Physiological strategies driving the reduction of poorly crystalline Fe(III) oxide by the thermophilic Gram-positive dissimilatory Fe(III)-reducing bacterium C. ferrireducens were evaluated. Direct cell-to-mineral contact appears to be the major physiological strategy for ferrihydrite reduction. This strategy is promoted by cell surface-associated c-type cytochromes, and the extracellular electron transfer to ferrihydrite is linked to energy generation via a membrane-bound electron transport chain. The involvement of pili-like appendages in ferrihydrite reduction has been detected for the first time in a thermophilic microorganism. A supplementary strategy for the utilization of a siderophore (DFO) in dissimilatory ferrihydrite reduction has also been characterized.  相似文献

This study explores the effects of early-life and middle-life conditions on exceptional longevity using two matched case-control studies. The first study compares 198 validated centenarians born in the United States between 1890 and 1893 to their shorter-lived siblings. Family histories of centenarians were reconstructed and exceptional longevity validated using early U.S. censuses, the Social Security Administration Death Master File, state death indexes, online genealogies, and other supplementary data resources. Siblings born to young mothers (aged less than 25 years) had significantly higher chances of living to 100 compared to siblings born to older mothers (odds ratio = 2.03, 95% CI = 1.33-3.11, p = .001). Paternal age and birth order were not associated with exceptional longevity. The second study explores whether people living to 100 years and beyond differ in physical characteristics at a young age from their shorter-lived peers. A random representative sample of 240 men who were born in 1887 and survived to age 100 was selected from the U.S. Social Security Administration database and linked to U.S. World War I civil draft registration cards collected in 1917 when these men were 30 years old. These validated centenarians were then compared to randomly selected controls who were matched by calendar year of birth, race, and place of draft registration in 1917. Results showed a negative association between "stout" body build (being in the heaviest 15 percent of the population) and survival to age 100. Having the occupation of "farmer" and a large number of children (4 or more) at age 30 increased the chances of exceptional longevity. The results of both studies demonstrate that matched case-control design is a useful approach in exploring effects of early-life conditions and middle-life characteristics on exceptional longevity.  相似文献

A number of recent experimental approaches demonstrated that a three-dimensional organization of the eukaryotic genome play an important role in the regulation of its activity. One of the most important results was a discovery of the genome separation into relatively independent topologically-associated domains (TADs). They restricted the action area of regulatory elements, i.e., they simultaneously were regulatory domains of the genome. In this connection, an understanding of the molecular mechanism of the TAD formation has become a very topical problem. Here, we review and discuss our recent data which demonstrated that the TAD formation was directed by simple physical laws and was based on establishing multiple internucleosomal contacts.  相似文献

The results of the evaluation of the immunogenic properties of B-antigen, earlier identified in the culture fluid of Yersinia pseudotuberculosis submerged culture, with respect to experimental plague and pseudotuberculosis are presented. B-antigen has been shown to produce protective effect in guinea pigs and, probably, hamadryas baboons, but not in white mice infected with the causative agent of plague. Immunizaton with B-antigen protects guinea pigs from primary pneumonic plague caused by both capsule-forming and noncapsular Y. pestis virulent strains. Passive immunization with antibodies to B-antigen induces limitedly pronounced protective effect in guinea pigs and is not effective for white mice with respect to experimental plague. No active or passive protection of white mice or guinea pigs, infected with Y. pseudotuberculosis cultures, has been achieved by the injection of B-antigen or antibodies to it.  相似文献

We used the 4C-Seq technique to characterize the genome-wide patterns of spatial contacts of several CpG islands located on chromosome 14 in cultured chicken lymphoid and erythroid cells. We observed a clear tendency for the spatial clustering of CpG islands present on the same and different chromosomes, regardless of the presence or absence of promoters within these CpG islands. Accordingly, we observed preferential spatial contacts between Sp1 binding motifs and other GC-rich genomic elements, including the DNA sequence motifs capable of forming G-quadruplexes. However, an anchor placed in a gene/CpG island-poor area formed spatial contacts with other gene/CpG island-poor areas on chromosome 14 and other chromosomes. These results corroborate the two-compartment model of the spatial organization of interphase chromosomes and suggest that the clustering of CpG islands constitutes an important determinant of the 3D organization of the eukaryotic genome in the cell nucleus. Using the ChIP-Seq technique, we mapped the genome-wide CTCF deposition sites in the chicken lymphoid and erythroid cells that were used for the 4C analysis. We observed a good correlation between the density of CTCF deposition sites and the level of 4C signals for the anchors located in CpG islands but not for an anchor located in a gene desert. It is thus possible that CTCF contributes to the clustering of CpG islands observed in our experiments.  相似文献

During intrauterine life microcirculatory bed of the muscular coat is forming and developing in accordance with its differentiation, growth and functional changes at various steps of its ontogenesis. This step-like differentiation in the parts composing the microcirculatory bed and the whole microcirculatory network of longitudinal and circulatory layers of the muscular coat is genetically connected with differentiation taking place in the wall of the sigmoid colon during the developmental period studied and, in the end, reflects morphological maturation of the sigmoid colon at different stages of its individual ontogenesis.  相似文献
