Protein synthesis with membrane-bound polysomes and albumin messenger RNA from livers of mutant mice
Summary Investigation of deficiencies in serum protein synthesis resulting from deletion-mutations at the albino locus in mice was
continued usingin vitro conditions. Previous work showed that although total protein synthesis was only slightly lower in livers from albinos, newly
synthesized protein appearing in plasma was 22% of that in controls. It was thought that the disorganized endoplasmic reticulum
and Golgi apparatus, characteristic for the liver (and kidney) of these mutants, might be responsible for the observed deficiencies.
In the present study mebrane-bound polysomes isolated from the livers of newborn albinos were 55% (c3H/c3H strain) and 62% (c14CoS/c14CoS strain) as efficient as those from normal littermates in incorporating radioactive leucine into protein in a cell-free system.
These differences could not be eliminated by the addition of excess liver mRNA, exogenous soluble factors or by the exchange
of cell sap between albino and control polysomes. In an earlier study albino liver slices synthesized only 13% (or 17% per
mg of total protein synthesized) as much albumin as controls. We have now found that the level of albumin poly(A)+-RNA isolated from albino livers and assayed with a reticulocyte lysate, was almost as high (85%) as in controls. It was concluded
that the very low level of albumin synthesis in albino livers did not result from a deficiency of albumin mRNA. Whether the
rate-limiting step in synthesis of albumin in mutant livers is at the level of translation or processing for secretion requires

Summary We examined heritabilities and correlations among measures of locomotor performance (speed, stamina) and among possible morphometric determinants of performance (hindlimb span, tail length) in families of hatchling lizards (Sceloporus occidentalis). We were particularly interested in determining whether these traits were heritable and thus might potentially respond genetically to selection. Moreover, we wished to determine whether speed and stamina are negatively genetically correlated, as suggested bya priori physiological and empirical considerations. All four traits appeared to be significantly heritable. Broadsense heritabilities were 0.33–0.36 for speed, 0.35–0.36 for stamina, 0.45–0.51 for hindlimb span, and 0.46–0.47 for tail length. Contrary to expectations, speed and stamina were not negatively genetically correlated. Hindlimb span and tail length, however, were negatively genetically correlated (but not phenotypically correlated). Hindlimb span and stamina were positively phenotypically correlated. Thus, for example, selection for longer hindlimb span could potentially result in shorter tails, contrary to evolutionary predictions based only on phenotypic correlations. 相似文献

A gas chromatographic method for the analysis of cresol metabolites of toluene and [2H8]toluene in urine was developed. Cresol glucuronides and sulfates in urine were hydrolyzed with β-glucuronidase and arylsulfatase. Following extraction with tert.-butyl methyl ether and solvent exchange into benzene, the cresols were derivatized with heptafluorobutyric anhydride to form the heptafluorobutyrate esters. The derivatives were analyzed by gas chromatography with electron capture detection. Chromatographic resolution was achieved between all cresol isomers and their 2H7 analogs. Calibration ranged from 0.001 to 500 μg/ml. Recoveries were 55–97% and showed no trend with respect to analyte concentration. Within-day precision of analyses of benchmark urine samples had a coefficient of variation of less than 4%. The assay sensitivity was limited by chromatographic background but was sufficient for quantification of the unlabeled cresols in urine from men with only environmental exposure to toluene. Average levels in urine samples from 45 men were 0.023, 0.054 and 37 μg/ml for o-, m- and p-cresol, respectively. 相似文献

Naphthalene is toxic to the eye and results in opacification of the lens. To investigate the metabolic events that may be occurring in the lens epithelial cells, a cell line of lens from a transgenic mouse was incubated with various metabolites of naphthalene. Naphthoquinone at 50 microM was toxic to most cells with a depletion of glutathione levels noted within 6 h of incubation. At 10 microM, naphthoquinone caused an increase in specific activity of the enzyme DT-diaphorase. This enzyme is thought to be a defense against quinones since semiquinone formation is thought to be lessened. Naphthalene-1,2-dihydrodiol at 50 microM also caused an increase in the specific activity of the DT-diaphorase, while at 10 microM no apparent change occurred in the cells. Although there was evidence of metabolic alterations in the cells with the metabolites of naphthalene, the protein profile by two-dimensional gel electrophoresis did not change and there was no indication of an increase in carbonyl formation in the soluble proteins of the cells. These experiments indicate that the metabolites of naphthalene can cause alteration in the metabolism of the lens cells but may not cause apparent changes in the major proteins within the lens epithelium. 相似文献
