The Vitiligo Global Issues Consensus Conference (VGICC), through an international e‐Delphi consensus, concluded that ‘repigmentation’ and ‘maintenance of gained repigmentation’ are essential core outcome measures in future vitiligo trials. This VGICC position paper addresses these core topics in two sections and includes an atlas depicting vitiligo repigmentation patterns and color match. The first section delineates mechanisms and characteristics of vitiligo repigmentation, and the second section summarizes the outcomes of international meeting discussions and two e‐surveys on vitiligo repigmentation, which had been carried out over 3 yr. Treatment is defined as successful if repigmentation exceeds 80% and at least 80% of the gained repigmentation is maintained for over 6 months. No agreement was found on the best outcome measure for assessing target or global repigmentation, therefore highlighting the limitations of e‐surveys in addressing clinical measurements. Until there is a clear consensus, existing tools should be selected according to the specific needs of each study. A workshop will be conducted to address the remaining issues so as to achieve a consensus.  相似文献

该文综述了北京地区全新世以来植被演替和气候变化的相关研究资料,这些资料反映了当前阶段对该地区该时段植被与气候环境格局特征的认识。北京地区全新世早期(约12 000–8 000 cal a B.P.)植被为森林草地和/或针叶树占主导的针阔混交林,森林中阔叶树类群逐渐增多,指示了气候由寒冷干燥转为温暖湿润;全新世中期(约8 000–2 000 cal a B.P.前后)植被为暖温带针阔混交林,指示暖湿气候;全新世晚期(约2 000 cal a B.P.以来)转为森林草地和/或针叶树占主导的针阔混交林,气候转向凉干。植被演替反映的湿润度变化与季风区其它地区变化趋势一致。值得注意的是,前人研究揭示北京地区山区与平原中植被类型和类群组成已经出现空间分异。今后如能深入开展定量古气候重建研究,有可能精确描述其气候变化的过程,增进中国不同地理单元同时期气候变化的对比。  相似文献

A well-designed three-way junction (TWJ) aptasensor for lysozyme detection was developed based on target-binding-induced conformational change of aptamer-complementary DNA (cDNA) as probe. A ferrocene (Fc)-tagged cDNA is partially hybridized with an anti-lysozyme aptamer to form a folded structure where there is a coaxial stacking of two helices and the third one at an acute angle. In addition, the fabrication of the sensor was achieved via the single-step method, which offered a good condition for sensing. In the absence of lysozyme, electron transfer (eT), through the coaxial two helices called "conductive path", is allowed between Fc-labeled moiety and the electrode. The binding of lysozyme to the aptamer blocks eT, leading to diminished redox signal. This aptasensor with an instinct signal attenuation factor shows a high sensitivity to lysozyme, and the response data is fitted by nonlinear least-squares to Hill equation. Detection limit is 0.2nM with a dynamic range extending to 100nM. Compared with existing electrochemical impedance spectroscopy (EIS)-based approaches, TWJ-DNA aptasensor was demonstrated to be more specific for detection and simpler for regeneration procedure.  相似文献

The random disposal of vegetable residues in north China has become an important obstacle for the sustainable development of vegetable industry. The composting treatment technology has been transformed into organic fertilizer by high temperature fermentation, which has become a widely promoted agricultural clean production technology. However, due to the voluntary nature of farmers' adoption and their reluctance to adopt, this study aimed at assessing the factors that influence their adoption will and evaluate the value of willingness to pay (WTP) for composting technology. Data were collected from 142 respondents through a household survey in Gaocheng District by using structural questionnaire of contingent valuation method (CVM). Some qualitative response models (Probit, Logistic and multiple linear regression models) were applied for examining the main factors influencing the vegetable residue compost adoption and estimating the WTP value. The findings showed that social resource factors play an important role in the respondents' behaviors toward composting technology adoption. The empirical results indeed highlighted that subsidy policy, top dressing time, age, scale, investment of irrigation and net income have significant positive influences on the WTP of compost, while work time and information sources have a negative impact. Government subsidy is a necessary premise for implementing the composting program. The governments should subsidize the remaining 97% of the construction costs to ensure the smooth implementation of composting technology. The findings specifically mentioned that the subsidy object should be the disadvantaged peasant groups with lower household income but more environmentally conscious. This paper is believed to not only assess the technical externality of vegetable residues for the first time but also provide decision reference for policymakers, especially in the background of rapid development of agricultural clean production technology, the accuracy and efficiency of subsidies should be improved.  相似文献

The metal ion coordinating properties of the ligands N,N-bis(2-methylquinoline)-2-(2-aminoethyl)pyridine (DQPEA) and N,N-bis(2-methylquinoline)-2-(2-aminomethyl)pyridine (DQPMA) are presented. DQPEA and DQPMA differ only in that DQPEA forms six-membered chelate rings that involve the pyridyl group, whereas DQPMA forms analogous five-membered chelate rings.These two ligands illustrate the application of a ligand design principle, which states that increase of chelate ring size in a ligand will result in increase in selectivity for smaller relative to larger metal ions. The formation constants (log K1) of DQPEA and DQPMA with Ni(II), Cu(II), Zn(II), Cd(II) and Pb(II) are reported. As expected from the applied ligand design principle, small metal ions such as Ni(II) and Zn(II) show increases in log K1 with DQPEA (six-membered chelate ring) relative to DQPMA (five-membered chelate ring), while large metal ions such as Cd(II) and Pb(II) show decreases in log K1 when the chelate ring increases in size. In order to further understand the steric origin of the destabilization of complexes of metal ions of differing sizes by the six-membered chelate ring of DQPEA, the structures of [Zn(DQPEA)H2O](ClO4)2 (1) [triclinic, , a = 9.2906(10), b = 10.3943(10), c = 17.3880(18) Å, α = 82.748(7)°, β = 88.519(7)°, γ = 66.957(6)°, Z = 4, R = 0.073] and [Cd(DQPEA)(NO3)2] (2) [monoclinic, C2/c, a = 22.160(3), b = 15.9444(18), c = 16.6962(18) Å, β = 119.780(3)°, Z = 8, R = 0.0425] are reported. The Zn in (1) is five-coordinate, with a water molecule completing the coordination sphere. The Cd(II) in (2) is six-coordinate, with two unidentate nitrates coordinated to the Cd. It is found that the bonds to the quinaldine nitrogens in the DQPEA complexes are considerably stretched as compared to those of analogous TPyA (tri(pyridylmethyl)amine) complexes, which effect is attributed to the greater steric crowding in the DQPEA complexes. The structures are analyzed for indications of the origins of the destabilization of the complex of the large Cd(II) ion relative to the smaller Zn(II) ion. A possible cause is the greater distortion of the six-membered chelate ring in (2) than in (1), as evidenced by torsion angles that are further away from the ideal values in (2) than in (1). Fluorescence properties of the DQPMA and DQPEA complexes of Zn(II) and Cd(II) are reported. It is found that the DQPEA complex of Zn(II) has increased fluorescence intensity compared to the DQPMA complex, while for the Cd(II) complex the opposite is found. This is related to the greater strain in the six-membered chelate ring of the Cd(II) DQPEA complex as compared to the Zn(II) complex, with resulting poorer overlap in the Cd-N bond, and hence greater ability to quench the fluorescence in the Cd(II) complex.  相似文献

Introduction

The Hepatitis B Virus (HBV) genome contains four ORFs, S (surface), P (polymerase), C (core) and X. S is completely overlapped by P and as a consequence the overlapping region is subject to distinctive evolutionary constraints compared to the remainder of the genome. Specifically, a non-synonymous substitution in one coding frame may produce a synonymous substitution in the alternative frame, suggesting a possible conflict between requirements for diversifying and purifying forces. To examine how these contrasting requirements are balanced within this region, we investigated the relationship amongst positive selection sites, conserved regions, epitopes and elements of protein structure to consider how HBV balances the contrasting evolutionary pressures.

Methodology/Results

323 HBV genotype D genome sequences were collected and analyzed to identify sites under positive selection and highly conserved regions. Epitopes sequences were retrieved from previously published experimental studies stored in the Immune Epitope Database. Predicted secondary structures were used to investigate the association between structure and conservation. Entropy was used as a measure of conservation and bivariate logistic regression was used to investigate the relationship between positive selection/conserved sites and epitope/secondary structure regions. Our results indicate: (i) conservation in S is primarily dictated by α-helix elements in the protein structure, (ii) variable residues are mainly located in PreS, the major hydrophilic region (MHR) and the C-terminus, (iii) epitopes in S, which are directly targeted by the host immune system, are significantly associated with sites under positive selection.

Conclusions

The highly variable spacer domain in P, which corresponds to PreS in S, appears to act as a harbor for the accumulation of mutations that can provide flexibility for conformational changes and responding to immune pressure.  相似文献

目的 探讨I型子宫内膜癌组织中HIF-1α的表达,以及调控Snail和E-cadherin对肿瘤侵袭性生物学行为的影响.方法 采用免疫组化方法,结合组织芯片技术,检测124例I型子宫内膜癌、28例内膜不典型增生、35例正常内膜组织中HIF-1α、Snail和E-cadherin的表达水平,分析三种蛋白表达之间的相关性及与临床病理因素的关系.结果 I型子宫内膜癌组织中HIF-1α、Snail和E-cadherin的表达率分别为61.3%、46.8%、36.3%,与正常内膜和不典型增生内膜组织相比,有显著统计学差异(P<0.01).HIF-1α表达与病理分级、肿瘤肌层浸润和淋巴结转移明显相关(P<0.05).Snail表达与FIGO分期、淋巴结转移明显相关(P<0.05).E-cadherin的缺失与肿瘤肌层浸润和淋巴结转移显著相关(P<0.01).I型子宫内膜癌组织中HIF-1α和Snail的表达呈明显正相关(r=0.214,P=0.017),而Snail和E-cadherin的表达存在负相关关系(r=–0.203,P=0.024).结论 HIF-1α可能通过上调Snail的表达和抑制E-cadherin的表达在I型子宫内膜癌发生、侵袭和转移中发挥重要作用.  相似文献

Background

Mesenchymal stem cells (MSC) represent a particularly attractive cell type for bone tissue engineering because of their ex vivo expansion potential and multipotent differentiation capacity. MSC are readily differentiated towards mature osteoblasts with well-established protocols. However, tissue engineering frequently involves three-dimensional scaffolds which (i) allow for cell adhesion in a spatial environment and (ii) meet application-specific criteria, such as stiffness, degradability and biocompatibility.

Methodology/Principal Findings

In the present study, we analysed two synthetic, long-term degradable polymers for their impact on MSC-based bone tissue engineering: PLLA-co-TMC (Resomer® LT706) and poly(ε-caprolactone) (PCL). Both polymers enhance the osteogenic differentiation compared to tissue culture polystyrene (TCPS) as determined by Alizarin red stainings, scanning electron microscopy, PCR and whole genome expression analysis. Resomer® LT706 and PCL differ in their influence on gene expression, with Resomer® LT706 being more potent in supporting osteogenic differentiation of MSC. The major trigger on the osteogenic fate, however, is from osteogenic induction medium.

Conclusion

This study demonstrates an enhanced osteogenic differentiation of MSC on Resomer® LT706 and PCL compared to TCPS. MSC cultured on Resomer® LT706 showed higher numbers of genes involved in skeletal development and bone formation. This identifies Resomer® LT706 as particularly attractive scaffold material for bone tissue engineering.  相似文献

WD repeat-containing protein 5 (WDR5) is a common component of mammalian mixed lineage leukemia methyltransferase family members and is important for histone H3 lysine 4 methylation (H3K4me), which has been implicated in control of activation of cell lineage genes during embryogenesis. However, WDR5 has not been considered to play a specific regulatory role in epigenetic programming of cell lineage because it is ubiquitously expressed. Previous work from our laboratory showed the appearance of histone H3K4me within smooth muscle cell (SMC)-marker gene promoters during the early stages of development of SMC from multipotential embryonic cells but did not elucidate the underlying mechanisms that mediate SMC-specific and locus-selective H3K4me. Results presented herein show that knockdown of WDR5 significantly decreased SMC-marker gene expression in cultured SMC differentiation systems and in Xenopus laevis embryos in vivo. In addition, we showed that WDR5 complexes within SMC progenitor cells contained H3K4 methyltransferase enzymatic activity and that knockdown of WDR5 selectively decreased H3K4me1 and H3K4me3 enrichment within SMC-marker gene promoter loci. Moreover, we present evidence that it is recruited to these gene promoter loci through interaction with a SMC-selective pituitary homeobox 2 (Pitx2). Taken together, studies provide evidence for a novel mechanism for epigenetic control of SMC-marker gene expression during development through interaction of WDR5, homeodomain proteins, and chromatin remodeling enzymes.  相似文献
