In systemic lupus erythematosus (SLE) it has been hypothesized that self-reactive B cells arise from virgin B cells that express low-affinity, nonpathogenic germline V genes that are cross-reactive for self and microbial antigens, which convert to high-affinity autoantibodies via somatic hypermutation. The aim of the present study was to determine whether the VH family repertoire and pattern of somatic hypermutation in germinal centre (GC) B cells deviates from normal in SLE. Rearranged immunoglobulin VH genes were cloned and sequenced from GCs of a SLE patient's spleen. From these data the GC V gene repertoire and the pattern of somatic mutation during the proliferation of B-cell clones were determined. The results highlighted a bias in VH5 gene family usage, previously unreported in SLE, and under-representation of the VH1 family, which is expressed in 20–30% of IgM+ B cells of healthy adults and confirmed a defect in negative selection. This is the first study of the splenic GC response in human SLE.  相似文献

Species that are dispersed across oceanic islands can have strong population structure due to genetic isolation, which makes it difficult to determine realistic and meaningful species boundaries. This becomes especially problematic when pest species are involved, and can result in undetected new invasions. The mango fruit fly, Bactrocera frauenfeldi (Schiner), is currently considered to be one of five morphologically similar members in a monophyletic species group distributed across Southeast Asia, Australasia, and Oceania, including three major pests. We used a phylogenomic approach with highly multiplexed amplicon sequencing to test species limits and evaluate the relationships among species in the B. frauenfeldi species complex and two closely related species. We obtained sequence data from 196 specimens for 395 nuclear DNA loci, totalling 102 kb, of which 2.2 kb were parsimony informative sites. Based on morphology, biogeography, and phylogenetic analyses, we conclude that there are five distinct species in the complex in our phylogeny. Our results show that the morphological differences between B. frauenfeldi and B. albistrigata (de Meijere) are part of a continuum that cannot be phylogenetically separated into monophyletic groups. We therefore synonymize the names of two major pests: B. albistrigata syn. rev. with B. frauenfeldi, making B. frauenfeldi now recognized as a widespread pest across Australasia and Southeast Asia. We evaluated the use of COI for pest recognition and conclude that it cannot reliably distinguish between six of the seven species we studied, thus new molecular approaches will be necessary for effective management and the prevention of incursions.  相似文献

Residential development of lakeshores is expected to change a variety of key lake features that include increased nutrient loading, increased invasion rate of nonnative species, increased exploitation rates of fishes by anglers, and alteration of littoral habitats. All of these factors may alter the capacity of lakes to support productive native fish populations. Fourteen north temperate lakes were surveyed to examine how growth rates of two common fish species (bluegill sunfish, Lepomis macrochirus; largemouth bass, Micropterus salmoides) varied along a residential development gradient. Size-specific growth rates for both species were negatively correlated with the degree of lakeshore residential development, although this trend was not statistically significant for largemouth bass. On average, annual growth rates for bluegill sunfish were 2.6 times lower in heavily developed lakes than in undeveloped lakes. This effect of lakeshore development on fish growth was not size specific for bluegills between 60 and 140 mm in total length. An index of population production rate that accounted for both the size-specific growth rate and the size distribution of fishes showed that bluegill populations were approximately 2.3 times less productive in highly developed lakes than in undeveloped lakes. Our results suggest that extensive residential development of lakeshores may reduce the fish production capacity of aquatic ecosystems. Received 29 April 1999; Accepted 26 October 1999.  相似文献

In skeletal muscle, excitation-contraction coupling involves a functional interaction between the ryanodine receptor (RyR) and the dihydropyridine receptor (DHPR). The domain corresponding to Thr671-Leu690 of the II-III loop of the skeletal DHPR α1-subunit is able to regulate RyR properties and calcium release from sarcoplasmic reticulum, whereas the domain corresponding to Glu724-Pro760 antagonizes this effect. Two peptides, covering these sequences (peptide ASk and CSk, respectively) were immobilized on polystyrene beads. We demonstrate that peptide ASk binds to the skeletal isoform of RyR (RyR1) whereas peptide CSk does not. Using surface plasmon resonance detection, we show that 1) domain Thr671-Leu690 is the only sequence of the II-III loop binding with RyR1 and 2) the interaction of peptide ASk with RyR1 is not modulated by Ca2+ (pCa 9-2) nor by Mg2+ (up to 10 mM). In contrast, this interaction is strongly potentiated by the immunophilin FKBP12 (EC50 = 10 nM) and inhibited by both rapamycin (IC50 = 5 nM) and FK506. Peptide ASk induces a 300% increase of the opening probability of the RyR1 incorporated in lipid bilayer. Removal of FKBP12 from RyR1 completely abolishes this effect of domain ASk on RyR1 channel behavior. These results demonstrate a direct interaction of the RyR1 with the discrete domain of skeletal DHPR α1-subunit corresponding to Thr671-Leu690 and show that the association of FKBP12 with RyR1 specifically modulates this interaction.  相似文献

Sec8 is highly expressed in mammalian nervous systems and has been proposed to play a role in several aspects of neural development and function, including neurite outgrowth, calcium-dependent neurotransmitter secretion, trafficking of ionotropic glutamate receptors and regulation of neuronal microtubule assembly.
