ABSTRACT. 1. The distribution of Dactylopius opuntiae (Cockerell) colonies on Opuntia ficus-indica (L.) Miller cladodes collected during a prolonged drought, was recorded for the proximal and distal portions of the cladodes, for the exposed and sheltered surfaces, the edges, and in relation to the proximity of the colonies to areoles at the base of the thorns.
2. Most of the colonies were found on the more sheltered parts of the cladodes and a disproportionately large number of the colonies on exposed surfaces had settled below the areoles.
3. Individuals in colonies on the exposed surfaces were all protected by a relatively hard, compacted wax cover and were thus less vulnerable to simulated rainfall than those on sheltered surfaces. Thorns provided some protection from simulated rainfall for individuals in colonies on exposed surfaces, but not for individuals on sheltered surfaces.
4. Natural rainfall greatly reduced the numbers of colonies, but did not significantly change the distribution of the colonies on the cactus cladodes.
5. Rainfall does not explain the observed distribution of D. opuntiae colonies on the cladodes, but does explain the poor performance of cochineal insects as biological control agents of prickly pear cacti in high rainfall areas.  相似文献

ABSTRACT. We have demonstrated previously that crystal violet induces a rapid, dose-related collapse of the inner mitochondrial membrane potential of Trypanosoma cruzi epimastigotes. In this work, we show that crystal violet-induced dissipation of the membrane potential was accompanied by an efflux of Ca2+ from the mitochondria. In addition, crystal violet inhibited the ATP-dependent, oligomycin-, and antimycin A-insensitive Ca2+ uptake by digitonin-permeabilired epimastigotes. Crystal violet also induced Ca2+ release from the mitochondria and endoplasmic reticulum of digitonin-permeabilized trypomastigotes. Furthermore, crystal violet inhibited Ca2+ uptake and the (Ca2+-Mg2+)ATPase of a highly enriched plasma membrane fraction of epimastigotes, thus indicating an inhibition of other calcium transport mechanisms of the cells. Disruption of Ca2+ homeostasis by crystal violet may be a key process leading to trypanosome cell injury by this drug.  相似文献

The effect of elevated Na+ concentration on Na+ permeability(PNa) and Na+ influx in the presence of two levels of externaldivalent cations was determined in Chara corallina and freshwater-culturedChara buckellii. When Na+ in the medium was increased from 1.0to 70 mol m–3, Na+ influx increased in both species ifCa2+ was low (0.1 mol m–3). If Ca2+ was increased to 7.0mol m–3 when Na+ was increased, Na+ influx remained atthe low control level in C. corallina, and showed only a temporaryincrease in C. buckellii. Mg2+ was a better substitute for Ca2+in C. buckellii than in C. corallina. Na+ permeability data suggest that when the external Ca2+ concentrationis low, PNa does not increase in the presence of elevated NaCl;the increase in Na+ influx appears to be due to the increasein external Na+ concentration alone. Ca2 + supplementation appearsto decrease PNa whereas supplemental Mg2+ has no effect. Na+ effluxes were computed from previously determined net fluxesand the influxes. It was found that for both species, fluxesin both directions were stimulated in response to all experimentaltreatments, but Na+ influx always exceeded efflux. This resultedin net Na+ accumulation in the vacuoles of both species. The results are discussed with reference to net flux and electrophysiologicaldata obtained previously under identical conditions, as wellas the comparative salinity tolerance of both species and theNa+/divalent cation ratio. Abstract.  The giant milkweed bug, Oncopeltus fasciatus , is an intermittent feeder, spending most of its time in compact groups away from the food, with feeding bouts lasting 30–60 min. They macerate a small area of the seed with their stylets, secreting large amounts of saliva (which suspends and emulsifies the seed tissue), and quickly suck the emulsion into the gut. Therefore, little digestive activity occurs in the seed tissue. Freshly ingested food in the first gut region (V1) consists of 50% water, 46% lipid, 5% protein, and 1% soluble carbohydrates. The junction of the midgut and hindgut lacks a lumen in the larvae stages, and a 3 μL large oil drop (> 85% triacylglycerides) accumulates in the third gut region (V3). Approximately 24 h after the imaginal moult, this large volume of oil and faeces is defecated, and thereafter an oily faeces is defecated several times daily. It is postulated that the delayed egestion in larvae protects them against suffocation due to self-soiling with oily faeces. Serine proteases are absent in the gut and, instead, a cystein proteinase, inhibited by E64, is present (gut pH 6.0). High lipase activity was found throughout the gut, but only limited amylase activities. Basal levels of all three enzymes occurred throughout the gut, but at lower levels in starved bugs. Salivary gland extracts contain a cysteine proteinase and a lipase, but no amylase.  相似文献

Adaptation of guppies (Poecilia reticulata) to contrasting upland and lowland habitats has been extensively studied with respect to behaviour, morphology and life history traits. Yet population history has not been studied at the whole‐genome level. Although single nucleotide polymorphisms (SNPs) are the most abundant form of variation in many genomes and consequently very informative for a genome‐wide picture of standing natural variation in populations, genome‐wide SNP data are rarely available for wild vertebrates. Here we use genetically mapped SNP markers to comprehensively survey genetic variation within and among naturally occurring guppy populations from a wide geographic range in Trinidad and Venezuela. Results from three different clustering methods, Neighbor‐net, principal component analysis (PCA) and Bayesian analysis show that the population substructure agrees with geographic separation and largely with previously hypothesized patterns of historical colonization. Within major drainages (Caroni, Oropouche and Northern), populations are genetically similar, but those in different geographic regions are highly divergent from one another, with some indications of ancient shared polymorphisms. Clear genomic signatures of a previous introduction experiment were seen, and we detected additional potential admixture events. Headwater populations were significantly less heterozygous than downstream populations. Pairwise FST values revealed marked differences in allele frequencies among populations from different regions, and also among populations within the same region. FST outlier methods indicated some regions of the genome as being under directional selection. Overall, this study demonstrates the power of a genome‐wide SNP data set to inform for studies on natural variation, adaptation and evolution of wild populations  相似文献

Recent Advances in Our Knowledge of the Myxozoa   总被引:20,自引:0,他引:20  
In the last few years two factors have helped to significantly advance our understanding of the Myxozoa. First, the phenomenal increase in fin fish aquaculture in the 1990s has lead to the increased importance of these parasites; in turn this has lead to intensified research efforts, which have increased knowledge of the development, diagnosis. and pathogenesis of myxozoans. The hallmark discovery in the 1980s that the life cycle of Myxobolus cerebralis requires development of an actinosporean stage in the oligochaete. Tubifex tubifex, led to the elucidation of the life cycles of several other myxozoans. Also, the life cycle and taxonomy of the enigmatic PKX myxozoan has been resolved: it is the alternate stage of the unusual myxozoan, Tetracapsula bryosalmonae, from bryozoans. The 18S rDNA gene of many species has been sequenced, and here we add 22 new sequences to the data set. Phylogenetic analyses using all these sequences indicate that: 1) the Myxozoa are closely related to Cnidaria (also supported by morphological data); 2) marine taxa at the genus level branch separately from genera that usually infect freshwater fishes; 3) taxa cluster more by development and tissue location than by spore morphology; 4) the tetracapsulids branched off early in myxozoan evolution, perhaps reflected by their having bryozoan, rather than annelid hosts; 5) the morphology of actinosporeans offers little information for determining their myxosporean counterparts (assuming that they exist); and 6) the marine actinosporeans from Australia appear to form a clade within the platysporinid myxosporeans. Ribosomal DNA sequences have also enabled development of diagnostic tests for myxozoans. PCR and in situ hybridisation tests based on rDNA sequences have been developed for Myxobolus cerebralis, Ceratomyxa shasta, Kudoa spp., and Tetracapsula bryosalmonae (PKX). Lectin-based and antibody tests have also been developed for certain myxozoans, such as PKX and C. shasta. We also review important diseases caused by myxozoans, which are emerging or re-emerging. Epizootics of whirling disease in wild rainbow trout (Oncorhynchus mykiss) have recently been reported throughout the Rocky Mountain states of the USA. With a dramatic increase in aquaculture of fishes using marine netpens, several marine myxozoans have been recognized or elevated in status as pathological agents. Kudoa thyrsites infections have caused severe post-harvest myoliquefaction in pen-reared Atlantic salmon (Salmo salar), and Ceratomyxa spp., Sphaerospora spp., and Myxidium leei cause disease in pen-reared sea bass (Dicentrarchus labrax) and sea bream species (family Sparidae) in Mediterranean countries.  相似文献

We examined latitudinal variation in adult and larval heat tolerance in Drosophila melanogaster from eastern Australia. Adults were assessed using static and ramping assays. Basal and hardened static heat knockdown time showed significant linear clines; heat tolerance increased towards the tropics, particularly for hardened flies, suggesting that tropical populations have a greater hardening response. A similar pattern was evident for ramping heat knockdown time at 0.06 °C min?1 increase. There was no cline for ramping heat knockdown temperature (CTmax) at 0.1 °C min?1 increase. Acute (static) heat knockdown temperature increased towards temperate latitudes, probably reflecting a greater capacity of temperate flies to withstand sudden temperature increases during summer in temperate Australia. Larval viability showed a quadratic association with latitude under heat stress. Thus, patterns of heat resistance depend on assay methods. Genetic correlations in thermotolerance across life stages and evolutionary potential for critical thermal limits should be the focus of future studies.  相似文献

1. Despite growing knowledge on the relationship between ecological variables and individual immune function, data on the spatial variability of immune defence in invertebrate natural populations are scarce. 2. Here, we use replicated populations of the butterfly Lycaena tityrus from different altitudes to investigate genetic variation in the melanin‐based encapsulation response. As high‐ and low‐altitude populations differ in cuticular pupal melanisation, we further tested for any associations between pupal melanisation and parasite resistance. 3. Although pupal melanisation was higher at higher compared with lower altitudes (and at a higher compared with a lower rearing temperature), any obvious relations to the encapsulation response were absent. Further phenotypic correlations within groups were significant in one out of four cases only, suggesting that in L. tityrus encapsulation operates largely independent of cuticular melanisation. 4. A significant interaction between altitude and temperature indicated that high‐altitude animals show a stronger melanisation response than low‐altitude ones at the lower temperature and vice versa, indicating local adaptation to different climates.  相似文献
