Two kinds of bioelectronic gas sensors (bio-sniffer) incorporating alcohol oxidase (AOD) and aldehyde dehydrogenase (ALDH) were developed for the convenient analysis of ethanol and acetaldehyde in expired gas, respectively. The sniffer devices for gaseous ethanol and acetaldehyde were constructed by immobilizing enzyme on electrodes covered with filter paper and hydrophilic PTFE membrane, respectively. The AOD and ALDH sniffers were used in the gas phase to measure ethanol vapor from 1.0 to 500 ppm, and acetaldehyde from 0.11 to 10 ppm covering the concentration range encountered in breath after alcohol consumption. Both bio-sniffers displayed good gas selectivity which was attributed to the substrate specificity of the relevant enzymes (AOD and ALDH) as gas recognition material. From the results of physiological application, the bio-sniffers could monitor the concentration changes in breath ethanol and acetaldehyde after drinking. The ethanol and acetaldehyde concentrations in expired air from ALDH2 [-] (aldehyde dehydrogenase type 2 negative) subjects were higher than that of the ALDH2 [+] (positive) subjects. The results indicated that the lower activity of ALDH2 induced an adverse effect on ethanol metabolism, leading to ethanol and acetaldehyde remaining in the human body, even human expired air. 相似文献

Spatial expansion and increased population abundance of masu salmon Oncorhynchus masou have been recently reported following habitat restoration in some rivers, but no studies have examined these types of changes under natural conditions. We found spatial expansion and increased population density of masu salmon parr in rivers of eastern Hokkaido where no habitat restoration had occurred during the period examined. Changes in the distribution and population densities of parr are often regarded as effects of river habitat destruction and restoration, yet such changes should be evaluated while taking into account the effects of other ocean environmental conditions as well. 相似文献

As a therapeutic treatment, recombinant human basic fibroblast growth factor (rhbFGF) is usually employed in tissue regeneration, and as an essential component in culture medium for maintaining the induced pluripotent stem (iPS) cell and embryonic stem (ES) cell in an undifferentiated state. Therefore, a large amount of biologically active rhbFGF is required. In this study, silkworm-baculovirus expression vector system (silkworm-BEVS) is employed to achieve a high productivity of recombinant rhbFGF with two small affinity tags (His-tag and STREP-tag) at the N or C-terminus. It is observed that rhbFGF with 30?K signal peptide of silkworm were successfully expressed but are not sufficiently secreted into the culture medium of cultured insect cells. Then we purified the N- or C-tagged intracellular rhbFGF protein and obtained a yield of about 0.7?mg/larva and 1.2?mg/larva, respectively. Although the final yield of the C-tagged rhbFGF is higher than that of the N-tagged, rhbFGF with N-tag demonstrated promising and comparable biological activity, which is evaluated through a mammalian cell proliferation assay. Taken together, these results indicate that silkworm-BEVS could contribute to the mass-production of the biologically active rhbFGF for medical uses. 相似文献

Vertical distribution of organic constituents, i.e. total organic carbon (TOC), hydrocarbons, fatty acids and hydroxy acids in water and sediment samples from Lake Fryxell (77° 35 S, 163° 15 E) of southern Victoria Land, Antarctica were studied to elucidate their features in relation to stratification of the lake waters and likely distribution of microorganisms. The TOC content of the surface water (5.0 m; just below the ice cover of 4.50 m thickness) was 1.4 mg l–1. It increased markedly with depth and attained a maximum value of 21.7 mg C l–1 at a depth of 17.5 m, but decreased to the bottom (13.3 mg C l–1). The high TOC content of the anoxic bottom layers (> 15 m) is attributable to the concentration of refractory organic substances over long periods following the degradation of labile organic constituents. Hydrocarbons were not found in the water column, but the major constituent of the bottom sediment was n-C29 : 2 alkene. Total concentrations of fatty acids in the oxic layers ( 10 m) were highest at 10.0 m and much higher than those in the anoxic layers (> 10 m), probably reflecting the phytoplankton population. The content of branched (iso and anteiso) fatty acids and 3-hydroxy acids in the anoxic layers were much greater than those in the oxic layers which would seem to reflect the distribution of bacterial abundance. The differences of organic composition between the water column and sediments imply that sinking dead organisms were quickly degraded in the lake bottom. Also, the composition of microorganisms in the water column must be very different from that in the sediments. 相似文献

Cooperation is one of the essential factors for all biological organisms in major evolutionary transitions. Recent studies have investigated the effect of migration for the evolution of cooperation. However, little is known about whether and how an individuals’ cooperativeness coevolves with mobility. One possibility is that mobility enhances cooperation by enabling cooperators to escape from defectors and form clusters; the other possibility is that mobility inhibits cooperation by helping the defectors to catch and exploit the groups of cooperators. In this study we investigate the coevolutionary dynamics by using the prisoner’s dilemma game model on a lattice structure. The computer simulations demonstrate that natural selection maintains cooperation in the form of evolutionary chasing between the cooperators and defectors. First, cooperative groups grow and collectively move in the same direction. Then, mutant defectors emerge and invade the cooperative groups, after which the defectors exploit the cooperators. Then other cooperative groups emerge due to mutation and the cycle is repeated. Here, it is worth noting that, as a result of natural selection, the mobility evolves towards directional migration, but not to random or completely fixed migration. Furthermore, with directional migration, the rate of global population extinction is lower when compared with other cases without the evolution of mobility (i.e., when mobility is preset to random or fixed). These findings illustrate the coevolutionary dynamics of cooperation and mobility through the directional chasing between cooperators and defectors. 相似文献

Spinal and bulbar muscular atrophy (SBMA) is an inherited motor neuron disease caused by the expansion of a polyglutamine (polyQ) tract within the androgen receptor (AR) gene. The pathologic features of SBMA are motor neuron loss in the spinal cord and brainstem, and diffuse nuclear accumulation and nuclear inclusions of mutant AR in residual motor neurons and certain visceral organs. AR‐associated coregulator 70 (ARA70) was the first coregulator of AR to be identified, and it has been shown to interact with AR and increase its protein stability. Here, we report that genistein, an isoflavone found in soy, disrupts the interaction between AR and ARA70 and promotes the degradation of mutant AR in neuronal cells and transgenic mouse models of SBMA. We also demonstrate that dietary genistein ameliorates behavioral abnormalities, improves spinal cord and muscle pathology, and decreases the amounts of monomeric AR and high‐molecular‐weight mutant AR protein aggregates in SBMA transgenic mice. Thus, genistein treatment may be a potential therapeutic approach for alleviating the symptoms of SBMA by disrupting the interactions between AR and ARA70. 相似文献
