Summary . Spatial clustering is commonly modeled by a Bayesian method under the framework of generalized linear mixed effect models (GLMMs). Spatial clusters are commonly detected by a frequentist method through hypothesis testing. In this article, we provide a frequentist method for assessing spatial properties of GLMMs. We propose a strategy that detects spatial clusters through parameter estimates of spatial associations, and assesses spatial aspects of model improvement through iterated residuals. Simulations and a case study show that the proposed method is able to consistently and efficiently detect the locations and magnitudes of spatial clusters. 相似文献

A bioreactor system composed of a stirred tank and three tubular bioreactors in series was established, and continuous ethanol fermentation was carried out using a general Saccharomyces cerevisiae strain and a very high gravity medium containing 280 g L(-1) glucose, supplemented with 5 g L(-1) yeast extract and 3 g L(-1) peptone. Sustainable oscillations of glucose, ethanol, and biomass were observed when the tank was operated at the dilution rate of 0.027 h(-1), which significantly affected ethanol fermentation performance of the system. After the tubular bioreactors were packed with 1/2' Intalox ceramic saddles, the oscillations were attenuated and quasi-steady states were achieved. Residence time distributions were studied for the packed bioreactors by the step input response technique using xylose as a tracer, which was added into the medium at a concentration of 20 g L(-1), indicating that the backmixing alleviation assumed for the packed tubular bioreactors could not be established, and its contribution to the oscillation attenuation could not be verified. Furthermore, the role of the packing's yeast cell immobilization in the oscillation attenuation was investigated by packing the tubular bioreactors with packings with significant difference in yeast cell immobilization effects, and the experimental results revealed that only the Intalox ceramic saddles and wood chips with moderate yeast cell immobilization effects could attenuate the oscillations, and correspondingly, improved the ethanol fermentation performance of the system, while the porous polyurethane particles with good yeast cell immobilization effect could not. And the viability analysis for the immobilized yeast cells illustrated that the extremely lower yeast cell viability within the tubular bioreactors packed with the porous polyurethane particles could be the reason for their inefficiency, while the yeast cells loosely immobilized onto the surfaces of the Intalox ceramic saddles and wood chips could be renewed during the fermentation, guaranteeing their viability and making them more efficient in attenuating the oscillations. The packing Raschig rings without yeast cell immobilization effect did not affect the oscillatory behavior of the tubular bioreactors, further supporting the role of the yeast cell immobilization in the oscillation attenuation. 相似文献

The objective of this work was to select and test systematically possible cryoprotective agents (CPAs) and to obtain a suitable formula for vitrification of corneal endothelial cells (CECs). Fresh bovine CECs were isolated and tested with an optimized vitrification protocol with multi-step CPA loading and removal. Three types of CPAs components, i.e. the penetrating CPAs, sugars and macromolecular compounds, were experimentally evaluated using the viability assayed by trypan blue. Dimethyl sulfoxide, ethylene glycol (EG), 1,2-propanediol, 2,3-butanediol, acetamide and ethylene glycol monomethyl ether were chosen as the penetrating CPA components. Sugars including xylose, fructose, mannose, glucose, maltose, sucrose and trehalose were tested. Ficoll (MW 7 kDa), dextran (MW 7 kDa), chondroitin sulfate (CS, MW 18-30 kDa), bovine serum albumin (MW 68 kDa) and polyethylene glycol (MW 6 kDa, 10 kDa and 20 kDa) were chosen as the macromolecular compounds. CECs were also preserved by slow freezing as a control. The results showed that EG was the most suitable penetrating CPA component and glucose the most suitable sugar, and CS the most suitable macromolecule. The optimized concentrations for each component in the vitrification solution were 52% (w/w) EG, 8% (w/w) glucose and 3% (w/w) CS. The CEC survival rate of 89.4 ± 2.1% (mean ± SD) was obtained using this formula and established vitrification protocol which was comparable to that by slow freezing. 相似文献

Extracellular signal-regulated kinase 5 (ERK5), a recently discovered mitogen-activated protein kinase (MAPK), plays a key role in the development and pathogenesis of cardiovascular disease. In order to clarify the pathophysiological significance of ERK5 in vascular remodeling, we investigated ERK5 phosphorylation in hypertrophy of human aortic smooth muscle cells (HASMCs) induced by angiotensin II (Ang II). The AT1 receptor was involved in Ang II-induced ERK5 activity. Hypertrophy was detected by the measurement of protein synthesis with [3H]-Leu incorporation in cultured HASMCs. Ang II rapidly induced phosphorylation of ERK5 at Thr218/Tyr220 residues in a time- and dose-dependent manner. Activation of myocyte enhancer factor-2C (MEF2C) by ERK5 was inhibited by PD98059. Transfecting HASMCs with small interfering RNA (siRNA) to silence ERK5 inhibited Ang II-induced cell hypertrophy. Thus, ERK5 phosphorylation contributes to MEF2C activation and subsequent HASMC hypertrophy induced by Ang II, for a novel molecular mechanism in cardiovascular diseases induced by Ang II. 相似文献

An innovative consecutive batch fermentation process was developed for very high gravity (VHG) ethanol fermentation with the
self-flocculating yeast under high biomass concentration conditions. On the one hand, the high biomass concentration significantly
shortened the time required to complete the VHG fermentation and the duration of yeast cells suffering from strong ethanol
inhibition, preventing them from losing viability and making them suitable for being repeatedly used in the process. On the
other hand, the separation of yeast cells from the fermentation broth by sedimentation instead of centrifugation, making the
process economically more competitive. The VHG medium composed of 255 g L−1 glucose and 6.75 g L−1 each of yeast extract and peptone was fed into the fermentation system for nine consecutive batch fermentations, which were
completed within 8–14 h with an average ethanol concentration of 15% (v/v) and ethanol yield of 0.464, 90.8% of its theoretical value of 0.511. The average ethanol productivity that was calculated
with the inclusion of the downstream time for the yeast flocs to settle from the fermentation broth and the supernatant to
be removed from the fermentation system was 8.2 g L−1 h−1, much higher than those previously reported for VHG ethanol fermentation and regular ethanol fermentation with ethanol concentration

Starch films were developed from oxidized potato starch (OPS) with glycerol as a plasticizer at different contents. The OPS films were transparent and flexible. The mechanical properties of these films were measured, and the results indicated that the film with 19.4% glycerol exhibited the desirable mechanical properties. X-ray diffraction study showed that the increase of glycerol content led to a decrease in the crystallinity for OPS films, and storage conditions such as storage time, storage temperature and relative humidity also had certain effects on the retrogradation of starch owing to re-crystallization. Anti-leakage, anti-crosslinking, and stability in acid or alkali solutions of the OPS films were also studied, and the results indicated that the OPS films had excellent anti-leakage ability for vegetable oil, good anti-crosslinking ability in saturated formaldehyde vapor, and good stability in acid aqueous medium, but poor stability in alkali aqueous medium. 相似文献
