Molecular characterization of an aberrant allele for the Gy3 glycinin gene: a chromosomal rearrangement.

The soybean variety Forrest contains an aberrant allele for the Gy3 glycinin gene. The aberrant allele is designated gy3 because mRNA for the G3 glycinin subunit is reduced to below detectable amounts in the seed. Molecular and genetic characterization of gy3 show it to be associated with a chromosomal rearrangement that causes the 5' halves and 3' halves of the gene to become separated from one another in the genome. An inversion is the simplest structural model that accounts for the genetic and molecular features of the chromosomal rearrangement involving gy3, although more complex models that involve reciprocal translocations are also consistent with the data.

In this review, we discuss the biological action and biochemical function of the v-erbA oncogene product, and the role of c-erbA proto-oncogene products as thyroid hormone receptors, as related to the molecular structure and function of the nuclear hormone receptors at large.     

Involvement of the region 13q14 in a patient with adamantinoma of the long bones

Summary A cytogenetic analysis of a lung metastasis of an adamantinoma of the tibia, a rare tumor of the long bones, revealed a karyotype 52, XY, t(7;13)(q32;q14), +7, +12, +13, +19, +der(7) t(7;13), +der(13) t(7;13). The t(7;13) was found to be constitutional in the patient and was also present in peripheral blood lymphocytes of his unaffected father. However, both subjects displayed normal levels of esterase D enzyme activity.     

In vitro phosphorylation of angiotensin analogs by tyrosyl protein kinases

Peptide analogs of angiotensin were phosphorylated in vitro by the src gene product, pp60src, of Rous sarcoma virus. The Km for the phosphorylation reaction varied from 1 to 5 mM and the Vmax varied from 2 to 10 nmol/min/mg. Tyrosine was the only residue phosphorylated in all analogs that were examined. The peptides were phosphorylated by tyrosyl protein kinases associated with several avian sarcoma viruses and by the epidermal growth factor-receptor kinase of A431 cells. Peptide substrate also was used to investigate the effectiveness of three different phosphatase inhibitors. Assay of tyrosyl kinase activities in whole cell lysates indicated that both p-nitrophenyl phosphate and sodium vanadate were potent inhibitors of phosphotyrosine phosphatases.     

The effects of starvation and temperature acclimation on pentose phosphate pathway dehydrogenases in brook trout liver

Temperature and starvation were found to be factors which affected the PPP dehydrogenase activities in brook trout liver. Fish acclimated at 5 °C possessed greater levels of G6PD, H6PD, and 6PGD activity than those fish maintained at 10 or 15 °C. This phenomenon was probably associated with increased lipogenesis during cold acclimation.During starvation hepatic G6PD and 6PGD activities decreased, whereas H6PD activity increased slightly. Upon refeeding, the G6PD level gradually increased, but the “overshoot” in enzyme activity reported in mammalian studies was not observed.When both cold acclimation and starvation were studied simultaneously, regulation by temperature was initially the dominant control factor. After 6 wk at 5 °C, there was no difference in specific activities between starved and fed fish. However, fish maintained at 5 °C for longer than 2 mo did show the normal response to starvation and refeeding. Therefore, regulation of the PPP by temperature appears to be a transitory phenomenon and may be associated with temporary metabolic reorganization in the fish.     

Development of defined and minimal media for the growth of Bacillus stearothermophilus.

Defined media, both solid and liquid, that support good growth of Bacillus stearothermophilus 1503 have been developed. Data are presented which indicate that manganese is required at relatively high concentrations for growth in a defined liquid medium. Phosphate concentrations higher than 5 times 10(-3) M have been shown to inhibit colony formation on solid media. Maximum viable counts of approximately 10(9) colony-forming units per ml were obtained in both the defined and minimal liquid media. Glucose, fructose, sucrose, glycerol, and starch support the growth of this obligate thermophile in the defined media, whereas citrate, alpha-ketoglutarate, succinate, fumarate, malate, acetate, and lactate do not. The described media have been utilized to isolate several amino acid-requiring mutants of B. stearothermophilus.     

Growth of Pseudomonas C on C1 compounds: a correction.

On reexamination Pseudomonas C was found to be incapable of growth on formaldehyde or formate as a sole carbon source and to contain a hexose phosphate synthase activity when grown on methanol.
