrophosphatase and analysis of the products by high voltage paper electrophoresis, indicated the presence of the sequence m7G(5)ppp(5)N at the 5-end of the mRNA. Ribonuclease T1 and A digests of the terminally labeled, and also of unlabeled mRNA followed by fractionation on denaturing polyacrylamide gels indicated the presence of polyadenylate tracts ranging in size from 45 to 165 nucleotides at the 3-end of the mRNA.  相似文献

DAPI (4'-6-diamidino-2-phenylindole), a fluorochrome specific for AT-rich DNA, was supplied for 24 h at various concentrations to human leukocytes in culture. This treatment caused the appearance on the chromosomes of specific areas lacking spiralization. In particular, the centromeric regions of chromosomes 1,9, and 16, a short region on the long arm of chromosomes 1 and 2, and the distal heterochromatic part of the long arm of the Y chromosome were despiralized. The despiralization pattern of DAPI is compared with those previously obtained with Hoechst 33258 and Distamycin A.  相似文献

A new virus, Mycoplasmatales virus-modicum 1 (MV-M1), was recovered from spontaneous plaques in lawns ofAcholeplasma modicum. Strain “mod” produced plaques onA. modicum strains but not on strains ofAcholeplasma laidlawii. Only MV-L3 of the three knownA. laidlawii viruses (MV-L1, MV-L2, and MV-L3) produced plaques onA. modicum. The MV-M1 virus was serologically distinct from the threeA. laidlawii viruses; filterable at 0.1 μm; partially sensitive to heat and Nonidet P-40; and chloroform labile. Spherical particles ranging from 105 to 160 nm were observed in electron micrographs of negatively stained preparations.  相似文献

Summary High-level mechanosensory interneurons integrate a substantial amount of polysynaptic input. We have used an identified interneuron, the crayfish (Procambarus clarki) caudal photoreceptor (CPR), to examine the extent and specificity of interneuronal input as received by a physiologically complex, smalldiameter sensory unit. Presynaptic central neurons were identified by antidromic stimulation of connective fibers and characterized physiologically relative to the bilateral responses observed in the paired CPR's. Eleven inhibitory interneurons have been identified, including cells with ipsilateral (Fig. 4), contralateral (Fig. 5) and bilateral effects (Fig. 6). Seven excitatory interneurons have been identified, including examples from each of the respective categories above (Figs. 7–9). The results of this survey are summarized in Table 1; axon locations are presented in Fig. 10.It has also been demonstrated that several of these fibers are themselves ascending mechanoreceptive interneurons (e.g., fiber 122, Fig. 1). Thus, for the encoding of environmental stimuli, these results indicate that central integration involves a lateral exchange of tactile information among a set of interrelated sensory interneurons. Summary Two 46,XX sibs, one of female, one of male gender, and both with ambiguous external genitalia and ovotestis, were H-Y positive. The mother was H-Y negative. It is assumed that the underlying mutation was transmitted by the father, resulting in an autosomal dominant mode of inheritance. Hospital activity analysis (HAA) was used to identify 2151 patients in whom the diagnostic code fell within the infectious disease categories of the International Classification of Disease. The clinical notes of these patients were examined by a doctor and in 414 (19.2%) the diagnostic coding was incorrect. Diarrhoea and viral infections were miscoded most often. The possibility of inaccuracy must be considered when HAA aggregated data are used for planning purposes. Accuracy could be improved if clinicians routinely completed the diagnostic sections of the HAA form.  相似文献

Displacement     
Displacement is a noncommital term for the reactions that occur when slides previously stained in phloxine or rose Bengal are immersed for varying lengths of time in a solution of another dye in ethyl Cellosolve. In most histotechnic tests Lendrum's (1947) phloxine-tartrazine is given as the stain for acidophilic inclusion bodies. However the lack of contrast between the phloxine and tartrazine has been a serious limitation. A number of dyes were tried as possible substitutes for the tartrazine. A rose Bengal-Bismark brown Y procedure was developed which stains similarly to Lendrum's phloxine-tartrazine and which doer have the needed contrast. After staining for 10 min in 1% aqueous rose Bengal and rinsing in isopropyl alcohol slides are placed for 20, 30, 40 and 50 min in 0.05% Bismark brown Y in ethyl Cellosolve. In various tissues and structures the rose Bengal is sequentially displaced by the Bismark brown Y. Thus collagen loses the red stain after 30 min while acidophilic structures like sperm heads and Paneth cell granules retain the red stain after 50 min in the displacement solution. The results are strikingly similar to staining with alkaline Biebrich scarlet.  相似文献
