Multiple forms of human renin. Purification and characterization.   总被引:10,自引:0,他引:10  
Human renin was purified from a juxtaglomerular cell tumor with a high renin content, 24.2 Goldblatt units/mg of protein. The purification procedure comprised three steps: gel filtration, DEAE-cellulose chromatography, and preparative isoelectric focusing. Five forms of renin amounting to 5.3 mg of enzyme were obtained with isoelectric points of 4.95, 5.10, 5.35, 5.55, and 5.70. They were all glycoproteins. The three major fractions had very similar specific activities, 868, 860, and 809 Goldblatt units/mg of protein. These fractions produced a single band on analytical isoelectric focusing and a single arc on immunoelectrophoresis. On polyacrylamide gel electrophoresis at pH 7.8, each fraction consisted of two renin bands with the same molecular weight, but different net charges. The molecular weight determined by gel filtration and Fergusson plot analysis on polyacrylamide gel was 38,000 to 42,000. The optimum pH determined on N-acetyltetradecapeptide substrate was 6.5, and the Km was 6.8 x 10(-6) M. These parameters were identical with those for standard human kidney renin. Antibodies raised against tumor renin completely inhibited the activity of both tumor and standard renin. Under dissociating conditions (sodium dodecyl sulfate-polyacrylamide gel electrophoresis and gel electrophoresis in the presence of 6 M urea), part of the purified enzyme dissociated into two smaller fragments (Mr = 20,000 and 25,000) containing renin activity.  相似文献

Chlorophyll was rapidly destroyed in the presence of bisulfite and linoleic acid hydroperoxide. Both bisulfite and linoleic acid hydroperoxide were required for chlorophyll destruction and both were consumed in the reaction; however, there was no oxygen requirement. Chlorophyll destruction occurred most readily in the slightly acidic pH region with little destruction occurring above pH 8. The free radical scavengers, hydroquinone and α-tocopherol, were very effective at inhibiting chlorophyll destruction, but the singlet oxygen quenchers, β-carotene, 2,5-dimethylfuran and 1,3-diphenylisobenzofuran, were only slightly effective. The addition of metal chelators indicated that metals were not participating in the reaction. The evidence indicates that chlorophyll was destroyed by a free radical mechanism. Based on the present results and that of others, it is suggested that chlorophyll was destroyed via oxidation by the alkoxy radical which was produced during the decomposition of linoleic acid hydroperoxide by bisulfite.  相似文献

The ultrastructure of mesophyll chloroplasts of maize (Zea mays L.) was more severely affected by iron deficiency that induced mild chlorosis than was the ultrastructure of bundle sheath plastids. Ferredoxin and ribulose diphosphate carboxylase levels were severely decreased by iron deficiency. Malic enzyme was less affected, and phosphoenolpyruvate carboxylase activity remained high even under severe iron deficiency. Iron deficient leaves fixed carbon into malic and aspartic acids but the rate of entrance of carbon into the sugar phosphates and sucrose was greatly reduced compared to the control. Chlorophyll a/b ratios ranged from low values of less than 2 in severely iron deficient leaves to high values exceeding 4 in leaves showing little iron deficiency.  相似文献

Transect surveys were carried out in northern Argentina during October and November 1977 in order to determine the distribution and abundance ofAotus trivirgatus. The monkeys were seen in pairs with one to two recent young and occurred at a density of approximately six family groups/km2.Aotus was only found in relatively moist, riparian, low forests. Some life history traits, such as diurnal activity and the lack of tree-hole use, are distinctive compared to more northern populations.  相似文献

We studied the reproductive ecology of Clintonia borealis, a clonal understory species, in eastern Ontario. Flowers are protogynous and require insect pollination for outcrossing and maximum seed set. Most pollination is done by nectar-gathering bumble bees. We recorded pollen deposition and flowering patterns in 1983. Flowering lasted for about 10 days in mid-June. Stigmatic pollen loads were lowest at the onset of blooming, reflecting the scarcity of male-phase flowers. Pollination increased significantly by peak bloom and subsequently remained high. However, since bees fly mostly between neighboring stems, much of the pollen transferred may have been geitonogamous. In 1983, seed set per flower was not increased by supplementing pollination or by reducing the number of fruits competing for resources per stem. Rather, flowers set more seeds (14%) only if both treatments were performed simultaneously. Seed size was increased by 25% when competitive fruits were removed and by 5% more when pollen was added under the removal treatment. Further work in 1984 showed that pollination effects may be related to changes in pollen source. Selfed flowers set fewer and smaller seeds than outcrossed ones when screened from insects and hand pollinated. Outcrossing distance had little effect on seed set or seed size. Thus, in C. borealis seed number and seed size are limited by a balance between maternal resource availability and the amount of outcrossing provided by pollinators.  相似文献

The purpose of this study was to measure the effect of enhanced venous return on atrial natriuretic factor (ANF) secretion during exercise and upright posture and the consequences on renin angiotensin aldosterone system (RAAS) activity. Six healthy male subjects were submitted to four different procedures. All procedures were performed in the same position, i.e. riding on a support with legs hanging. Two procedures were performed at rest: the subjects were studied after a 25-min rest in this position, with and without the lower limb fitted with an anti-G suit inflated to 60 mmHg. Two procedures were carried out with physical exercise; arm-cranking was performed in the same position with and without the anti-G suit inflated to 60 mmHg. Venous blood was collected before and after each procedure in order to measure plasma ANF, plasma aldosterone concentration (PAC), plasma renin activity (PRA), corticotrophin (ACTH) and catecholamine level. The data mean +/- SEM showed that the ANF plasma level decreased significantly (p less than 0.05) from 32.5 +/- 4 to 28 +/- 6 pg.ml-1 after a 20-min rest in the upright posture, whereas this effect was absolished with anti-G suit inflation. Physical exercise with and without the anti-G suit increased the ANF level above control values (60 +/- 13.6 pg.ml-1 and 53 +/- 13 pg.ml-1): anti-G suit inflation had no significant effect. To evaluate to what extent opioid secretion in exercise induces the release of atrial natriuretic factor (ANF), six healthy male volunteers who were trained subjects, were submitted to two maximal exercise tests with and without (control) opioid receptor blockade by Naltrexone. Blood samples were drawn before (rest) and after exercise (post-exercise) in order to measure human ANF (alpha h ANF), beta-endorphin, plasma aldosterone concentration (PAC) plasma renin activity (PRA) and adreno-cortico trophic hormone (ATCH) by radio-immunological methods. Expired gas was collected during exercise to measure oxygen consumption. On average, the same maximal oxygen consumption (VO2max) during exercise was reached by all subjects with and without treatment. Plasma ANF level at rest slightly decreased after administration of Naltrexone; the response to physical exercise was significantly reduced by Naltrexone. There was no statistical difference between plasma levels of beta-endorphin, PRA and ACTH at rest nor in the post-exercise situation under the influence of Naltrexone. The PAC increased significantly at rest after Naltrexone administration but there was no statistical difference between both values after exercise. These data demonstrate that: (1) ANF secretion during exercise is influenced by the level of beta-endorphin in the plasma; (2) the possible inhibitory role of ANF on aldosterone secretion during exercise is probably over-ruled by the increase in plasma ACTH and PRA.  相似文献

Summary The flowers of the alpine snow buttercup Ranunculus adoneus track the sun's movement from early morning until mid-afternoon. Individual blooms last up to a week: younger female stage flowers show greater solar tracking fidelity than older hermaphrodite or dehisced flowers. Flowers aligned parallel to the sun's rays reach mean internal temperatures several degrees Celsius above ambient air temperature. As a flower's angle of deviation from the sun increases beyond 45 degrees, internal flower temperature is significantly reduced. Fly pollinators are seen disproportionately often on flowers aligned with the sun; this is due, in part, to their greater residence time on tracking flowers. Fly visitation is important to fecundity. When flies were excluded from flowers, some selfed seeds were matured, but total seed production was much less than in either handoutcrossed or open-pollinated flowers. Flowers that were tethered at random angles to prevent solar tracking set fewer, smaller seeds than unmanipulated control flowers. Reductions in seed weight were statistically consistent at different times in the season, but tethering influenced seed number per flower most strongly in the early season. Effects of tethering on maternal reproduction could be due to either pollinator diserimination, post-pollination developmental processes, or both. In a second series of experiments using fluorescent dye particles as pollen analogs, tethered donor flowers dispersed dye to as many recipients as paired control flowers, suggesting that solar tracking fidelity may have little effect upon this component of male reproduction.  相似文献
