Changes in the level of dopamine and its metabolites in brain structures and immunocompetent organs in the formation of immune response

By the method of liquid chromatography with electrochemical detection dopamine level increase was shown within 2 min after immunization of rats with sheep red blood cells in n. caudatus. In 20 min after the immune response the acceleration of dopamine metabolism was observed in nuclei A9, A10, A11, accumbens, in amygdala, hypothalamus and hippocampus. In immunocompetent organs dopamine level was changed in 24 h after the immunization.     

Polypharmacy in the management of patients with schizophrenia on risperidone in a tertiary-care hospital in Malaysia

The present study was conducted primarily to determine the occurrence of polypharmacy in patients with schizophrenia on risperidone. The secondary aim was to ascertain the incidence of inappropriate prescribing with anticholinergics. A retrospective review of the medical records of all patients who were being followed up at the out-patient clinic of a tertiary-care hospital in Malaysia was conducted. Only patients who were being prescribed risperidone between 1 June 2008 and 31 December 2008 were included in the study. Demographic data such as patient’s age, gender and race were obtained from the patient’s medical records. In total, 113 patients met the selection criteria. Polypharmacy was found to occur in 34 patients (30.09%), with the majority (76.47%) being on two antipsychotics. In total, 27 patients (34.18%) on monotherapy with risperidone were prescribed an anticholinergic on scheduled dosing, while 19 patients (24.05%) were prescribed it on an as-needed basis. Of the patients on polypharmacy, 26 (76.47%) were on scheduled dosing of anticholinergics, while three (8.82%) were taking the medication on an as-needed basis. Polypharmacy should be avoided, and the use of anticholinergics should be closely reviewed. By adopting more efficient prescribing practices, costs can be reduced and financial resources can instead be channelled towards more beneficial areas for the patients.

Action of strong and weak nitrogenous bases on respiratory chain enzymes of mitochondria

The action of nitrogenous basis--electroneutral hydrazides (pK less than 7,50 and positive charged arylhydrazones (pK greater than 8)--on the respiratory chain enzymes and the influence of the electric charge and the size of alkoxylic group on biological activity compounds have been investigated. It has been shown that the size of alkoxylic group defines the selective action of nitrogenous basis on the enzymes of mitochondrial respiratory chain. The nitrogenous basis with a long alkoxylic group is shown to be inhibitors of NADH-dehydrogenase, their action is similar to rotenone. At the same time compounds with a short group are more effective in the inhibition of the enzymes of the initial segment in the respiratory chain mitochondria. The affinity of the organic cations of arylhydrazones to NADH-dehydrogenase is 100-1000 times higher than the affinity of electric neutral compounds.     

Effect of high-altitude hypoxia on the effectiveness of chemotherapy in tumors

It was shown in rat experiments that high-altitude hypoxia inhibits the growth of transplantable tumors and enhances the efficacy of antitumor treatment with chemotherapeutic drugs (thiotepa for Heren's carcinoma, and cyclophosphamide for sarcoma 45). Under high-altitude conditions, the cyclophosphamide toxicity as regards blood cells was modified, manifesting in the absence of anemization and less considerable reduction in the platelet and lymphocyte counts. The animals' adaptation to high-altitude hypoxia was followed by a rise in the blood content of sulfhydryl groups and increase in cancerolytic activity of the blood serum.
