Inclusion bodies of recombinant human growth hormone (r-hGH) were isolated from Escherichia coli, enriched and solubilized in 100 mM Tris buffer containing 6 M n-propanol and 2 M urea. Around 4 mg/ml of r-hGH from inclusion bodies were solubilized in 6 M n-propanol-based buffer containing 2 M urea. Existence of native-like secondary structure of r-hGH in 6 M n-propanol solution was confirmed by CD and fluorescence spectra. Solubilized r-hGH was subsequently refolded by pulsatile dilution, purified to homogeneity and found to be functionally active. Tris buffer containing 6 M n-propanol and 2 M urea also effectively solubilized a number of proteins expressed as inclusion bodies in E. coli. Mild solubilization of inclusion body proteins, chaotropic effect of n-propanol at high concentration and kosmotropic effect at lower concentration helped in improved refolding of the solubilized protein. Around 40% of the r-hGH in the form of inclusion body aggregates was refolded into bioactive form while using n-propanol as solubilization agent. Solubilization with 6 M n-propanol solution thus can be a viable alternative for achieving high throughput recovery of bioactive protein from inclusion bodies of E. coli. 相似文献

Eight years after DNA barcoding was formally proposed on a large scale, CO1 sequences are rapidly accumulating from around the world. While studies to date have mostly targeted local or regional species assemblages, the recent launch of the global iBOL project (International Barcode of Life), highlights the need to understand the effects of geographical scale on Barcoding's goals. Sampling has been central in the debate on DNA Barcoding, but the effect of the geographical scale of sampling has not yet been thoroughly and explicitly tested with empirical data. Here, we present a CO1 data set of aquatic predaceous diving beetles of the tribe Agabini, sampled throughout Europe, and use it to investigate how the geographic scale of sampling affects 1) the estimated intraspecific variation of species, 2) the genetic distance to the most closely related heterospecific, 3) the ratio of intraspecific and interspecific variation, 4) the frequency of taxonomically recognized species found to be monophyletic, and 5) query identification performance based on 6 different species assignment methods. Intraspecific variation was significantly correlated with the geographical scale of sampling (R-square = 0.7), and more than half of the species with 10 or more sampled individuals (N = 29) showed higher intraspecific variation than 1% sequence divergence. In contrast, the distance to the closest heterospecific showed a significant decrease with increasing geographical scale of sampling. The average genetic distance dropped from > 7% for samples within 1 km, to < 3.5% for samples up to > 6000 km apart. Over a third of the species were not monophyletic, and the proportion increased through locally, nationally, regionally, and continentally restricted subsets of the data. The success of identifying queries decreased with increasing spatial scale of sampling; liberal methods declined from 100% to around 90%, whereas strict methods dropped to below 50% at continental scales. The proportion of query identifications considered uncertain (more than one species < 1% distance from query) escalated from zero at local, to 50% at continental scale. Finally, by resampling the most widely sampled species we show that even if samples are collected to maximize the geographical coverage, up to 70 individuals are required to sample 95% of intraspecific variation. The results show that the geographical scale of sampling has a critical impact on the global application of DNA barcoding. Scale-effects result from the relative importance of different processes determining the composition of regional species assemblages (dispersal and ecological assembly) and global clades (demography, speciation, and extinction). The incorporation of geographical information, where available, will be required to obtain identification rates at global scales equivalent to those in regional barcoding studies. Our result hence provides an impetus for both smarter barcoding tools and sprouting national barcoding initiatives-smaller geographical scales deliver higher accuracy. 相似文献

The enzymatic cleavage products of β-endorphin (β-endorphin1-27 and Gly-Gln) reduce voluntary alcohol consumption in alcohol-preferring (P) rats. Gly-Gln also inhibits the reward-benefiting effects of morphine and nicotine. It would be useful for the investigation of these effects to have an analytical method suitable for Gly-Gln detection and quantitation. Given the now widespread availability of liquid chromatography-tandem mass spectrometry (LC-MS/MS) instruments, the development of an LC-MS/MS-based approach seemed a viable option. An LC-MS/MS method for Gly-Gln quantitation was developed based on derivatization with Marfey's reagent. The Marfey's adduct of Gly-Gln (Mar-Gly-Gln) was chromatographically resolved and readily detected and quantitated by LC-MS/MS. Precursor/product positive ions of 456.2/366.2, 456.2/237.2, and 456.2/147.0 were used for detection and quantitation. This method shows good linearity from 1 to 500 pmol of Mar-Gly-Gln (R2 > 0.99). The assay also demonstrated good accuracy and precision, with an average percentage standard deviation for Gly-Gln over the range of the assay of less than 5%. A combination of multiple reaction monitoring (MRM) fragment ratio normalization and chromatographic peak shifting was used to ensure that the LC-MS/MS peak for Mar-Gly-Gln was free from possible isobar interferences. This assay was then demonstrated for the determination of in vivo Gly-Gln levels in P and Sprague-Dawley rat cortex and nucleus accumbens samples. 相似文献

Delayed cerebral ischemia resulting from extracellular hemoglobin is an important determinant of outcome in subarachnoid hemorrhage. Hemoglobin is scavenged by the CD163-haptoglobin system in the circulation, but little is known about this scavenging pathway in the human CNS. The components of this system were analyzed in normal cerebrospinal fluid and after subarachnoid hemorrhage. The intrathecal presence of the CD163-haptoglobin-hemoglobin scavenging system was unequivocally demonstrated. The resting capacity of the CD163-haptoglobin-hemoglobin system in the normal CNS was 50 000-fold lower than that of the circulation. After subarachnoid hemorrhage, the intrathecal CD163-haptoglobin-hemoglobin system was saturated, as shown by the presence of extracellular hemoglobin despite detectable haptoglobin. Hemoglobin efflux from the CNS was evident, enabling rescue hemoglobin scavenging by the systemic circulation. Therefore, the CNS is not capable of dealing with significant intrathecal hemolysis. Potential therapeutic options to prevent delayed cerebral ischemia ought to concentrate on augmenting the capacity of the intrathecal CD163-haptoglobin-hemoglobin scavenging system and strategies to encourage hemoglobin efflux from the brain. 相似文献

Sexual conflict can drive intersexual arms races, with female resistance and male persistence traits coevolving antagonistically. Such arms races are well documented in some diving beetles, although the extent of sexual conflict in this family remains unclear. The European dytiscid Agabus uliginosus has a strikingly dimorphic female; individuals from most regions are smooth and male‐like, whereas those from some populations have a strongly roughened dorsum, a trait that has attracted the name dispar. We demonstrate that rough and smooth females differ consistently in the development of dorsal surface microreticulation, and that these females are associated with males that differ in the development of their persistence traits. These findings extend the occurrence of pre‐insemination sexual conflict and associated intrasexual dimorphism in Dytiscidae, and suggest that such mating systems are relatively widespread in these beetles. 相似文献

Managed wetlands provide critical foraging and roosting habitats for shorebirds during migration; therefore, ensuring their availability is a priority action in shorebird conservation plans. Contemporary shorebird conservation plans rely on a number of assumptions about shorebird prey resources and migratory behavior to determine stopover habitat requirements. For example, the US Shorebird Conservation Plan for the Southeast-Caribbean region assumes that average benthic invertebrate biomass in foraging habitats is 2.4 g dry mass m?2 and that the dominant prey item of shorebirds in the region is Chironomid larvae. For effective conservation and management, it is important to test working assumptions and update predictive models that are used to estimate habitat requirements. We surveyed migratory shorebirds and sampled the benthic invertebrate community in coastal managed wetlands of South Carolina. We sampled invertebrates at three points in time representing early, middle, and late stages of spring migration, and concurrently surveyed shorebird stopover populations at approximately 7-day intervals throughout migration. We used analysis of variance by ranks to test for temporal variation in invertebrate biomass and density, and we used a model based approach (linear mixed model and Monte Carlo simulation) to estimate mean biomass and density. There was little evidence of a temporal variation in biomass or density during the course of spring shorebird migration, suggesting that shorebirds did not deplete invertebrate prey resources at our site. Estimated biomass was 1.47 g dry mass m?2 (95 % credible interval 0.13–3.55), approximately 39 % lower than values used in the regional shorebird conservation plan. An additional 4728 ha (a 63 % increase) would be required if habitat objectives were derived from biomass levels observed in our study. Polychaetes, especially Laeonereis culveri (2569 individuals m?2), were the most abundant prey in foraging habitats at our site. Polychaetes have lower caloric content than levels assumed in the regional plan; when lower caloric content and lower biomass levels are used to determine habitat objectives, an additional 6395 ha would be required (86 % increase). Shorebird conservation and management plans would benefit from considering the uncertainty in parameters used to derive habitat objectives, especially biomass and caloric content of prey resources. Iterative testing of models that are specific to the planning region will provide rapid advances for management and conservation of migratory shorebirds and coastal managed wetlands. 相似文献

Most shallow-water teleosts have moderate levels of trimethylamine N-oxide (TMAO; approximately 50 mmol/kg wet mass), a common osmolyte in many other marine animals. Recently, muscle TMAO contents were found to increase linearly with depth in six families. In one hypothesis, this may be an adaptation to counteract the deleterious effects of pressure on protein function, which TMAO does in vitro. In another hypothesis, TMAO may be accumulated as a by-product of acylglycerol (AG) production, increasing with depth because of elevated lipid metabolisms known to occur in some deep-sea animals. Here we analyze muscle TMAO contents and total body AG (mainly triacyglycerol [TAG]) levels in 15 species of teleosts from a greater variety of depths than sampled previously, including eight individual species caught at two or more depths. Including data of previous studies (total of 17 species, nine families), there is an apparent sigmoidal increase in TMAO contents between 0- and 1.4-km depths, from about 40 to 150 mmol/kg. From 1.4 to 4.8 km, the increase appears to be linear (r2=0.91), rising to 261 mmol/kg at 4.8 km. The trend also occurred within species: in most cases in which a species was caught at two or more depths, TMAO was higher in the deeper-caught specimens (e.g., for Coryphaenoides armatus, TMAO was 173, 229, and 261 mmol/kg at 1.8, 4.1, and 4.8 km, respectively). TMAO contents not only were consistent within species at a given depth but also did not vary with season or over a wide range of body masses or TAG contents. Thus, no clear link between TMAO and lipid was found. However, TMAO contents might correlate with the rate (rather than content) of TAG production, which could not be quantified. Overall, the data strongly support the hypothesis that TMAO is adaptively regulated with depth in deep-sea teleosts. Whether lipid metabolism is the source of that TMAO is a question that remains to be tested fully. 相似文献

Marine sediments are frequently covered by mats of the filamentous Beggiatoa and other large nitrate-storing bacteria that oxidize hydrogen sulfide using either oxygen or nitrate, which they store in intracellular vacuoles. Despite their conspicuous metabolic properties and their biogeochemical importance, little is known about their genetic repertoire because of the lack of pure cultures. Here, we present a unique approach to access the genome of single filaments of Beggiatoa by combining whole genome amplification, pyrosequencing, and optical genome mapping. Sequence assemblies were incomplete and yielded average contig sizes of approximately 1 kb. Pathways for sulfur oxidation, nitrate and oxygen respiration, and CO2 fixation confirm the chemolithoautotrophic physiology of Beggiatoa . In addition, Beggiatoa potentially utilize inorganic sulfur compounds and dimethyl sulfoxide as electron acceptors. We propose a mechanism of vacuolar nitrate accumulation that is linked to proton translocation by vacuolar-type ATPases. Comparative genomics indicates substantial horizontal gene transfer of storage, metabolic, and gliding capabilities between Beggiatoa and cyanobacteria. These capabilities enable Beggiatoa to overcome non-overlapping availabilities of electron donors and acceptors while gliding between oxic and sulfidic zones. The first look into the genome of these filamentous sulfur-oxidizing bacteria substantially deepens the understanding of their evolution and their contribution to sulfur and nitrogen cycling in marine sediments. 相似文献
