Sex is genetically determined in Histoplasma capsulatum, governed by a sex-specific region in the genome called the mating-type locus (MAT1). We investigate the distribution of isolates of two H. capsulatum mating types in the clades circulating in Buenos Aires, Argentina. Forty-nine H. capsulatum isolates were obtained from the culture collection of the Mycology Center. The MAT1 locus was identified by PCR from the yeast suspension. The analysis of forty-eight isolates from clinical samples exhibited a ratio of 1.7 (MAT1-1:MAT1-2) and the only isolate from soil was MAT1-1. Forty-five H. capsulatum isolates belonged to the LAm B clade (H. capsulatum from Latin American group B clade) and showed a ratio of 1.8 (MAT1-1:MAT1-2). These results suggest an association between the mating types in isolates belonging to the LAm B clade. It remains to be defined whether a greater virulence should be attributed to the differences between the strains of the opposite mating type of the LAm B clade.
Messenger RNA encoded signals that are involved in programmed -1 ribosomal frameshifting (-1 PRF) are typically two-stemmed hairpin (H)-type pseudoknots (pks). We previously described an unusual three-stemmed pseudoknot from the severe acute respiratory syndrome (SARS) coronavirus (CoV) that stimulated -1 PRF. The conserved existence of a third stem–loop suggested an important hitherto unknown function. Here we present new information describing structure and function of the third stem of the SARS pseudoknot. We uncovered RNA dimerization through a palindromic sequence embedded in the SARS-CoV Stem 3. Further in vitro analysis revealed that SARS-CoV RNA dimers assemble through ‘kissing’ loop–loop interactions. We also show that loop–loop kissing complex formation becomes more efficient at physiological temperature and in the presence of magnesium. When the palindromic sequence was mutated, in vitro RNA dimerization was abolished, and frameshifting was reduced from 15 to 5.7%. Furthermore, the inability to dimerize caused by the silent codon change in Stem 3 of SARS-CoV changed the viral growth kinetics and affected the levels of genomic and subgenomic RNA in infected cells. These results suggest that the homodimeric RNA complex formed by the SARS pseudoknot occurs in the cellular environment and that loop–loop kissing interactions involving Stem 3 modulate -1 PRF and play a role in subgenomic and full-length RNA synthesis. 相似文献
Phytoplankton communities dominating Musgos and Papúa ponds with differing trophic states were sampled over 3 days enabling the detection of the physiological and population responses of microalgae to short-scale changes in biotic and abiotic factors, rather than frequently analyzed changes in community composition responses to long-scale environmental changes. We hypothesized that both environments undergoing diel changes would be dominated by phytoplankton with generalist strategies, while community structure would be mostly dictated by the trophic state of each water body. The phytoplankton biovolumes of both ponds were strongly dominated by euplanktonic nanoflagellated Chlorophyta, while phycocyanin-rich picocyanobacteria dominated the picophytoplankton. Parallel diel cycles of air and water temperatures were more pronounced on a sunny, warm day which prompted algal photosynthesis, revealed by strong increases in dissolved oxygen and pH. Nutrient and phytoplanktonic chlorophyll a confirmed the hypertrophic condition of Papúa pond. This accounted for the distinct community composition encountered in each pond, which remained stable throughout the study, as revealed by the SIMI index. The inverse relationship between the chl a/abundance ratio and the abundances of dominant species together with varying net growth rates (k′) showed algal reproduction, yet densities remained rather stable in both cases. In Musgos pond, fluctuations in k′ for small and median ciliates shadowed those of pico- and nanophytoplankton, respectively, strongly suggesting that they can control algal growth in these 2-level trophic chains. 相似文献
Salt stress is an increasing problem in agricultural soils in many parts of the world, and salt tolerant cropping systems are in great demand. We investigated the effect of co-inoculation of Galega officinalis with Rhizobium galegae and two plant growth promoting Pseudomonas species on plant growth, nodulation, and N content under salt stress.
Methods
The effect of inoculation with R. galegae sv. officinalis HAMBI 1141 alone and in combination with the root-colonizing Pseudomonas extremorientalis TSAU20 or Pseudomonas trivialis 3Re27 on the growth of G. officinalis exposed to salt stress (50 and 75 mM NaCl) was studied under gnotobiotic and greenhouse conditions.
Results
The growth of goat’s rue was reduced at 50 and 75 mM NaCl both in the gnotobiotic sand system and in low-fertilized potting soil in the greenhouse. Co-inoculation of unstressed and salt-stressed goat’s rue with R. galegae HAMBI 1141 and either P. extremorientalis TSAU20 or P. trivialis 3Re27 significantly improved root and shoot growth and increased nodulation of plant roots in both growth systems compared with plants inoculated with R. galegae alone. The nitrogen content of co-inoculated plant roots was significantly increased at 75 mM NaCl in potting soil. Co-inoculation of G. officinalis with either of the two plant growth promoting (PGPR) Pseudomonas strains also improved root tip-colonization by R. galegae cells.
Conclusions
The co-inoculation of goat’s rue with Rhizobium and PGPR Pseudomonas strains alleviated salt stress of plants grown in NaCl-amended gnotobiotic sand systems and in potting soil in the greenhouse. 相似文献
Ichnodiversity has been used as a proxy for environmental stress and stability in facies interpretations and to reconstruct evolutionary radiations and colonization histories in evolutionary palaeoecology. The three components of global diversity are exported from ecology and adapted for ichnology. Alpha ichnodiversity is used for palaeoenvironmental characterization, being assessed for individual facies. Beta ichnodiversity is commonly overlooked, although it may provide information about degree of similarity between ichnofaunas formed along environmental gradients. Gamma ichnodiversity may provide clues to detect ichnofossil provincialism. The concept of disparity may also prove to be significant in ichnological studies. Whereas ichnodiversity refers to ichnotaxonomic richness, ichnodisparity provides a measure of the variability of morphological plans in biogenic structures. Changes in global ichnodiversity do not necessarily parallel changes in ichnodisparity. For example, while the Cambrian explosion involved a dramatic increase in both, the Ordovician radiation essentially reflects an increase only in the former. Ichnodiversity and ichnodisparity should be used with caution because they are both affected by taphonomic processes. High diversity of superficial to shallow‐tier trace fossils may result from enhanced preservation due a poorly developed mixed layer, rather than a true reflection of ecosystem performance, as shown by underexploited infaunal ecospace after biotic crises (e.g. end‐Permian mass extinction). 相似文献
The stability of the erythrocyte membrane, which is essential for the maintenance of cell functions, occurs in a critical region of fluidity, which depends largely on its composition and the composition and characteristics of the medium. As the composition of the erythrocyte membrane is influenced by several blood variables, the stability of the erythrocyte membrane must have relations with them. The present study aimed to evaluate, by bivariate and multivariate statistical analyses, the correlations and causal relationships between hematologic and biochemical variables and the stability of the erythrocyte membrane against the chaotropic action of ethanol. The validity of this type of analysis depends on the homogeneity of the population and on the variability of the studied parameters, conditions that can be filled by patients who undergo bariatric surgery by the technique of Roux-en-Y gastric bypass since they will suffer feeding restrictions that have great impact on their blood composition. Pathway analysis revealed that an increase in hemoglobin leads to decreased stability of the cell, probably through a process mediated by an increase in mean corpuscular volume. Furthermore, an increase in the mean corpuscular hemoglobin (MCH) leads to an increase in erythrocyte membrane stability, probably because higher values of MCH are associated with smaller quantities of red blood cells and a larger contact area between the cell membrane and ethanol present in the medium. 相似文献
The important role of the CD8+ T-cell response on HIV control is well established. Moreover, the acute phase of infection represents a proper scenario to delineate the antiviral cellular functions that best correlate with control. Here, multiple functional aspects (specificity, ex vivo viral inhibitory activity [VIA] and polyfunctionality) of the HIV-specific CD8+ T-cell subset arising early after infection, and their association with disease progression markers, were examined. Blood samples from 44 subjects recruited within 6 months from infection (primary HIV infection [PHI] group), 16 chronically infected subjects, 11 elite controllers (EC), and 10 healthy donors were obtained. Results indicated that, although Nef dominated the anti-HIV response during acute/early infection, a higher proportion of early anti-Gag T cells correlated with delayed progression. Polyfunctional HIV-specific CD8+ T cells were detected at early time points but did not associate with virus control. Conversely, higher CD4+ T-cell set points were observed in PHI subjects with higher HIV-specific CD8+ T-cell VIA at baseline. Importantly, VIA levels correlated with the magnitude of the anti-Gag cellular response. The advantage of Gag-specific cells may result from their enhanced ability to mediate lysis of infected cells (evidenced by a higher capacity to degranulate and to mediate VIA) and to simultaneously produce IFN-γ. Finally, Gag immunodominance was associated with elevated plasma levels of interleukin 2 (IL-2) and macrophage inflammatory protein 1β (MIP-1β). All together, this study underscores the importance of CD8+ T-cell specificity in the improved control of disease progression, which was related to the capacity of Gag-specific cells to mediate both lytic and nonlytic antiviral mechanisms at early time points postinfection. 相似文献
In 2004, the urban presence of Lutzomyia longipalpis was recorded
for the first time in Formosa province. In 2006, the first autochthonous case of
human urban visceral leishmaniasis (VL) was recorded in Misiones in the presence of
the vector, along with some canine VL cases. After this first case, the vector began
to spread primarily in northeast Argentina. Between 2008-2011, three human VL cases
were reported in Salta province, but the presence of Lu. longipalpis
was not recorded. Captures of Phlebotominae were made in Tartagal, Salta, in 2013,
and the presence of Lu. longipalpis was first recorded in northwest
Argentina at that time. Systematic sampling is recommended to observe the
distribution and dispersion patterns of Lu. longipalpis and consider

This cross-sectional retrospective study evaluated 440 leprosy patients; 57% (251/440) had leprosy reactions during and/or after multidrug therapy, 80.5% (202/251) of whom presented with multibacillary leprosy. At diagnosis, positive bacterial index (BI) [odds ratio (OR) = 6.39; 95% confidence interval (CI): 4.1-10.1)] or polymerase chain reaction (PCR) (OR = 9.15; 95% CI: 5.4-15.5) in skin smears, anti-phenolic glycolipid-1 (anti-PGL-1) ELISA (OR = 4.77; 95% CI: 2.9-7.9), leucocytosis (OR = 9.97; 95% CI: 3.9-25.7), thrombocytopenia (OR = 5.72; 95% CI: 2.3-14.0) and elevated lactate dehydrogenase (OR = 2.38; 95% CI: 1.4-4.0) were potential markers for the development of reactions during treatment. After treatment, positive BI (OR = 8.47; 95% CI: 4.7-15.3) and PCR (OR = 6.46; 95% CI: 3.4-12.3) in skin smears, anti-PGL-1 ELISA (OR = 2.25; 95% CI: 1.3-3.9), anaemia (OR = 2.36; 95% CI: 1.2-4.5), leucocytosis (OR = 4.14; 95% CI: 1.5-11.6) and thrombocytopenia (OR = 3.70; 95% CI: 1.3-2.2) were risk factors for the occurrence of reactions during the study period. The identification of groups with an increased risk for developing reactions will allow for the timely development of a treatment plan to prevent nerve damage and, therefore, the appearance of the disabling sequelae associated with the stigma of leprosy. 相似文献
