Summary Bioelectrical parameters and unidirectional sodium and chloride fluxes were measured under voltageclamp conditions in groups of lizards submitted to single or chronic aldosterone treatment. Both acute (AT) and chronic (CT) treatment induced significant increases in the short-circuit current (I sc), as well as in the mucosa-to-serosa (J m-s Na ) and net sodium flux (J net Na ). In AT tissues, aldosterone did not change net chloride flux (J net Cl ) but did so in CT tissues. Amiloride reduced the aldosterone-increased I sc in AT and CT tissues, inhibited J net Na in AT tissues and abolished it in CT colons. J net Cl was also reduced by the diuretic in the group of AT colons, whereas no changes were observed in the CT tissues. Addition of luminal DIDS reduced Na+ absorption and totally inhibited Cl- absorption in the AT tissues, but did not change I sc. However, in CT tissues neither Na+ nor Cl- transport were affected by DIDS. A good relationship between I sc and J m-s Na was apparent after DIDS treatment in AT tissues. In this group, simultaneous addition of DIDS and amiloride totally abolished J net Na and reduced I sc to untreated control values. Addition of serosal ouabain abolished I sc and Na+ absorption in AT and CT colons, but Cl- absorption was only altered in AT tissues. Summary The white-rot fungus Ganoderma australis selectively degrades lignin in the ecosystem palo podrido. Using conditions that simulate those of palo podrido in the laboratory, it was found that low nitrogen content and low O2 tension stimulate the productio of manganese peroxidase and lignin degradation, and depress cellulose degradation and cellulase production. The inverse is found at high nitrogen concentration and high O2 tension. This agrees with previous results indicating that low O2 tension and low nitrogen stimulate selective lignin degradation by this fungus. Correspondence to: J. Eyzaguirre  相似文献

The oxygenation of trout red cells opens a Cl-dependent K pathway inhibited by furosemide, and by inhibitors of the erythrocyte anion exchanger such as DIDS and niflumic acid. The trigger is the deoxy-oxy conformational change of hemoglobin. The binding of carbon monoxide to heme, which induces a similar conformational change, mimics the effect of oxygen. The possible mechanisms enabling molecular oxygen to control the transport protein are discussed. This oxygenation-activated K transport appears to play a regulatory role in the control of the extracellular K concentration.  相似文献

An improved method for the isolation of leaf mesophyll protoplasts ofSolanum tuberosum L. cvs. Xenia and Bintje, their culture and regeneration to plants is described. The improvements involve the culture of nodal stem explants on Schenk and Hildebrandt (SH) medium supplemented with AgNO3 (7.5 mg 11) and Alar 85 (5 mg 1?1), and the pre-treatment of donor plants for 2–3 weeks at a short photoperiod (6 h), a low irradiance (17 (μmol m?2s?1) and a low temperature (20 °C). Regeneration of cell wals was observed within 2–3 d and cell divisions within 7–12 d after resuspending freshly isolated protoplasts in Sidorov et al. (SW) medium. The resulting microcalli were cultured on solid media containing zeatin (1–2,5 mg 1?1) and/or NAA (0.1 and/or 0.01 mg 1?1) and subsequently on medium with BAR (0.25 mg1?1) and GA?3 (0.1 mg 11) for shoot regeneration.  相似文献

The paper summarizes data concerning the biology and ecology ofOenanthe aquatica. The species is commonly distributed all over Czechoslovakia, from lowlands to the submontane belt, especially in fishpond and river basins.O. aquatica shows no special relationship to the chemical and physical soil properties, and is well adapted to habitats with a changing water level. The reproduction ofO. aquatica depends on emerging of the bottom. The most developed populations are formed by biennial plants and arise in the year following summer or autumn drainage.  相似文献

The inhibitory capacity of strain S-48 ofEnterococcus faecalis ssp.liquefaciens was studied. The strain produces a broad-spectrum peptide antibiotic (AS-48) that has been characterized elsewhere. The isolation of mutants from S-48 after mutagenic treatment revealed another inhibitory substance which remained masked in the wild strain. The protein nature and restricted spectrum of this substance points to its being a bacteriocin (Bc-48).  相似文献

Purified transverse tubule membranes from normal and dystrophic chicken skeletal muscle were isolated by a calcium-loading procedure. Normal and dystrophic T-tubules were similar in cholesterol content and (Na+,K+)-ATPase and 5'-nucleotidase activities but a significant decrease of Mg2(+)-ATPase activity was observed in dystrophic membranes. A comparative analysis of the enzyme properties revealed that the kinetic parameters were altered in dystrophic T-tubules and the ATP-hydrolyzing activity was differently affected by the ionic strength. However, the influence of temperature and the regulatory effect of concanavalin A were the same as in normal T-tubules. Membrane fluidity was similar in both preparations as estimated by fluorescence polarization of 1,6-diphenyl-1,3,5-hexatriene and trimethylammonium diphenylhexatriene. These results point to an impairment in the function of Mg2(+)-ATPase due to structural alterations of the enzyme.  相似文献

Summary The distribution of three biochemical markers, U-1, CM-4 and Aphv-a, -b, among wheat-Aegilops addition lines carrying Mv chromosomes from Aegilops ventricosa (genomes DvMv) has been investigated. Addition lines which had been previously grouped together on the basis of common non-biochemical characters carried marker U-1, a protein component from the 2M urea extract. The added chromosome, in the appropriate genetic background, seems to confer a high level of resistance to the eyespot disease, caused by the fungus Cercosporella herpotrichoides. The other two markers were concomitantly associated with another similarly formed group of addition lines. Both CM-4, a protein component from the chloroform:methanol extract, and Aphv-a, -b, alkaline phosphate isozymes, have been previously shown to be associated with homoeologous chromosome group 4, which suggests that the added chromosome in the second group of addition lines is 4Mv.  相似文献
