Secondary-site attachment of coliphage lambda near the thr operon   总被引:4,自引:0,他引:4  
Phage λ has been integrated at a secondary attachment site near the thr oporon of Escherichia coli. The integration caused a pleiotropic effect since both thrA and thrB enzyme activities were lost. Lysogenic, thr+ revertants regained thrA and thrB activities although the enzymes were synthetized constitutively at low levels. Four classes of prophage deletion strains were isolated with deletions extending into trpR and the thr operon. Genetic and biochemical analyses indicated a gene order: trpR .. λ .. thrA .. B .. C. Defective transducing phage carrying thr A, B,C were also isolated.  相似文献

Summary The red-absorbing form (P r ) and the far-red absorbing form (P fr ) of undergraded, high-molecular-weight phytochrome from rye (Secale cereale L.) seedlings were examined for their reactivity toward N-ethyl-[14C]maleimide ([14C]-NEM). After pre-treatment of P r with cold NEM and extensive dialysis, photoconversion to P fr and treatment with [14C]NEM resulted in an approximately 70% increase in incorporation of radioactivity over the dark control. These results are discussed in relation to the view that phytochrome undergoes a protein conformational change upon phototransformation.  相似文献

Scanning and transmission electron microscopy were utilized in studying the cephalic abnormalities of the tu-h strain of Drosophila melanogaster. Abnormalities, appearing as protuberances (growths) of different shapes and sizes, were observed only on or in close proximity to receptor cites. Compound eyes of some flies, besides having such protuberances, either had modified corneal lenses or were absent altogether. Cuticles of the growths were thinner than that of the normal surrounding layer. The epidermal cells associated with setae on the growths or underlying modified corneal lenses remained undifferntiated. Both setae and growths lacked innervation. The receptor portions (retinular sensory cells and secondary pigment cells) of the abnormal compound eye, where the dioptric portions were replaced by protuberances or remained undifferentiated, were unaffected by the mutation. Nuclei of several cells, including oenocytes and fat bodies, close to or underlying epidermal cells of abnormalities, were large and contained compact nucleoli without nucleonemas. Viruslike particles were observed in several nuclei of growth cells. It was concluded from the present study that the alterations induced by the mutation can be only a localized phenomenon restricted to at least several cephalic epidermal cells and/or their derivatives.  相似文献

When hypothyroid rat liver nuclei labeled in vivo with [125 I]L-triiodothyronine are incubated with micrococcal nuclease, the nuclear chromatin is digested and chromatin particles are released into the medium. The nuclease-treated nuclei contain intact nucleoli and a residual chromatin fraction. When this residual chromatin is purified, it contains only a small percentage of the initial nuclear DNA but is strikingly enriched in [125 I]L-triiodothyronine. This chromatin fraction has many of the characteristics of nucleolar chromatin including a high protein to DNA ratio, an abundance of nonhistone proteins, and a relatively high RNA to DNA ratio. An association of thyroid hormone receptors with a nucleolar component implicates this organelle in the early events of thyroid hormone action.  相似文献

Sprague-Dawley and Fischer rat embryo cells became spontaneously transformed about 20 passages after release of endogenous ecotropic type C virus (SD-RaLV and F-RaLV). The virus-producing transformed cells showed loss of contact inhibition, increased growth rate, and tumorigenicity in vivo. Exogenous infection of other Fischer rat embryo cultures in early passage with SD-RaLV and F-RaLV markedly accelerated their rates of transformation.  相似文献

One case of giant-cell pneumonia and two of encephalopathy, all due to measles infection in children in leukaemic remission on immunosuppressive treatment, were seen recently. The clinical syndromes were variable and atypical and the antibody responses unpredictable. Conventional doses of pooled immunoglobulin failed to protect the two children to whom it was given. Degeneration rather than inflammation seems to characterise the encephalopathy in immunosuppressed children infected with measles virus.  相似文献

Eight out of 115 infants admitted to hospital with respiratory syncytial (RS) virus infection had been breast-fed compared with 46 out of 167 controls; this difference was statistically significant. Twenty-one specimens of human colostrum were examined, and all contained RS virus neutralising activity. Specific IgA and IgG were detected in 18 specimens, whereas IgM was detected in none. The titre of IgA antibody was usually higher and correlated more closely to the titre of neutralising activity than that of IgG. Infants inhale milk feeds and regurgitate them through the nose, and the IgA collecting in the respiratory tract might protect against severe respiratory infection. Alternatively, if severe RS virus illness is a sign of hypersensitivity to the virus breast-feeding might protect the infant from an early sensitising infection.  相似文献

The methodology of density labeling of proteins by biosynthetic incorporation of 2H, 13C, 15N-amino acids into newly synthesized polypeptide chains allows the direct measurement of the turnover rate of the acetylcholine receptor in cultured chick skeletal muscle. In this study, receptors synthesized in medium containing 2H, 13C, 15N-amino acids were resolved from 1H, 12C, 14N-receptors by velocity sedimentation in sucrose-deuterium oxide gradients, and their proportions were determined by computer analysis of the gradient profiles. The kinetics of turnover of acetylcholine receptors are identical for developing chick muscle fibers grown in medium containing 2H, 13C, 15N-amino acids or 1H, 12C, 14N-amino acids, and the high degree of substitution of normal aminoacyl residues by 2H, 13C, 15N-residues does not affect the turnover rate of the denser receptor. Comparison of the turnover rates in continuous and pulse-labeling experiments gave independent confirmation of these results. The application of a potent, essentially irreversible blocking agent, alpha-bungarotoxin, increases the median lifetime of receptors from 17 hr for the native unbound receptor to 22 hr for the alpha-bungarotoxin-receptor complex. As predicted, the total number of alpha-bungarotoxin binding sites increased in the continued presence of alpha-bungarotoxin due to extension of receptor lifetime. To determine whether other cholinergic agents affect the turnover rate of the receptor, measurements were performed on cultures grown in the presence of 10(-4) M d-tubocurare or 10(-4) M carbachol, a reversible antagonist and a reversible agonist, respectively, of the nicotinic acetylcholine receptor. The receptor degradation rates of the drug-treated cells were identical to control values. The total number of alpha-bungarotoxin binding sites was reduced by 30% in the presence of carbachol, indicating that this agent affects the rate of synthesis of the acetylcholine receptor. Data formerly interpreted as suggesting a cycling of receptor-containing plasma membrane out of and back into the sarcolemma are now understood to reflect the alteration in receptor lifetime upon complexing with alpha-bungarotoxin. The intracellular "hidden" receptor sites were found to remain inside the myotubes and thus do not signify an intracellular pool of recycling plasma membrane.  相似文献
