The aim of this work was to evaluate, by comet assay, the possible inducing of DNA lesions in peripheral blood mononuclear cells of rats subjected to acute or chronic food deprivation. Wistar male rats were subjected to 72 h of partial (50%), or total acute food deprivation, and then allowed to recover for different time periods (24, 48 and 72 h). In other experiments, comet scores were determined in peripheral blood mononuclear cells of rats subjected to chronic food deprivation (25% and 50%) for 50 days. Blood aliquots were obtained before, during and after food deprivation. Comet assay was carried out, the comet units photographed and scored (class 0 up to 3). Acute and chronic food-deprived rats presented peripheral blood mononuclear cells with DNA lesions (comet classes 1, 2 and 3) and a significant increase (p<0.05) in the number of comet units compared with its basal level. The increase was proportional to acute food deprivation time, but after being taken off, it progressively returned to basal level after 48 h (partial group) or 72 h (total group). Chronic food-deprived rats presented a progressive increase of comet score up to 5 days, and a decrease thereafter to reach a basal level. Possible mechanisms of DNA lesions are discussed.  相似文献

SMOC-2 is a recently discovered member of the BM-40/SPARC/osteonectin family of extracellular multidomain proteins of so far unknown function. While we have shown earlier that the homologous protein SMOC-1 is associated with basement membranes, in this study we demonstrate that, in the mouse, SMOC-2 could be detected in a large number of non-basement membrane localizations, often showing a diffuse tissue distribution. A more distinct expression pattern was seen in skin where SMOC-2 is mainly present in the basal layers of the epidermis. Functionally, recombinant SMOC-2 stimulated attachment of primary epidermal cells as well as several epidermal-derived cell lines but had no effect on the attachment of non-epidermal cells. Inhibition experiments using blocking antibodies against individual integrin subunits allowed the identification of αvβ6 and αvβ1 integrins as important cellular receptors for SMOC-2. Cell attachment as well as the formation of focal adhesions could be attributed to the extracellular calcium-binding domain. The calcium-binding domain also stimulated migration, but not proliferation of keratinocyte-like HaCaT cells. We conclude that SMOC-2, like other members of the BM40/SPARC family, acts as a regulator of cell–matrix interactions.  相似文献

PACSINs are intracellular adapter proteins involved in vesicle transport, membrane dynamics and actin reorganisation. In this study, we report a novel role for PACSIN proteins as components of the centrosome involved in microtubule dynamics. Glutathione S-transferase (GST)-tagged PACSIN proteins interacted with protein complexes containing α- and γ-tubulin in brain homogenate. Analysis of cell lysates showed that all three endogenous PACSINs co-immunoprecipitated dynamin, α-tubulin and γ-tubulin. Furthermore, PACSINs bound only to unpolymerised tubulin, not to microtubules purified from brain. In agreement, the cellular localisation of endogenous PACSIN 2 was not affected by the microtubule depolymerising reagent nocodazole. By light microscopy, endogenous PACSIN 2 localised next to γ-tubulin at purified centrosomes from NIH 3T3 cells. Finally, reduction of PACSIN 2 protein levels with small-interfering RNA (siRNA) resulted in impaired microtubule nucleation from centrosomes, whereas microtubule centrosome splitting was not affected, suggesting a role for PACSIN 2 in the regulation of tubulin polymerisation. These findings suggest a novel function for PACSIN proteins in dynamic microtubuli nucleation.  相似文献

Ranunculus peltatus Schrank is an aquatic macrophyte spreading in northeastern France. As with other Ranunculus species, its growth demonstrates considerable plasticity. A preliminary understanding of its development and precise adaptive strategy is possible through functional ecology. The present study took morphological traits into account in order to analyse R. peltatus' development and morphological plasticity in relation to environmental parameters. During the five-month growing season, field measurements of ten different morphological traits were performed monthly at twelve sampling sites presenting various physical and chemical characteristics. Three overall growth stages occurred between April and August 2000: a stage of elongation of the main stem, a stage of branching and flowering and a stage of decline associated with vegetative dispersion. The sampling sites were defined according to environmental parameters and divided into three groups corresponding to three morphologically different R. peltatus populations. Plants in upstream, nutrient-poor, undisturbed sites were small and achieved little sexual reproduction. Plants in nutrient-rich, undisturbed sites had long shoots and were branched. Plants in weakly shaded, disturbed sites were small but produced many flowers. Correlations between morphological traits and environmental parameters showed significant relationships between chemical parameters and some characteristic vegetative growth traits. Physical environmental parameters were found to be less well correlated with morphological traits than the chemical parameters. The phenology and morphological plasticity of R. peltatus confer competitive advantages that could explain its ability to spread in some circumstances. Finally, the previous classification of R. peltatus as a Grime CSR species is discussed in relation to its plasticity. According to its morphological characteristics, R. peltatus tended indeed to adopt a C-strategy in nutrient-rich undisturbed sites, a S-strategy in nutrient-poor undisturbed sites and a R-strategy in disturbed sites.  相似文献

Human bone sialoprotein (BSP) comprises 15% of the total noncollagenous proteins in bone and is thought to be involved in bone mineralization and remodeling. Recent data suggest a role for BSP in breast cancer and the development of bone metastases. We have produced full-length recombinant BSP in a human cell line and purified the protein from human bone retaining the native structure with proper folding and post-translational modifications. Mass spectrometry of bone-derived BSP revealed an average mass of 49 kDa and for recombinant BSP 57 kDa. The post-translational modifications contribute 30-40%. Carbohydrate analysis revealed 10 different complex-type N-glycans on both proteins and eight different O-glycans on recombinant BSP, four of those were found on bone-derived BSP. We could identify eight threonines modified by O-glycans, leaving the C terminus of the protein free of glycans. The recombinant protein showed similar secondary structures as bone-derived BSP. BSP was visualized in electron microscopy as a globule linked to a thread-like structure. The affinity for hydroxyapatite was higher for bone-derived BSP than for recombinant BSP. Cell adhesion assays showed that the binding of BSP to cells can be reversibly diminished by denaturation.  相似文献

In this study, we contrast brain morphology from hatchery and wild reared stocks to examine the hypothesis that in salmonid fishes, captive rearing produces changes in brain development. Using rainbow trout, Oncorhynchus mykiss, as a model, we measured eight regions of the salmonid brain to examine differences between wild and hatchery reared fish. We find using multiple analysis of covariance (MANCOVA), analysis of covariance (ANCOVA) and discriminant function analysis (DFA) that the brains of hatchery reared fish are relatively smaller in several critical measures than their wild counterparts. Our work may suggest a mechanistic basis for the observed vulnerability of hatchery fish to predation and their general low survival upon release into the wild. Our results are the first to highlight the effects of hatchery rearing on changes in brain development inbreak fishes.  相似文献

A methodology has been developed and used to investigate a direct link between juvenile whitebait (Hyperlophus vittatus), the key prey species sampled from the stomachs of Little Penguins (Eudyptula minor), and the specific nursery area of that prey. A unique application of existing methodology initially involved the measurement of the stable isotopes of oxygen (δ18O:δ16O) and carbon (δ13C:δ12C) in sagittal otolith carbonate, using standard mass spectrometric techniques. The results indicated that the inshore distribution of juvenile (0+ year old) whitebait between Cockburn Sound and Koombana Bay, Western Australia consisted of a number of separate assemblages. Measured differences in stable oxygen isotope ratios were attributed to variations in freshwater input to the embayments that provided the whitebait habitats. In contrast, the measured stable carbon isotope ratios probably resulted from the different isotopic compositions of the food webs in the various habitats. Secondly, a comparison of the average value of carbon and oxygen isotope signatures of pooled otoliths from samples of whitebait from a number of different nearshore coastal sites (assemblages), with that of whitebait obtained from the stomachs of penguins at their main breeding site (Penguin Island) indicated that the values from the penguins resemble most closely those of the average otolith values obtained from whitebait from only one site (Becher Point). Assuming that the whitebait sampled were representative of the whitebait in the nearshore habitats and the diets of the penguins, then these results imply that at the time of sampling the penguins were feeding on whitebait from only one site.  相似文献

We measured net ecosystem CO2 exchange (NEE) in Panamá over C4 pasture plots that varied in grazing intensity. After adjusting for variation in light, there were noticeable effects of grazing‐related variables on CO2 exchange that were largely dependent on the developmental stage of the plant canopy. Above‐ground productivity was positively related to grazing intensity (r2=0.30). Two experimentally grazed fields had significantly lower standing crop biomass but no significant difference in CO2 uptake (24.2 μmol/m2/s) compared with two ungrazed fields (20.3 μmol/m2/s). Grazed fields had significantly lower ecosystem respiration rates (10.3 μmol/m2/s) than did ungrazed fields (17.6 μmol/m2/s). These results suggest that, although these pastures were possible sources of CO2 during the time intervals sampled, the size of the sources tended to be dampened by cattle grazing through reductions in ecosystem respiration. Thus, it appears that disturbance caused by cattle grazing will not always result in an increase in CO2 release from tropical pastures to the atmosphere.  相似文献
