Abstract

Bacterial cellulose (BC) is one of the prominent biopolymers that has been acquiring attention currently due to its distinctive properties and applications in various fields. The current work presents the isolation of Komagataeibacter saccharivorans strain BC1 isolated from rotten green grapes, followed by biochemical and genotypic characterization, which confirmed that the strain is capable of synthesizing cellulose. Further, production media was designed and certain variables such as carbon, nitrogen sources, pH, and temperature were optimized in order to obtain the maximum concentration of cellulose production. We found mannitol to be the ideal carbon source and yeast extract as the ideal nitrogen source with a highest BC dry yield of 1.81?±?0.25?g/100?mL at pH 5.76 for a week at 30?°C.The charcterization of pellicles by FTIR spectrum depicted similar functional groups present in synthesized BC as that of the commercial cellulose. X-ray diffraction revealed that BC showed 82% crystallinity. Surface morphology of the dried pellicle was studied by SEM image which showed that the BC surface was tightly packed with thin fibers with less porosity. Hence the study demonstrates that the isolates of K.saccharivorans could be used to produce a biopolymer in a short period of time using a modified production medium.  相似文献

Staphylococcus aureus is a commensal gram positive bacteria which causes severe and non severe infections in humans and livestock. In India, ST772 is a dominant and ST672 is an emerging clone of Staphylococcus aureus. Both cause serious human diseases, and carry type V SCCmec elements. The objective of this study was to characterize SCCmec type V elements of ST772 and ST672 because the usual PCR methods did not amplify all primers specific to the type. Whole genome sequencing analysis of seven ST772 and one ST672 S. aureus isolates revealed that the SCCmec elements of six of the ST772 isolates were the smallest of the extant type V elements and in addition have several other novel features. Only one ST772 isolate and the ST672 isolate carried bigger SCCmec cassettes which were composites carrying multiple ccrC genes. These cassettes had some similarities to type V SCCmec element from M013 isolate (ST59) from Taiwan in certain aspects. SCCmec elements of all Indian isolates had an inversion of the mec complex, similar to the bovine SCCmec type X. This study reveals that six out of seven ST772 S. aureus isolates have a novel type V (5C2) SCCmec element while one each of ST772 and ST672 isolates have a composite SCCmec type V element (5C2&5) formed by the integration of type V SCCmec into a MSSA carrying a SCC element, in addition to the mec gene complex inversions and extensive recombinations.  相似文献

The major aspects of using plant-derived medications are significantly safer and secure than synthetic ones. The n-hexane seed extract of ayurvedic medicinal plants Myristica fragrans was also utilized as food ingredients have analyzed for phytochemical existence by Gas Chromatography-mass spectrometry (GC–MS). Twenty-three phytoconstituents were identified with elemicin (24.44%) as the major constituent. Lipid peroxidase, catalase and DPPH assays were performed using the isolated elemicin and the results revealed significant antioxidant activity. The antibacterial study revealed that elemicin showed MIC of 31.25 μg/mL against Escherichia coli, Pseudomonas aeruginosa, and Salmonella typhi, and 62.5 μg/mL against Klebsiella pneumonia and Staphylococcus aureus. Elemicin exhibited better antifungal activity against Candida tropicalis and Aspergillus flavus than Aspergillus niger, Penicillium chrysogenum and Trichophyton rubrum. The study implies that the elemicin isolated from Myristica fragrans possess promising bioactive properties and can be crucially utilized in the development of therapeutic agents and food preservatives.  相似文献

Signaling of information in the vertebrate central nervous system is often carried by populations of neurons rather than individual neurons. Also propagation of suprathreshold spiking activity involves populations of neurons. Empirical studies addressing cortical function directly thus require recordings from populations of neurons with high resolution. Here we describe an optical method and a deconvolution algorithm to record neural activity from up to 100 neurons with single-cell and single-spike resolution. This method relies on detection of the transient increases in intracellular somatic calcium concentration associated with suprathreshold electrical spikes (action potentials) in cortical neurons. High temporal resolution of the optical recordings is achieved by a fast random-access scanning technique using acousto-optical deflectors (AODs)1. Two-photon excitation of the calcium-sensitive dye results in high spatial resolution in opaque brain tissue2. Reconstruction of spikes from the fluorescence calcium recordings is achieved by a maximum-likelihood method. Simultaneous electrophysiological and optical recordings indicate that our method reliably detects spikes (>97% spike detection efficiency), has a low rate of false positive spike detection (< 0.003 spikes/sec), and a high temporal precision (about 3 msec) 3. This optical method of spike detection can be used to record neural activity in vitro and in anesthetized animals in vivo3,4.  相似文献

Periodontitis is attributed to the dental biofilm formation. Red complex organisms are a group of organisms linked with periodontal diseases. Therefore, it is of interest to identify potential targets from the red complex organisms to bind with the herbal compound resveratrol (E - 5 - (4 - hydroxy styryl) benzene 1,3 diol). We report a list of potential proteins having optimal drug like binding features with the herbal agent Resveratrol for further consideration. We used the STITCH v.5 pipeline VICMPred and VirulentPred tools to identify such targets as potential virulent factors in the red complex organisms. We considered the strains of Porphyromonas gingivalis ATCC 33277, Treponema denticola ATCC 35405 and Tannerella forsythia ATCC 43037 in the red complex pathogens for this analysis. Protein targets in the red complex organisms with optimal binding features with the herbal compound resveratrol were thus identified and reported for further consideration.  相似文献

Epidermal growth factor receptor (EGFR) is a potential target with disease modifying benefits against Alzheimer's disease (AD). Repurposing of FDA approved drugs against EGFR have shown beneficial effect against AD but are confined to quinazoline, quinoline and aminopyrimidines. Futuristically, the possibility of acquiring drug resistance mutation as seen in the case of cancer could also hamper AD treatment. To identify novel chemical scaffolds, we rooted on phytochemicals identified from plants such as Acorus calamus, Bacopa monnieri, Convolvulus pluricaulis, Tinospora cordifloia, and Withania somnifera that have well-established records in the treatment of brain disorders. The rationale was to mimic the biosynthetic metabolite extension process observed in the plants for synthesizing new phytochemical derivates. Thus, novel compounds were designed computationally by fragment-based method followed by extensive in silico analysis to pick potential phytochemical derivates. PCD1, 8 and 10 were predicted to have better blood brain barrier permeability. ADMET and SoM analysis suggested that these PCDs exhibited druglike properties. Further simulation studies showed that PCD1 and PCD8 stably interact with EGFR and have the potential to be used even in cases of drug-resistance mutations. With further experimental evidence, these PCDs could be leveraged as potential inhibitors of EGFR.  相似文献
