The nucleotide sequences, genetic organization, and distribution of plasmids pEU30 (30,314 bp) and pEL60 (60,145 bp) from the plant pathogen Erwinia amylovora are described. The newly characterized pEU30 and pEL60 plasmids inhabited strains isolated in the western United States and Lebanon, respectively. The gene content of pEU30 resembled plasmids found in plant-associated bacteria, while that of pEL60 was most similar to IncL/M plasmids inhabiting enteric bacteria.  相似文献

Photoacclimation involves the modification of components of the light and dark reactions to optimize photosynthesis following changes in available light. All of the energy required for photosynthesis comes from linear electron transport through PSII and PSI and is dependent upon the amount of light harvested by PSII relative to PSI (a*PSII and a*PSI). The amount of light harvested is determined by the effective absorption cross‐sections (σPSII, σPSI) and cellular contents of the PSII and PSI reaction center complexes (RCII, RCI). Here, we examine the effective absorption cross‐sections and reaction center contents for calcifying (B11) and noncalcifying (B92) strains of the globally important coccolithophorid Emiliania huxleyi (Lohmann) W. H. Hay et H. Mohler when grown under various photon flux densities (PFDs). The two strains displayed different “strategies” of acclimation. As growth PFD increased, B11 preferentially changed σ and the cellular content of chl a per cell over PSU “size” (the total cellular chl a content associated with the reaction center complexes); strain B92 preferentially changed PSU size over the cellular content of reaction complexes. Neither strategy was specifically consistent with the majority of previous studies from other microalgal species. For both strains, cellular light absorption for PSII and PSI was maintained close to unity across the range of growth PFDs since changes of σPSII and σPSI were reciprocated by those of RCIIs and RCIs per cell. Our results demonstrate a significant adaptive flexibility of E. huxleyi to photoacclimate. Finally, we calculated the amount of chl a associated with either photosystem to consider our interpretations of photoacclimation based on conventional determinations of PSU size.  相似文献

Summary Giemsa staining procedures have been used on human lymphocyte preparations to locate constitutive heterochromatin (C-bands) and to produce distinct banding patterns for each individual chromosome (G-bands). A metacentric C group chromosome has been shown by both techniques to be chromosome number 9.

Beide Techniken erlaubten, ein metazentrisches C-Chromosom als Nr. 9 zu identifizieren.

A nonpathogenic mutant of Colletotrichum magna (path-1) was previously shown to protect watermelon (Citrullus lanatus) and cucumber (Cucumis sativus) seedlings from anthracnose disease elicited by wild-type C. magna. Disease protection was observed in stems of path-1-colonized cucurbits but not in cotyledons, indicating that path-1 conferred tissue-specific and/or localized protection. Plant biochemical indicators of a localized and systemic (peroxidase, phenylalanine ammonia-lyase, lignin, and salicylic acid) “plant-defense” response were investigated in anthracnose-resistant and -susceptible cultivars of cucurbit seedlings exposed to four treatments: (1) water (control), (2) path-1 conidia, (3) wild-type conidia, and (4) challenge conditions (inoculation into path-1 conidia for 48 h and then exposure to wild-type conidia). Collectively, these analyses indicated that disease protection in path-1-colonized plants was correlated with the ability of these plants to mount a defense response more rapidly and to equal or greater levels than plants exposed to wild-type C. magna alone. Watermelon plants colonized with path-1 were also protected against disease caused by Colletotrichum orbiculare and Fusarium oxysporum. A model based on the kinetics of plant-defense activation is presented to explain the mechanism of path-1-conferred disease protection.  相似文献

Data on activity states were collected from 29 group-housed capuchin monkey (Cebus apella) infants for 3 h each week from birth to 11 weeks of age. The amounts of time spent in sleeping/drowsy, alert–quiet, and alert–active states were measured in these subjects. Videotaped observations of these infants were recorded 3 times/week in the home cage over the first year of life and were scored for a number of social and exploratory behaviors. The extent to which early infant activity state scores predicted later behavior in the home cage was examined. Infant state measures correlated significantly with home cage behavior during months 2–6 in that infants that had been more active in early infancy spent more time alone, with other animals, and in exploration and play and less time with mothers than did quieter infants. Early state measures were less successful in predicting home cage scores beyond 8 months of age, whereas differences in behavior attributable to housing variables became more salient in the latter part of the first year. There was also a negative correlation between mother and infant activity in months 2 and 3, in that more sedentary mothers tended to have more active infants. Am. J. A characteristic of neurological disorders is the loss of critical populations of cells that the body is unable to replace,thus there has been much interest in identifying methods of generating clinically relevant numbers of cells to replace those that have been damaged or lost.The process of neural direct conversion,in which cells of one lineage are converted into cells of a neural lineage without first inducing pluripotency,shows great potential,with evidence of the generation of a range of functional neural cell types both in vitro and in vivo,through viral and non-viral delivery of exogenous factors,as well as chemical induction methods.Induced neural cells have been proposed as an attractive alternative to neural cells derived from embryonic or induced pluripotent stem cells,with prospective roles in the investigation of neurological disorders,including neurodegenerative disease modelling,drug screening,and cellular replacement for regenerative medicine applications,however further investigations into improving the efficacy and safety of these methods need to be performed before neural direct conversion becomes a clinically viable option.In this review,we describe the generation of diverse neural cell types via direct conversion of somatic cells,with comparison against stem cell-based approaches,as well as discussion of their potential research and clinical applications.  相似文献

Genes of the Schlafen family, first discovered in mouse, are expressed in hematopoietic cells and are involved in immune processes. Previous results showed that they are candidate genes for two major phenomena: meiotic drive and embryonic lethality (DDK syndrome). However, these genes remain poorly understood, mostly due to the limitations imposed by their similarity, close location and the potential functional redundancy of the gene family members.  相似文献

Malus sieversii (Lebed.) M. Roem. is a wild progenitor species of the domesticated apple. It is found across a mountainous region of central Asia and has been the focus of several collection expeditions by the USDA-ARS-National Plant Germplasm System. This study used microsatellite variation at seven loci to estimate diversity and differentiation within M. sieversii using several complimentary approaches. Multilocus genotypes were amplified from 949 individuals representing seedling trees from 88 half-sib families from eight M. sieversii populations collected in Kazakhstan. Apportioning of genetic variation was estimated at both the family and site level. Analyses using a hierarchical model to estimate F st showed that differentiation among individual families is more than three times greater than differentiation among sites. In addition, average gene diversity and allelic richness varied significantly among sites. A rendering of a genetic network among all sites showed that differentiation is largely congruent with geographical location. In addition, nonhierarchical Bayesian assignment methods were used to infer genetic clusters across the collection area. We detected four genetic clusters in the data set. The quality of these assignments was evaluated over multiple Markov Chain Monte Carlo runs using both posterior likelihood and stability of the assignments. The spatial pattern of genetic assignments among the eight collection sites shows two broadly distributed and two narrowly distributed clusters. These data indicate that the southwestern collection sites are more admixed and more diverse than the northern sites.  相似文献

Aim Urbanization and deforestation are important drivers of biodiversity change. However, long‐term changes in faunal communities within urbanizing regions are poorly understood. We investigated how well observed community changes in both space and time agree with expectations based on current paradigms in urban ecology. Location Greater Brisbane region, Australia. Methods We compared bird assemblages in two time‐periods 15 years apart, at multiple sites in remnant forest and residential suburbs across an urbanizing landscape. Differences in assemblage composition, species abundances and functional groupings were assessed within and between habitats. Results Compared with forest, suburbs in both time‐periods had over twice the total bird abundance, a different species composition, greater between‐site community similarity, a greater proportion of non‐native species and greater dominance by large‐bodied species. These differences corresponded with changes in sites whose habitat was converted from forest to suburb. Between time‐periods, abundances of 58% of suburban species changed significantly compared with those of 11% in forest. Increaser species outnumbered decreasers in suburbs, with the reverse in forest. Abundance of small‐bodied birds decreased 70% in suburbs and 20% in forest. Broad‐spectrum competitors and nest predators were common among suburban increasers. Among invasive species, the number of increasers was counterbalanced by decreasers. Both site‐scale species richness and between‐site community similarity increased to a small extent in both habitats. Main conclusions Species composition and ecological function of suburban bird communities were very dynamic. Suburban assemblages were neither a subset of forest species nor an increasingly non‐native compilation. Communities in large forest patches were comparatively stable. The notion of habitat‐specific species turnover better characterizes the nature of most changes than either species decline or homogenization, even though both of these were evident. There is considerable scope for careful urban planning, focused on both among‐ and within‐habitat variety, to sustain bird diversity in urbanizing landscapes.  相似文献
