Four-day-old etiolated cucumber seedlings (Cucumis sativus L.) were transferred to cool-white-fluorescent light (15 mumol m-2 s-1) for 1 h and 24 hours and etiochloroplasts and chloroplasts were isolated from developing cotyledons. Plastids were fractionated to stroma, envelope and thylakoid or inner membranes and the pigment contents of all these different fractions were analysed. In intact cucumber chloroplast protochlorophylide was present in significant amounts whereas protoporphyrin IX and Mg-protoporphyrin plus its monoester were present only in very small quantities. Out of the total chloroplastic protochlorophylide pool 1.0% was partitioned to envelope membranes and 99.0% was partitioned to thylakoids. Stroma had only trace amounts of protochlorophylide. In contrast to chloroplasts, etiochloroplasts had, besides protochlorophylide, significant amounts of other chlorophyll biosynthetic intermediates. In etiochloroplasts, protoporphyrin IX primarily partitioned to inner membranes (59.1%) followed by stroma (37.7%) and envelope (3.21%). The content of Mg-protoporphyrin IX plus its monoester in different subplastidic fractions was 74.4% for inner membranes, 22.58% for stroma and 3.02% for envelope. Protochlorophyllide primarily partitioned to inner membranes (95.79%), followed by envelope (4.15%) and, to a negligible extent (0.06%), into stroma. The sub-plastidic distribution of chlorophyll biosynthetic intermediates in etiochloroplasts was, therefore, different than that of chloroplasts. The significance of differential distribution of chlorophyll biosynthetic intermediates among thylakoids, envelope and stroma in developing and mature plastids is discussed in relation to chloroplast biogenesis. 相似文献

While polyamines (PAs) have been suggested to protect cells against Reactive Oxygen Species (ROS), their catabolism is known to generate ROS. We compared the activities of several enzymes and cellular metabolites involved in the ROS scavenging pathways in two isogenic cell lines of poplar (Populus nigra × maximowiczii) differing in their PA contents. Whereas the control cell line was transformed with β-glucuronidase (GUS), the other, called HP (High Putrescine), was transformed with a mouse ornithine decarboxylase (mODC) gene. The expression of mODC resulted in several-fold increased production of putrescine as well its enhanced catabolism. The two cell lines followed a similar trend of growth over the seven-day culture cycle, but the HP cells had elevated levels of soluble proteins. Accumulation of H2O2 was higher in the HP cells than the control cells, and so were the activities of glutathione reductase and monodehydroascorbate reductase; the activity of ascorbate peroxidase was lower in the former. The contents of reduced glutathione and glutamate were significantly lower in the HP cells but proline was higher on some days of analysis. There was a small difference in mitochondrial activity between the two cell lines, and the HP cells showed increased membrane damage. In the HP cells, increased accumulation of Ca was concomitant with lower accumulation of K. We conclude that, while increased putrescine accumulation may have a protective role against ROS in plants, enhanced turnover of putrescine actually can make them vulnerable to increased oxidative damage. 相似文献

Atrial natriuretic peptide (ANP), the C-terminal peptide comprising residues 99-126 of the pro-ANP hormone, has been studied for 3 decades for its cardiovascular effects. Recent reports suggest that it plays a significant role in modulation of the immune system. Immune cells, including macrophages, dendritic cells, and T lymphocytes, express receptors for ANP. ANP plays a significant role in shaping the early immune response to environmental antigens and may play a critical role in the interaction between cells of the innate and adaptive immune systems; it also appears to be involved in polarizing the immune response to allergens. Thus, ability to alter the magnitude of natriuretic peptide receptor A (NPRA) signaling could be exploited to develop therapeutics for several allergic diseases, including asthma. Recent modern techniques of propagation have been developed which could help growers to meet the demand of the horticultural industry in the next century. An overview on the in vitro propagation via thin cell layer, meristem culture, regeneration via organogenesis and somatic embryogenesis is presented. Available methods for the transfer of genes could significantly simplify the breeding procedures and overcome some of the agronomic and environmental problems, which other wise would not be achievable through conventional propagation methods. The development and remarkable achievements with biotechnology in ornamental pot plants made during the three decades have been reviewed. Response surface methodology was used to develop a fermentation medium for the enhanced biosynthesis of a novel sulfide oxidase by Arthrobacter species strain FR‐3. The interactive effect of the medium components – such as glucose as the carbon source, and tryptone and yeast extract as the nitrogen source – was evaluated by a 23‐factorial central composite statistical design. Glucose and yeast extract were found to be the more influential medium constituents compared to tryptone since they had lower coefficients of linear effect, P‐values (< 0.02). The optimal fermentation medium components for the enhanced production of sulfide oxidase were recorded as glucose (8.98 g/L), tryptone (10.62 g/L) and yeast extract (7.3 g/L). Optimization of the medium constituents increased the experimental enzyme yield by 54 % compared to the unoptimized medium.
