Protein phosphorylation is reversibly regulated by the interplay between kinases and phosphatases. Recent developments within the field of proteomics have revealed the extent of this modification in nature. To date there is still a lack of information about phosphatase specificity for different proteomes and their conditions to achieve maximum enzyme activity. This information is important per se, and in addition often requested in functional and biochemical in vitro studies, where a dephosphorylated sample is needed as a negative control to define baseline conditions. In this study, we have addressed the effectiveness of two phosphatases endogenously present in the heart (protein phosphatases 1 and 2A) and two generic phosphatases (alkaline phosphatase and lambda protein phosphatase) on three cardiac subproteomes known to be regulated by phosphorylation. We optimized the dephoshorylating conditions on a cardiac tissue fraction comprising cytosolic and myofilament proteins using 2DE and MS. The two most efficient conditions were further investigated on a mitochondrial-enriched fraction. Dephosphorylation of specific proteins depends on the phosphatase, its concentration, as well as sample preparation including buffer composition. Finally, we analyzed the efficiency of alkaline phosphatase, the phosphatase with the broadest substrate specificity, using TiO(2) peptide enrichment and 2DLC-MS/MS. Under these conditions, 95% of the detected cardiac cytoplasmic-enriched phospho-proteome was dephosphorylated. In summary, targeting dephosphorylation of the cardiac muscle subproteomes or a specific protein will drive the selection of the specific phosphatase, and each requires different conditions for optimal performance.  相似文献

Sarco(endo)plasmic reticulum calcium ATPases (SERCA) are cellular pumps that transport Ca2+ into the sarcoplasmic reticulum (SR). Serca2 is the most widely expressed gene family member. The very early embryonic lethality of Serca2null mouse embryos has precluded further evaluation of loss of Serca2 function in the context of organ physiology. We have generated mice carrying a conditional Serca2flox allele which allows disruption of the Serca2 gene in an organ-specific and/or inducible manner. The model was tested by mating Serca2flox mice with MLC-2vwt/Cre mice and with αMHC-Cre transgenic mice. In heterozygous Serca2wt/floxMLC-2vwt/Cre mice, the expression of SERCA2a and SERCA2b proteins were reduced in the heart and slow skeletal muscle, in accordance with the expression pattern of the MLC-2v gene. In Serca2flox/flox Tg(αMHC-Cre) embryos with early homozygous cardiac Serca2 disruption, normal embryonic development and yolk sac circulation was maintained up to at least embryonic stage E10.5. The Serca2flox mouse is the first murine conditional gene disruption model for the SERCA family of Ca2+ ATPases, and should be a powerful tool for investigating specific physiological roles of SERCA2 function in a range of tissues and organs in vivo both in adult and embryonic stages.  相似文献

Ligand-induced phosphorylation of the receptor-regulated Smads (R-Smads) is essential in the receptor Ser/Thr kinase-mediated TGF-beta signaling. The crystal structure of a phosphorylated Smad2, at 1.8 A resolution, reveals the formation of a homotrimer mediated by the C-terminal phosphoserine (pSer) residues. The pSer binding surface on the MH2 domain, frequently targeted for inactivation in cancers, is highly conserved among the Co- and R-Smads. This finding, together with mutagenesis data, pinpoints a functional interface between Smad2 and Smad4. In addition, the pSer binding surface on the MH2 domain coincides with the surface on R-Smads that is required for docking interactions with the serine-phosphorylated receptor kinases. These observations define a bifunctional role for the MH2 domain as a pSer-X-pSer binding module in receptor Ser/Thr kinase signaling pathways.  相似文献

A central theme in life history theory is to determine how reproduction varies with age in iteroparous organisms. Evidence of ageing and senescence, defined as the progressive loss of function accompanied by decreased performance with age, remains poorly documented for large herbivores, in particular as it relates to reproduction. Analyses of body weight of 87 532 domestic sheep lambs demonstrates that onset of reproductive senescence in ewes occurs already at 5 and 6 years of age when measured, respectively, as lamb weight and litter size produced. This provides convincing evidence of early onset of reproductive senescence in this highly domesticated sheep breed. As this is earlier than indicated for other Ovis species as well as for the Soay sheep, an ancient and lightly domesticated sheep, we hypothesize that there may be a cost of selection for large litter size in mammalian herbivores.  相似文献

This study was designed to determine the spring, summer, autumn, and early winter distribution, migration routes, and timing of migration of the Atlantic population of Steller's eiders Polysticta stelleri . Satellite transmitters were implanted in 20 eiders captured in April 2001 at Vadsø, Norway, and their locations were determined from 5 May 2001 to 6 February 2002. Regions where birds concentrated from spring until returning to wintering areas included coastal waters from western Finnmark, Norway, to the eastern Taymyr Peninsula, Russia. Novaya Zemlya, Russia, particularly the Mollera Bay region, was used extensively during spring staging, moult, and autumn staging; regions of the Kola, Kanin, and Gydanskiy peninsulas, Russia, were used extensively during spring and moult migrations. Steller's eiders migrated across the Barents and Kara seas and along the Kara Sea and Kola Peninsula coastal waters to nesting, moulting, and wintering areas. The majority of marked eiders (9 of 15) were flightless in near-shore waters along the west side of Novaya Zemlya. Eiders were also flightless in northern Norway and along the Kanin and at Kola Peninsula coasts. We compare and contrast natural history characteristics of the Atlantic and Pacific populations and discuss evolutionary and ecological factors influencing their distribution.  相似文献

A case of intracellular systemic infection with the diplomonad flagellate Spironucleus barkhanus in farmed Arctic char Salvelinus alpinus is described. The parasites were widely disseminated throughout the vasculature and in most organs. Aggregates of the parasites were seen within well-defined structures regarded as host cells in capillaries and sinusoids of the liver, spleen and head kidney. Intracellular infection with Spironucleus spp. has never previously been reported. The prevalence of infection and mortality in the affected farm was low. In contrast to systemic spironucleosis in farmed Atlantic salmon, and despite huge numbers of flagellates in the vasculature, the tissues of the organs were remarkably unaffected. The relatively few gross and histopathological lesions may indicate that Arctic char are more tolerant to this parasite than Atlantic salmon.  相似文献

The type I TGF beta receptor (T beta R-I) is activated by phosphorylation of the GS region, a conserved juxtamembrane segment located just N-terminal to the kinase domain. We have studied the molecular mechanism of receptor activation using a homogeneously tetraphosphorylated form of T beta R-I, prepared using protein semisynthesis. Phosphorylation of the GS region dramatically enhances the specificity of T beta R-I for the critical C-terminal serines of Smad2. In addition, tetraphosphorylated T beta R-I is bound specifically by Smad2 in a phosphorylation-dependent manner and is no longer recognized by the inhibitory protein FKBP12. Thus, phosphorylation activates T beta R-I by switching the GS region from a binding site for an inhibitor into a binding surface for substrate. Our observations suggest that phosphoserine/phosphothreonine-dependent localization is a key feature of the T beta R-I/Smad activation process.  相似文献
