E. coli cells produce acetate as an extracellular coproduct of aerobic cultures. Acetate is undesirable because it retards growth and inhibits protein formation. Most process designs or genetic modifications to minimize acetate formation aim at balancing growth rate and oxygen consumption. In this research, three genetic approaches to reduce acetate formation were investigated: (1) direct reduction of the carbon flow to acetate (ackA-pta, poxB knock-out); (2) anticipation on the underlying metabolic and regulatory mechanisms that lead to acetate (constitutive ppc expression mutant); and (3) both (1) and (2). Initially, these mutants were compared to the wild-type E. coli via batch cultures under aerobic conditions. Subsequently, these mutants were further characterized using metabolic flux analysis on continuous cultures. It is concluded that a combination of directly reducing the carbon flow to acetate and anticipating on the underlying metabolic and regulatory mechanism that lead to acetate, is the most promising approach to overcome acetate formation and improve recombinant protein production. These genetic modifications have no significant influence on the metabolism when growing the micro-organisms under steady state at relatively low dilution rates (less than 0.4 h(-1)).  相似文献

Using genetic engineering technologies, the chitin-binding domain (ChBD) of the human macrophage chitotriosidase has been inserted into the host protein BlaP, a class A beta-lactamase produced by Bacillus licheniformis. The product of this construction behaved as a soluble chimeric protein that conserves both the capacity to bind chitin and to hydrolyze beta-lactam moiety. Here we describe the biochemical and biophysical properties of this protein (BlaPChBD). This work contributes to a better understanding of the reciprocal structural and functional effects of the insertion on the host protein scaffold and the heterologous structured protein fragments. The use of BlaP as a protein carrier represents an efficient approach to the functional study of heterologous protein fragments.  相似文献

Using radioimmunoassay for total immunoreactive blood neurophysins (I.R.Neur.) we demonstrate a significant increase 30' after the onset of suckling in 26 normal carefully studied lactating women. On the first day postpartum a constant increase is only observed in 12 patients whilst a significant increase is clearly shown for all patients from the 2nd to the 6th day. These variations are similar to those observed for prolactin blood levels; there is however no relation between basal, suckling stimulated, or ergocryptin inhibited prolactin blood level on the one hand and I.R.Neur. blood level on the other hand.  相似文献

Two male Florida manatees (Trichechus manatus latirostris) demonstrated sensitive tactile discrimination in a two‐alternative forced choice task, using a modified staircase method. Stimuli were acrylic plates with vertical gratings of ridges and grooves. The standard stimulus, present on every trial, had 2 mm gratings and the comparison stimuli had wider gratings. The blindfolded subjects were trained to demonstrate discrimination by pressing the target with wider gratings. Discrimination thresholds (75% correct) for the subjects were 2.05 mm and 2.15 mm, corresponding to Weber fractions of 0.025 and 0.075, respectively. These results indicate thresholds on similar stimuli comparable to humans (index finger tasks) and better than harbor seals, Phoca vitulina, and the closely related Antillean manatee, Trichechus manatus manatus. Memory for the tactile task was quite stable for both subjects, over 2 yr in the case of one of the subjects. Video analysis of responses indicated that bristle‐like hairs, perioral bristles, and skin on the oral disk were involved in the discrimination response.  相似文献
