Plasma urea levels and hepatic urea production in the euryhaline bull shark, Carcharhinus leucas, acclimated to freshwater and seawater environments were measured. It was found that plasma urea concentration increased with salinity and that this increase was, in part, the result of a significant increase in hepatic production of urea. This study provides direct evidence that hepatic production of urea plays an important role in the osmoregulatory strategy of C. leucas. 相似文献

The construction cost of a plant tissue, i.e. the amount of
photoassimilates used in the synthesis of a unit weight, varies with its
biochemical composition. Crop modellers use standard values published for a
few groups of cultivated species. Yet, there are also intraspecific
variations in the construction cost in relation with the development of the
plant or organ. This research aimed at analysing the ontogenic changes in
the construction cost of leaves, stems, roots, and fruits of tomato plants
and the specific contribution of the mineral content to these changes. For
that purpose, samples were harvested from the vegetative phase to the
beginning of fruit production. The estimation of the construction cost was
based on the contents of carbon, nitrogen and ash. In leaves, the
construction cost decreased with the physiological age whereas, in stem
internodes, it varied with the sympod number. These ontogenic changes could
partly be explained by different accumulations of minerals. In contrast,
the construction cost and the mineral content of fruits and roots remained
fairly stable. On a whole plant basis, the construction cost of the bulk of
each category of organs varied much less. Most of the increase in the mean
construction cost of the whole plant during the experiment was due to
changes in the allocation ratio between the vegetative parts and the
fruits. Attention of crop modellers is drawn to the importance of a precise
estimation of the construction cost and to the existence of ontogenic
changes at the whole plant and organ levels.Key words:

Hydrazides of five N-acylamino acids have been used alone as substrates for papain catalysis to yield N1,N2-diacylhydrazines. With the exception of N-(benzyloxycarbonyl)(Z)-
-alanine hydrazide, they were very effective as both acylating agents of the enzyme and nucleophiles in attacking the enzyme-substrate intermediate. Although Z-
-alanine hydrazide was a minimal acylating agent, it was a satisfactory nucleophile. The most favorable reaction involved Z-
-alanine hydrazide in producing N1,N2-bis(Z-
-alanyl)hydrazine. When Z-
-alanine hydrazide was the substrate, this same chiral diacylhydrazine was formed along with meso N1-(Z-
-alanyl)-N2-(Z-
-alanyl)hydrazine. For the acylation step, the enzyme displayed powerful, essentially stereospecific, bias toward the
enantiomer. Once the thioester intermediate was formed, little preference was detected for attack by the enantiomers as nucleophiles. The most direct procedure for synthesis of substrates was conversion of Z-amino acids to their esters by means of dry HCl in an absolute alcohol. In past attempts, the experimental germination of the seeds of Euonymus americanus L. in vitro has had little success. However, treatment of seeds with ruminal fluid containing viable microflora has been successful in stimulating germination. In the presence of the cellulolytic ruminal bacterium, Clostridium cellobioparum ATCC 15832, seeds of E. americanus were stimulated to germinate. Subsequent studies were designed to determine whether the bacterium synthesized a cellulolytic enzyme responsible for initiating germination. The cell-free endocellulase from C. cellobioparum induced germination of the seeds. To support the hypothesis that the endocellulase from C. cellobioparum was responsible for triggering germination, a 1,4-beta-d-glucan glucanohydrolase (EC 3.2.1.4) from Penicillum funiculosum was used to treat the seeds. In addition, no germination was obtained from seeds treated with a commercial exocellulase enzyme. Also, Ruminococcus flavefaciens FD-1 was found to initiate germination of E. americanus seeds. Thus, cellulase activity is indicated in the degradation of the testa of the seed, allowing imbibition and germination. 相似文献

The culture of human embryonic stem (hES) cells in defined and xenogeneic-free conditions will contribute substantially to future biotechnological and medical applications. To achieve this goal, we developed the first fully defined synthetic polymer coating poly[2-(methacryloyloxy)ethyl dimethyl-(3-sulfopropyl)ammonium hydroxide] (PMEDSAH) that sustains long-term growth of hES cells in different culture media. Here we describe a detailed protocol for the reproducible fabrication of PMEDSAH coating on tissue culture polystyrene dishes, and for the feeder-free culture of hES cells on PMEDSAH coating in defined culture medium. Fruit bats of the genus Pteropus are important contributors to ecosystem maintenance on islands through their roles as pollinators and seed dispersers. However,
island faunas are the most prone to extinction and there is a real need to assess the possible genetic implications of population
reductions in terms of extinction risk. An effective method of ameliorating extinction risk in endangered species is the establishment
of captive populations ex situ. The effectiveness of captive breeding programmes may be assessed by comparing the genetic
variability of captive colonies to that of wild counterparts. Here, we use polymorphic microsatellite loci to assess genetic
variability in wild, critically endangered Rodrigues fruit bats (Pteropus rodricensis, Dobson 1878) and we compare this variability to that in a captive colony. We document remarkable conservation of genetic
variability in both the wild and captive populations, despite population declines and founder events. Our results demonstrate
that the wild population has withstood the negative effects of population reductions and that captive breeding programmes
can fulfil the goals of retaining genetic diversity and limiting inbreeding. 相似文献

Experiments were conducted to investigate the influence of endurance exercise training on protein synthesis in skeletal muscle, heart, and liver. Training decreased incorporation of [14C]-leucine into proteins of the stromal fraction of muscle but there was no change in amino acid incorporation into proteins of the sarcoplasmic and myofibrillar fractions. Incorporation of [14C]-leucine into the protein of heart, liver, and plasma was depressed in trained rats compared to untrained rats. The specific radioactivity of [14C]-leucine was similar in tissues of trained and untrained rats and thus the depressed amino acid incorporation represents a decrease in the rate of protein synthesis. These observations demonstrate that the adaptation of muscle protein metabolism to endurance training is quite different than the alterations during work-induced hypertrophy of muscle. The difference in adaptation probably relates to the functional differences between the types of exercise. However depression of protein synthesis in trained rats is a general effect in several tissues and not an effect localized in muscle tissue. 相似文献
