A major human acute phase protein, the serum amyloid A protein, has been tested in vitro for its effect on lymphocyte proliferation, the formation of E-stable rosettes, as well as the growth of HeLa and MRC5 cell cultures. Serum amyloid A protein has been found to be markedly inhibitory at 30, 100, 200, and 300 micrograms/mL, and is a very potent inhibitor of in vitro biological functions.  相似文献

The development of nonviral gene delivery vehicles for therapeutic applications requires methods capable of quantifying the association between the genes and their carrier counterparts. Here we investigate the potential of fluorescence cross-correlation spectroscopy (FCCS) to characterize and optimize the assembly of nonviral cationic liposome (CL)-DNA complexes based on a CL formulation consisting of the cationic lipid DOTAP and zwitterionic lipid DOPC. We use a DNA plasmid for lipoplex loading encoding the Oct4 gene, critically involved in reprogramming somatic cells into induced pluripotent stem cells. We demonstrate that FCCS is able to quantitatively determine the extent of the association between DNA and the liposomes and assess its loading capacity. We also establish that the cationic lipid fraction, being proportional to the liposome membrane charge density, as well as charge ratio between the CLs and anionic DNA play an important role in the degree of interaction between the liposomes and DNA.  相似文献

The detailed spatial organization of cytoskeletal proteins in an immortalized sympathoadrenal precursor cell line, termed MAH, was studied when the cells were grown on cellular substrates and when treated with combinations of recombinant nerve growth factor, ciliary neurotrophic factor and basic fibroblast growth factor. In response to growth factors, MAH cells expressed appropriate distributions of phosphorylated and non-phosphorylated neurofilaments, and dendrite and axon specific microtubule associated proteins. Sequential stages of maturation and axon formation were identified as the MAH cells established neuronal polarity and developed into sympathetic-like neurons. Combinations of the growth factors initiated growth associated protein-43 expression in processes and promoted the MAH cells to acquire sympathetic-like neuron characteristics with long, thin processes that branched and often terminated in elaborate growth cones. When treated with the three trophic factors, 15% of the MAH cells differentiated into sympathetic-like neurons, in contrast to less than 10% when cultured with ciliary neurotrophic factor plus nerve growth factor. An enhanced cholinergic phenotype was evident in the MAH cells when grown with ciliary neurotrophic factor. MAH cells also expressed neuron-specific markers when co-cultured on enriched substrates of smooth muscle, fibroblasts or Schwann cells. The results indicate that this sympathoadrenal cell lineage, carrying the v-myc oncogene, can express appropriate cytoskeletal markers in the process of neuronal differentiation when induced by neurotrophic factors or by specific cellular conditions in vitro.  相似文献

The rooting capacities of tips of seedling, juvenile and mature shoots of Sequoiadendron giganteum were compared on different rooting media (inductive and expressive media) after passage on an elongating medium. None of the cuttings rooted when continuously kept on medium containing the auxin NAA and vitamin D2. Peroxidase activity of all those cuttings on NAA+D2 first increased during the 7–9 first days and decreased in the days after. Rooting was obtained by transfer of the cuttings after periods longer than 7–9 days from the NAA+D2 inductive medium to a basal medium supplemented or not with rutin (expressive medium). The rooting capacity was emphasized by rutin treatment and was in correlation with the peroxidase peak reached on the NAA+D2 medium. A leaf protein concentrate (LPC) extracted from shelled plants of pea (Pisum sativum L.) yielded 108 kg DM ha?1. Dry matter (DM) was analysed for ether extract (13.59%), neutral detergent fibre (NDF, 1.79%), acid detergent fibre (ADF, 0.81%) and total ash (10.57%). The main nitrogen fraction was represented by protein-N (92% of total N). Mineral composition showed an adequate ratio of Ca to P (3). Lysine was present in nutritionally suitable amounts (7.42 g per 100 g recovered amino acids), but methionine was not (1.78). Apparent digestibility (AD; 75.5), true digestibility (TD; 87.6), biological value (BV; 68.8) and net protein utilisation (NPU; 59.7) were determined with Wistar albino rats. Methionine addition improved AD (81.3), BV (80.6) and NPU (70.5), although not TD (87.5) which was already very high.  相似文献

n-Alkane and long-chain alcohol (LCOH) profiles of needle cuticular waxes of nine provenances of three Pinus species (P. taeda. P. pinaster and P. pinea) were determined and their chemotaxonomic importance was studied. n-Alkanes concentrations were very low in all Pinus spp. A lack of ability of these compounds to differentiate Pinus spp. and their provenances was observed. LCOH concentrations were much higher, being nonacosan-10-ol (10-C29-OH) the most abundant one in all Pinus spp. Total LCOH concentrations varied (P<0.001) between Pinus spp. with P. taeda presenting the highest (P<0.05) value and P. pinea the lowest one. Differences in LCOH profiles were large (P<0.001) and allowed separation of Pinus spp. in the Principal Component Analysis (PCA). Although a noticeable separation of individuals was not observed. cluster analysis on LCOH concentrations allowed a clear distinction between species, indicating their potential to be used as chemotaxonomic markers to differentiate provenances of different Pinus spp.  相似文献
