Rapamycin, an inhibitor of the mechanistic target of rapamycin (mTOR), robustly extends the lifespan of model organisms including mice. We recently found that chronic treatment with rapamycin not only inhibits mTOR complex 1 (mTORC1), the canonical target of rapamycin, but also inhibits mTOR complex 2 (mTORC2) in vivo. While genetic evidence strongly suggests that inhibition of mTORC1 is sufficient to promote longevity, the impact of mTORC2 inhibition on mammalian longevity has not been assessed. RICTOR is a protein component of mTORC2 that is essential for its activity. We examined three different mouse models of Rictor loss: mice heterozygous for Rictor, mice lacking hepatic Rictor, and mice in which Rictor was inducibly deleted throughout the body in adult animals. Surprisingly, we find that depletion of RICTOR significantly decreases male, but not female, lifespan. While the mechanism by which RICTOR loss impairs male survival remains obscure, we find that the effect of RICTOR depletion on lifespan is independent of the role of hepatic mTORC2 in promoting glucose tolerance. Our results suggest that inhibition of mTORC2 signaling is detrimental to males, which may explain in part why interventions that decrease mTOR signaling show greater efficacy in females.  相似文献

Background  

The complexity of mitochondrial complex I (CI; NADH:ubiquinone oxidoreductase) has increased considerably relative to the homologous complex in bacteria. Comparative analyses of CI composition in animals, fungi and land plants/green algae suggest that novel components of mitochondrial CI include a set of 18 proteins common to all eukaryotes and a variable number of lineage-specific subunits. In plants and green algae, several purportedly plant-specific proteins homologous to γ-type carbonic anhydrases (γCA) have been identified as components of CI. However, relatively little is known about CI composition in the unicellular protists, the characterizations of which are essential to our understanding of CI evolution.  相似文献

Conquest of indigenous peoples in North America is understood primarily through ethnohistorical documents, archaeological evidence, and osteological analyses. However, in the Central Andes, the colonial enterprise and its effects are understood only from postcontact historical and ethnohistorical sources. Few archaeological and bioarchaeological studies have investigated Spanish Conquest and colonialism in the Andean region [for exceptions see Klaus and Tam: Am J Phys Anthropol 138 (2009) 356–368; Wernke, in press ; and Quilter, in press ]. Here we describe bioarchaeological evidence of violence from the cemeteries of Huaquerones and 57AS03 within the archaeological zone of Puruchuco‐Huaquerones, Peru (circa A.D. 1470–1540). A total of 258 individuals greater than 15 years of age were analyzed for evidence of traumatic injuries. Individuals were examined macroscopically and evidence of traumatic injuries was analyzed according to the skeletal element involved, the location of the injury on the skeletal element, and any additional complications of the injury. This study examines and compares the evidence of perimortem injuries on skeletonized individuals from the two cemeteries and focuses specifically on the interpretation of weapon‐related perimortem injuries. Evidence of perimortem trauma is present in both cemeteries (18.6%, 48/258); however, the frequency of injuries in 57AS03 is greater than that in Huaquerones (25.0% vs. 13.0%). Several injuries from 57AS03 are consistent with documented cases of injuries from firearms and 16th Century European weapons. We believe that the nature and high frequency of perimortem trauma at 57AS03 provide evidence of the violence that occurred with Spanish Conquest of the Inca Empire. Abstract

In April 1976 President Ford signed Public Law 94–265, the Fishery Conservation and Management Act of 1976. As this legislation comes of age, a far‐reaching and new approach to the management of our nation's fishery resources will be set into motion. Some of the most significant provisions of the Act are the restriction on the amount of foreign fishing, the charging of fees for foreign fishing privileges, and an enormous expansion of the area over which the United States exercises fisheries management control. This article briefly explains why the Act came into existence and what its key provisions are. It outlines the proposed foreign fee system for FY1977 and discusses the important economic impacts upon the United States from foreign fishing fees and restricted amounts of foreign fishing. It then examines possible policy questions and the important economic information necessary to assess them.  相似文献

Aim In the Indo‐Pacific, the mass of islands of the Indonesian archipelago constitute a major biogeographical barrier (the Indo‐Pacific Barrier, IPB) separating the Pacific and Indian oceans. Evidence for other, more localized barriers include high rates of endemism at the Marquesas and other isolated peripheral islands in the Pacific. Here we use mitochondrial‐sequence comparisons to evaluate the efficacy of biogeographical barriers on populations of the snappers Lutjanus kasmira and Lutjanus fulvus across their natural ranges. Location Pacific and Indian oceans. Methods Mitochondrial cytochrome b sequence data were obtained from 370 individuals of L. kasmira and 203 individuals of L. fulvus collected from across each species’ range. Allele frequency data for two nuclear introns were collected from L. kasmira. Phylogenetic and population‐level analyses were used to determine patterns of population structure in these species and to identify barriers to dispersal. Results Lutjanus kasmira lacks genetic structure across the IPB and throughout 12,000 km of its central Indo‐Pacific range. In contrast, L. fulvus demonstrates high levels of population structure at all geographical scales. In both species, highly significant population structure results primarily from the phylogenetic distinctiveness of their Marquesas Islands populations (L. kasmira, d = 0.50–0.53%; L. fulvus, d = 0.87–1.50%). Coalescence analyses of the L. kasmira data indicate that populations at opposite ends of its range (western Indian Ocean and the Marquesas) are the oldest. Coalescence analyses for L. fulvus are less robust but also indicate colonization from the Indian to the Pacific Ocean. Main conclusions The IPB does not act as a biogeographical barrier to L. kasmira, and, in L. fulvus, its effects are no stronger than isolating mechanisms elsewhere. Both species demonstrate a strong genetic break at the Marquesas. Population divergence and high endemism in that archipelago may be a product of geographical isolation enhanced by oceanographic currents that limit gene flow to and from those islands, and adaptation to unusual ecological conditions. Lutjanus kasmira shows evidence of Pleistocene population expansion throughout the Indo‐central Pacific that originated in the western Indian Ocean rather than the Marquesas, further demonstrating a strong barrier at the latter location.  相似文献
