ERG and EP of the visual cortex (VC) and superior colliculus (SC) were investigated under experimental dystrophy of the retina induced by intravenous injection of monoiodoacetic acid in different doses. The relative resistance of the VC to the derangement of retinal function was established. It was manifested by less marked suppression of its EP as compared with ERG. Taking into consideration the selective projection to the SC of the rod system that was damaged to a greater degree under pathology in question, it is suggested that functional suppression of the SC might facilitate the conduction of visual information through the main retino-geniculo-cortical canal. This might be one of the factors of VC resistance to the impairment of retinal function. This assumption was confirmed in experiments with SC electrocoagulation in which one could observe the facilitation of EP formation of the contralateral VC. 相似文献

Genes of human neurotrophins NGF, BDNF, NT-3 were cloned, and the corresponding proteins and their fragments were expressed in Escherichia coli BL-21 (DE3lambda) cells. Their intracellular localization was determined. The conditions for isolation and purification of the target recombinant proteins and for folding of BDNF and NT-3 precursors were selected. On Wistar rats characteristics were studied of investigating behaviour in the open field, of learning of conditioned food-reinforced reaction and also of BA and their metabolites content in various brain structures under local intracerebral injections of specific neurotoxins; 6-hydroxydopamine (6-OHDA) and 5,7-dihydroxytryptamine (5,7-DHT), abolishing correspondingly catecholaminergic and serotoninergic terminals. Bilateral injection of 6-OHDA in the neocortex led to a weakening of rats investigating activity in the open field and to an increase of the time of fulfillment of the forming of conditioned food-reinforced reaction. Administration of 5,7-DHT was accompanied by an increase of the investigating behaviour in the open field and a reduction of the duration of the forming of conditioned reaction. Administration of 6-OHDA to the neocortex caused a lowering of catecholamines level in the frontal area of the neocortex and the hippocampus. Analogous administration of 5,7-DHT elicited simultaneously with a deep level lowering of 5-HT and its metabolite in these structures, a change of catecholamines content which testifies to a lesser specificity of the neurotoxin 5,7-DHT in comparison with 6-OHDA. Structures lesion of serotoninergic and catecholaminergic systems of the frontal cortex and the hippocampus brought about by a local administration of 6-OHDA and 5,7-DHT in the neocortex was accompanied by differently directed changes in animals behaviour. 相似文献

Some literature and author's experimental data are presented on functional and morphological characteristics of nerve cells by their electrophysiological parameters. The proposition is substantiated that in multineuron discharges reflecting the activity of microgroups of neighbouring cells, the absolute size of impulses indicates the distance of cortical neurones from the tip of the electrode, while amplitudes correlation may characterize the size of the cell and some of its functional properties. Arguments for this proposition are: latencies of impulse responses to afferent and antidromic stimulations and amplitude changes of multineuronal activity during movement of the electrode through the cortex thickness in acute experiments. As the electrode (with the tip diameter of 50 mcm) approaches a microgroup of neurons, the amplitude of the recorded impulse discharges increases, while the relation of big spikes to small ones remains almost the same. 相似文献

In has been established that intrahippocampal bilateral injection of NA did not influence common frequency of lateral hypothalamic self stimulation. After the destruction of hippocampal NA - terminals of 6-OHDA increased the frequency of self stimulation and rearing. Binding of Gd3+ ions to linear dsDNA in water-salt solution and cholesteric liquid-crystalline dispersion (CLCD) drastically changes the optical and X-ray scattering characteristics of the samples. Depending on the complexing conditions, Gd3+ can bind to nucleobases and to phosphates, which differentially affects the properties of CLCD formed of such complexes. DNA packing in such dispersions changes with increasing Gd3+ occupancy, which is reflected in the decline of the Bragg reflection maximum in SAXS. The gene of Serratia proteamaculans proteinase, protealysin, was cloned, sequenced, and expressed in Escherichia coli. The gene encoded a precursor of 341 amino acids (AAs) with a significant homology to thermolysin-like proteinases (TLPs). The molecular weight of the purified mature active recombinant protein was 32 kDa, the N-terminal amino acid sequence was AKTSTGGEVI. The optimum pH for azocasein hydrolysis by protealysin was seven and it was completely inhibited by o-phenanthroline. The enzyme hydrolyzed 3-(2-furyl)acryloyl-glycyl-L-leucine amide, the standard substrate for TLPs, with k(cat)/K(m) ratio of (2.52 +/- 0.02) x 10(2) M(-1) s(-1). Protealysin maturation removes 50 AA from the N-terminus of the precursor. The removed region had no similarity with the preprosequence of thermolysin (232 AA) but was homologous to some other TLPs. These proteins shared a conserved 7-AA motif near the initial methionine. Such motif was also found in some nonproteolytic putative proteins; two of them were eukaryotic. 相似文献
