Background

Anadromous migratory fish species such as Atlantic salmon (Salmo salar) have significant economic, cultural and ecological importance, but present a complex case for management and conservation due to the range of their migration. Atlantic salmon exist in rivers across the North Atlantic, returning to their river of birth with a high degree of accuracy; however, despite continuing efforts and improvements in in-river conservation, they are in steep decline across their range. Salmon from rivers across Europe migrate along similar routes, where they have, historically, been subject to commercial netting. This mixed stock exploitation has the potential to devastate weak and declining populations where they are exploited indiscriminately. Despite various tagging and marking studies, the effect of marine exploitation and the marine element of the salmon lifecycle in general, remain the "black-box" of salmon management. In a number of Pacific salmonid species and in several regions within the range of the Atlantic salmon, genetic stock identification and mixed stock analysis have been used successfully to quantify exploitation rates and identify the natal origins of fish outside their home waters - to date this has not been attempted for Atlantic salmon in the south of their European range.

Results

To facilitate mixed stock analysis (MSA) of Atlantic salmon, we have produced a baseline of genetic data for salmon populations originating from the largest rivers from Spain to northern Scotland, a region in which declines have been particularly marked. Using 12 microsatellites, 3,730 individual fish from 57 river catchments have been genotyped. Detailed patterns of population genetic diversity of Atlantic salmon at a sub-continent-wide level have been evaluated, demonstrating the existence of regional genetic signatures. Critically, these appear to be independent of more commonly recognised terrestrial biogeographical and political boundaries, allowing reporting regions to be defined. The implications of these results on the accuracy of MSA are evaluated and indicate that the success of MSA is not uniform across the range studied; our findings indicate large differences in the relative accuracy of stock composition estimates and MSA apportioning across the geographical range of the study, with a much higher degree of accuracy achieved when assigning and apportioning to populations in the south of the area studied. This result probably reflects the more genetically distinct nature of populations in the database from Spain, northwest France and southern England. Genetic stock identification has been undertaken and validation of the baseline microsatellite dataset with rod-and-line and estuary net fisheries of known origin has produced realistic estimates of stock composition at a regional scale.

Conclusions

This southern European database and supporting phylogeographic and mixed-stock analyses of net samples provide a unique tool for Atlantic salmon research and management, in both their natal rivers and the marine environment. However, the success of MSA is not uniform across the area studied, with large differences in the relative accuracy of stock composition estimates and MSA apportioning, with a much higher degree of accuracy achieved when assigning and apportioning to populations in the south of the region. More broadly, this study provides a basis for long-term salmon management across the region and confirms the value of this genetic approach for fisheries management of anadromous species.  相似文献

以北京九龙山自然保护区幼龄侧柏人工林为研究对象,对其不同生长季节叶、枝、根(0—10 cm、10—20 cm土层)的碳(C)、氮(N)、磷(P)含量及其生态化学计量学特征进行了分析,深入探讨了生长季节与器官以及两因素交互作用对以上特征的影响,研究有助于理解植物各性状之间的相互作用以及植物生长过程中资源的利用和分配状况。结果表明:1)不同器官间C含量为414.97—461.58 g/kg,枝最大,根(0—10 cm)最小;N含量为6.57—14.28 g/kg,叶最大,枝最小;P含量为0.39—1.28 g/kg,叶最大,根(10—20 cm)最小;C∶N为31.76—70.98,枝最大,叶最小;C∶P为369.93—1099.20,根(10—20 cm)最大,叶最小;N∶P为9.21—23.81,根(0—10 cm)最大,枝最小。整个生长季节中侧柏各器官C含量最稳定,变异系数均小于7%;P含量变异性最大,变异系数均超过15%,N含量变异性介于两者之间;各器官中C∶N和N∶P较C∶P更为稳定,C、N与P具有较好的耦合协同性,C∶P和N∶P的变化主要取决于P的变化。2)器官对C、N、P含量及其化学计量关系均存在显著影响,生长季节对N和P含量存在显著影响,两者交互作用只对P含量存在显著影响,器官对侧柏C、N、P含量及其化学计量变异的贡献大于生长季节。3)侧柏各器官间C、N、P含量及其化学计量比相关性多数未达到显著性水平,仅有叶与枝中的P及C∶P显著相关,说明侧柏器官分化过程中各器官对元素的吸收利用具有特异性。侧柏叶片N∶P14,说明生长季节里幼龄侧柏人工林更多受到N限制。  相似文献

Genus Merluccius is considered one of the most important groups within the Teleostei, and comprises 12 extant species distributed along the coasts of America, Europe and Africa, being its fisheries very important in these continents. Despite their noticeable economical importance for humans, to date the phylogeny of hakes has not been clearly established. In this study we used mitochondrial sequences (the ribosomal genes 12S rDNA and 16S rDNA, the coding gene cytochrome b and the control region) and the nuclear 5S rDNA conserved region in order to determine the phylogenetic and biogeographical relationships within the genus Merluccius. This is the first time that all the species of this genus recognized by the FAO are included in a phylogeny. Maximum Parsimony, Maximum Likelihood and Bayesian analyses of the mitochondrial sequences suggest that the geographical origin of the genus was the North Atlantic Ocean, and indicate that two main clades, early separated in the evolution, exist within the genus: one American (7 species) and one Euro-African (5 species). Among the American species, M. bilinearis seems to be the most ancient one, and the rise of the Panama Isthmus could act as a physical barrier leading to further processes of speciation. Within the Euro-African clade, successive events of geographical differentiation could explain the observed pattern of species distribution. Therefore, we propose both vicariant speciation and geographical dispersion as main mechanisms to explain the evolutionary history of the genus Merluccius.  相似文献

In situ RNA hybridization and immunocytochemistry were used to establish the cellular distribution of monoterpenoid indole alkaloid biosynthesis in Madagascar periwinkle (Catharanthus roseus). Tryptophan decarboxylase (TDC) and strictosidine synthase (STR1), which are involved in the biosynthesis of the central intermediate strictosidine, and desacetoxyvindoline 4-hydroxylase (D4H) and deacetylvindoline 4-O-acetyltransferase (DAT), which are involved in the terminal steps of vindoline biosynthesis, were localized. tdc and str1 mRNAs were present in the epidermis of stems, leaves, and flower buds, whereas they appeared in most protoderm and cortical cells around the apical meristem of root tips. In marked contrast, d4h and dat mRNAs were associated with the laticifer and idioblast cells of leaves, stems, and flower buds. Immunocytochemical localization for TDC, D4H, and DAT proteins confirmed the differential localization of early and late stages of vindoline biosynthesis. Therefore, we concluded that the elaboration of the major leaf alkaloids involves the participation of at least two cell types and requires the intercellular translocation of a pathway intermediate. A basipetal gradient of expression in maturing leaves also was shown for all four genes by in situ RNA hybridization studies and by complementary studies with dissected leaves, suggesting that expression of the vindoline pathway occurs transiently during early leaf development. These results partially explain why attempts to produce vindoline by cell culture technology have failed.  相似文献

Life history traits determine fitness and hence the ability of populations and species to survive through adverse conditions. Therefore, identifying temporal changes in life history traits over large timescales is necessary to understand and predict the effect of current global change on wild populations. In this study, we compare life history traits between Palaeolithic and present-day vertebrates, analysing the number of winters spent in the river and at sea by North Iberian salmonids (Salmo salar and Salmo trutta) from two separate time frames: the Upper Palaeolithic, based on archaeological remains, and the present day, based on sport catches. The river stage did not change significantly, but the marine period has been shortened in modern anadromous specimens, accordingly shortening mean generation time. Population growth rates have been estimated through Bayesian analyses of the mitochondrial DNA control region of modern specimens for the two Salmo species using two different mutation rates (1% and 3.6%). Coincidence of coalescent N(e) estimates with independent N(e) calculated from catches suggests that the 3.6% mutation rate fits better the evolution of the studied populations. Population growth rate declines would have occurred in the last millennium and could be explained by a combination of climatic events and anthropogenic activities.  相似文献

目的:制备链亲和素标记的人白细胞介素-2(SA-hIL2)融合蛋白,并研究其生物学功能。方法:构建SA-L-IL2-pET24重组表达质粒,在大肠杆菌中表达SA-hIL2融合蛋白,对表达的SA-hIL2融合蛋白采用镍金属螯合(Ni-NTA)层析柱进行纯化,透析复性。CCK-8法检测SA-hIL2融合蛋白对PHA刺激的人外周血淋巴细胞的增值活性,流式细胞仪分析SA-hIL2融合蛋白对生物素化的B16.F10肿瘤细胞表面锚定修饰效率。结果:SA-hIL2在大肠杆菌中实现了高效表达,约占菌体总蛋白的20%,制备的SA-hIL2融合蛋白纯度达到95%,并具有双重活性,即hIL-2促进PHA刺激的人外周血淋巴细胞的增值活性和SA介导的高效结合至已生物素化的B16.F10肿瘤细胞表面的功能(表面锚定修饰效率约95%)。结论:研制的SA-hIL2融合蛋白具有双重活性,可为研制表面修饰的新型肿瘤细胞疫苗提供基础。  相似文献

We analysed micronuclei in brown trout Salmo trutta specimens sampled in the Trubia River, upstream and downstream of the emissions from a Spanish military factory to assess genotoxicity risks derived from military wastes. A significant exponential increase in micronuclei counts was found in fish living downstream of the military wastes with respect to fishes inhabiting upstream areas of the same river. In comparison, we only found a linear increase in micronuclei counts in a control stream where an old military factory had been demolished 6 months before sampling. This difference suggests that active discharge of armament factory wastes can directly induce micronuclei and therefore represents a genotoxic risk for the ecosystem.  相似文献
