Polyploidy is often thought to be associated with asexual reproduction, self-compatibility and self-pollination. Polyploids are also often regarded as being less sensitive than diploids to the negative effects of inbreeding. The reproductive biology of two lilioid species, the diploid Anthericum ramosum and the tetraploid A. liliago , was compared in a series of greenhouse and field experiments. Pollination experiments were carried out to study agamospermy (seed set without fertilization), self-compatibility and early inbreeding depression for both species. Tetraploid A. liliago showed no greater ability to self-pollinate than diploid A. ramosum and neither of the species showed agamospermy. Both species were self-compatible, but levels of fruit and seed set were lower after self-fertilization than after cross-fertilization in A. ramosum ( P <0.001). No reduction in fecundity after self-fertilization was observed in A. liliago . Fruit set in natural populations of A. ramosum increased significantly ( P <0.001) after emasculation (which excluded the possibility of autodeposition of pollen within flowers). In contrast, emasculation of flowers reduced the fruit set within natural populations of A. liliago ( P <0.05). The reproductive biology of diploid A. ramosum and tetraploid A. liliago is discussed in the context of polyploidization and the reduced fruit and seed set after self-fertilization in A. ramosum is discussed in terms of early inbreeding depression. 相似文献

Two sets of experiments were done to quantify the effects of chronic copper exposure on natural peri‐ phyton in a nonpolluted calcareous river. The results of short‐term (up to 6 h exposure) experiments corroborated the significance of pH on copper toxicity. Copper toxicity increased when pH was reduced from 8.6 to 7.7, and this was related to the effect of pH on copper speciation (free copper concentration increased from 0.2% to 2.3% of total copper). Longer term experiments demonstrated that periphyton communities exposed to copper under pH variation (8.2–8.6) were already affected at 10 μg·L ? 1 (20–80 ng·L ? 1 Cu2 + ) after 12 days of exposure. Copper exposure caused stronger effects on structural (algal biomass and community structure) than on functional (photosynthetic efficiency) parameters of peri‐ phyton. Changes in community composition included the enhancement of some taxa (Gomphonema gracile), the inhibition of others (Fragilaria capucina and Phormidium sp.), and the appearance of filament malformations (Mougeotia sp.). The results of our study demonstrated that several weeks of exposure to copper (10–20 μg·L ? 1) were sufficient to cause chronic changes in the periphyton of oligotrophic calcareous rivers. This degree of copper pollution can be commonly found in the Mediterranean region as a result of agricultural practices and farming activities. 相似文献

The matrilins form a family of non-collagenous adaptor proteins in the extracellular matrix. The extracellular ligand interactions of matrilins have been studied in some detail, while the potential interplay between matrilins and cells has been largely neglected. Except for matrilin-4, all matrilins mediate cell attachment, but only for matrilin-1 and -3 the binding is clearly dose dependent and seen already at moderate coating concentrations. Even so, much higher concentrations of matrilin-1 or -3 than of fibronectin are required for cell attachment to reach plateau values. Integrins contribute to the matrilin-mediated cell attachment, but the binding does not lead to formation of focal contacts and reorganisation of the actin cytoskeleton. Cells deficient in beta1 integrins are able to adhere, although weaker, and matrilins do not bind the soluble integrin alpha1beta1 and alpha2beta1 ectodomains. Cell surface proteoglycans may promote the attachment, as cells deficient in glycosaminoglycan biosynthesis adhere less well to matrilin-3. Even so, exogenous glycosaminoglycans are not able to compete for the attachment of HaCaT cells to matrilins. 相似文献

Chlorpromazine, dextroamphetamine and methylphenidate were significantly superior to placebo in producing overall improvement in the behaviour of hyperactive children. Chlorpromazine was effective for the majority of the children, but reduced only hyperactivity, having no demonstrable effect on distractibility, aggressivity or excitability. Both stimulants produced more goal-oriented behaviour and reduced distractibility. Methylphenidate was the most effective of the drugs in prpducing exceptional improvement. All three active drugs had to be discontinued in a few of the children because of side effects. Not all hyperactive children were benefited by the drugs.No background variables (with the exception of mother-child relationship) were found in the present studies to predict favourable response to the drugs.Methylphenidate became our drug of choice for this group of hyperactive children. 相似文献

Invasive plant species such as Ludwigia hexapetala might have a competitive advantage if they produce allelopathically active compounds against primary producers. Both phytoplankton and plant community structure may be affected due to different, species‐specific sensitivity to allelochemicals. Moreover, such allelopathic interactions could vary over the year depending on (i) the plant's phenological stage and (ii) the abilities of the native macrophytes to suppress—or the non‐native macrophytes to stimulate—the non‐native macrophyte population. We tested the allelopathic effects of aqueous leaf extracts of L. hexapetala on the photosynthetic activity of three target phytoplankton strains (Scenedesmus communis, a toxic Microcystis aeruginosa strain and a non‐toxic Microcystis aeruginosa strain) over three seasons of development (spring, summer and autumn). We also tested seasonal allelopathic effects of aqueous leaf extracts of both L. hexapetala (i.e. the non‐native invasive species) and the native Mentha aquatica on L. hexapetala seed germination. Finally, we identified three main secondary compounds present in the aqueous leaf extracts of L. hexapetala and we tested each individual compound on the phytoplankton's photosynthetic activity and on L. hexapetala seed germination. We observed marked seasonal and species‐specific patterns of L. hexapetala allelopathy on phytoplankton. The photosynthetic activities of S. communis and the toxic M. aeruginosa strain were stimulated by L. hexapetala aqueous leaf extracts in autumn and spring, respectively, whereas the non‐toxic M. aeruginosa strain was strongly inhibited in these two seasons. In summer, photosynthesis of all phytoplankton strains was inhibited. The germination rate of L. hexapetala seeds was stimulated by both L. hexapetala and M. aquatica aqueous leaf extracts, especially in summer, concomitant with the strong negative effects observed on the three phytoplankton strains. Three flavonoid glycosides (myricitrin, prunin and quercitrin) were identified as the main secondary compounds present in the L. hexapetala aqueous leaf extracts. The photosynthetic activity of S. communis was slightly stimulated by the three compounds. The photosynthetic activity of the toxic M. aeruginosa strain was stimulated by myricitrin and quercitrin, whereas that of the non‐toxic M. aeruginosa strain was inhibited by prunin. Finally, the germination rate and the germination velocity of L. hexapetala seeds were stimulated by myricitrin and prunin. These findings suggest that L. hexapetala could favour the photosynthetic activity of toxic cyanobacteria in spring and reduce their photosynthetic activity in summer, potentially leading to drastic changes in the phytoplankton communities and therewith ecological functioning of invaded ponds. Collagen VI, a collagen with uncharacteristically large N- and C-terminal non-collagenous regions, forms a distinct microfibrillar network in most connective tissues. It was long considered to consist of three genetically distinct α chains (α1, α2, and α3). Intracellularly, heterotrimeric molecules associate to form dimers and tetramers, which are then secreted and assembled to microfibrils. The identification of three novel long collagen VI α chains, α4, α5, and α6, led to the question if and how these may substitute for the long α3 chain in collagen VI assembly. Here, we studied structural features of the novel long chains and analyzed the assembly of these into tetramers and microfibrils. N- and C-terminal globular regions of collagen VI were recombinantly expressed and studied by small angle x-ray scattering (SAXS). Ab initio models of the N-terminal globular regions of the α4, α5, and α6 chains showed a C-shaped structure similar to that found for the α3 chain. Single particle EM nanostructure of the N-terminal globular region of the α4 chain confirmed the C-shaped structure revealed by SAXS. Immuno-EM of collagen VI extracted from tissue revealed that like the α3 chain the novel long chains assemble to homotetramers that are incorporated into mixed microfibrils. Moreover, SAXS models of the C-terminal globular regions of the α1, α2, α4, and α6 chains were generated. Interestingly, the α1, α2, and α4 C-terminal globular regions dimerize. To assess whether cells react differently towards a population of several laminin isoforms, as found in vivo, vs. a single isoform, we have compared the biological activity of kidney laminins to that of pure laminin 1. The kidney laminin preparation contained laminin 1 and further isoforms. Both substrates induced adhesion of a large spectrum of cell types, with kidney laminins being the most active. Unfolding of the coil-coiled conformation of the kidney isoforms negatively affected cell adhesion-promoting activity, which indicated that conformation-dependent cell binding is a characteristic feature of many or all laminins. Cellular interactions with kidney laminins were mediated by alpha3beta1 and alpha6beta1 integrins, with the contribution of alpha3beta1 being apparently lower than that of alpha6beta1 integrins. Immunofluorescence staining of vinculin and integrin subunits decorated focal adhesions on kidney laminins which differed in morphology from those formed on laminin 1 alone, in spite of the presence of the latter in the kidney preparation. These observations collectively indicate that tissue specific but often overlapping expression of laminin isoforms might modulate cell behavior by the activation of distinct sets of integrins and by the induction of distinct molecular assemblies within the cell adhesion signaling complexes. 相似文献

BackgroundThe pork tapeworm (Taenia solium) is a parasitic helminth that imposes a major health and economic burden on poor rural populations around the world. As recognized by the World Health Organization, a key barrier for achieving control of T. solium is the lack of an accurate and validated simulation model with which to study transmission and evaluate available control and elimination strategies. CystiAgent is a spatially-explicit agent based model for T. solium that is unique among T. solium models in its ability to represent key spatial and environmental features of transmission and simulate spatially targeted interventions, such as ring strategy.Methods/Principal findingsWe validated CystiAgent against results from the Ring Strategy Trial (RST)–a large cluster-randomized trial conducted in northern Peru that evaluated six unique interventions for T. solium control in 23 villages. For the validation, each intervention strategy was replicated in CystiAgent, and the simulated prevalences of human taeniasis, porcine cysticercosis, and porcine seroincidence were compared against prevalence estimates from the trial. Results showed that CystiAgent produced declines in transmission in response to each of the six intervention strategies, but overestimated the effect of interventions in the majority of villages; simulated prevalences for human taenasis and porcine cysticercosis at the end of the trial were a median of 0.53 and 5.0 percentages points less than prevalence observed at the end of the trial, respectively.Conclusions/SignificanceThe validation of CystiAgent represented an important step towards developing an accurate and reliable T. solium transmission model that can be deployed to fill critical gaps in our understanding of T. solium transmission and control. To improve model accuracy, future versions would benefit from improved data on pig immunity and resistance, field effectiveness of anti-helminthic treatment, and factors driving spatial clustering of T. solium infections including dispersion and contact with T. solium eggs in the environment. 相似文献
