Tobacco callus fed L-tyrosine-UL?14C was sampled at 3-day intervals for 15 days, homogenized and studied with respect to distribution of incorporated radioactivity. The supernatant obtained by centrifuging of the homogenates at 270 g contained the bulk of the radioactivity although significant activity was also detected in the pellet. Sucrose density gradient centrifugation of the supernatant showed over 90% of the recovered label to be associated with a fraction designated as “less dense than mitochondria”, with the remainder being found in the fraction identified as “mitochondria”. During tissue culture, virtually all of the radioactivity in the fraction “less dense than mitochondria” was recovered in the supernatant obtained by centrifugation at 100,000 g. From 4 to 18% of the labeling in the 100,000 g supernatant fraction was attributable to tyrosine-containing protein, and the rest to free tyrosine and unidentified anionic constituents. The highest proportions of radio-activity in the 270 g pellet were associated with substances extractable with NaCl, pronase, 4.6 N NaOH, and acetolyzing reagent. Low but substantial labeling characterized the extracts obtained with Triton X-100 and 1 N NaOH. The final unextractable residue contained 20% of the 270 g pellet radioactivity. 相似文献

Benzyladenine (BA) was applied to intact bean (Phaseolus vulgaris) leaves at different stages of their growth. Changes in the amounts of cellular constituents resulting from the different treatments were followed and compared. RNA, protein, and chlorophyll contents, dry weight, fresh weight, and leaf area per single leaf continued increasing when leaves were treated with BA from an early stage, whereas in untreated leaves all these values levelled off or declined with advancing age. Besides these changes, BA treatment induced an increase in the DNA content. Changes in RNA content was more remarkable in response to application or deprival of BA treatment than the corresponding ones in protein and chlorophyll contents. The pattern of response to BA varied greatly according to the age at which the leaf received the treatment. As leaves aged, they lost the ability to increase their area and fresh weight in response to BA. Calmodulin activity was detected and assayed in barley aleuronecells. The effect of calmodulin antagonists on GA3-induced enzymesynthesis and secretion in barley aleurone layers was also investigated.These calmodulin antagonists (chlorpromazine, haloperidol) inhibitedonly GA2-induced -amylase secretion. 为要了解七星瓢虫(Coccinella septempunctata)成虫不同发育时间及不同发育部位脂肪体细胞核DNA合成的情况及激素对DNA合成的调节,我们以扫描显微分光光度法测定了脂肪体细胞核DNA的相对含量,并以该虫精细胞核作为单倍体参考标准,从而计算出脂肪体细胞核的倍体值。本文对雄成虫及不同发育阶段的雌成虫脂肪体细胞核DNA含量进行了测定,结果表明有多倍化现象发生;而且随着生殖腺的成熟,DNA 分布趋向于较高倍体。用ZR-512处理雌虫,经72小时后与对照相比脂肪体细胞核DNA合成较多,而且脏壁层脂肪体较体壁层脂肪体出现更为复杂的倍性变化,表明 ZR-512对DNA合成有促进作用。 相似文献

Melittin, which is known to stimulate phospholipase A , in many cells, caused as much elevation of fertilization membranes and increase in respiration of unfertilized eggs of the sea urchins Anthocidaris crassispina and Hemicentrotus pulcherrimus as normal fertilization.
In melittin-activated eggs, amino acid transport was decreased to less than that of unfertilized eggs, nucleoside transport was only slightly, activated, protein synthesis was rather inhibited and neither DNA synthesis nor cleavage was observed. Unfertilized eggs of sea urchins, Hemicentrotus pulcherrimus and Pseudocentrotus depressus, were treated with 4–5% butyric acid-sea water for 40–60 sec so that they were activated partheno-genetically without visible cortical changes. When these insufficiently activated eggs were inseminated 90–120 min after butyric acid-treatment, they divided much earlier than the control eggs in the first cleavage cycle. In the present paper, it becomes clear that if eggs are put into m /2,000-m /16,000 DNP-sea water at 60 min after insufficient activation and 30 min later, returned to normal sea water and then inseminated, they still show acceleration of the first cleavage in the same degree as the eggs which are not treated with DNP, while if eggs are exposed to DNP for 30 min prior to the insufficient activation or within 60 min after the activation, they do not show any acceleration of the cleavage. From these results, it may be concluded that some preparations for cleavage acceleration which are arrested by DNP become ready in the eggs at an early period in the first cleavage cycle and these preparations cannot be cancelled by DNP-treatment once they have been completed. 相似文献

为探讨超声波辅助提取黑老虎叶总黄酮的最佳提取工艺条件及其抗氧化活性,该文以黑老虎叶为研究对象,采用超声波提取法提取黑老虎叶总黄酮,通过单因素试验研究提取时间、乙醇浓度、提取温度、料液比对黑老虎叶总黄酮提取率的影响,在单因素试验的基础上,采用正交试验优化其提取工艺条件,测试了最优条件下提取的黑老虎叶总黄酮对DPPH自由基、·OH自由基及超氧阴离子的清除能力。结果表明:黑老虎叶总黄酮超声辅助提取最佳提取条件为提取时间 35 min、乙醇浓度80%、提取温度50 ℃、料液比1:20 g·mL-1,最佳条件下提取率为4.83%。抗氧化活性测试结果显示,黑老虎叶总黄酮表现出较好的清除DPPH自由基、·OH自由基及超氧阴离子能力,其抗氧化能力为清除DPPH自由基能力>清除超氧阴离子能力>清除·OH自由基能力。在浓度为0.8 mg·mL-1时,黑老虎叶总黄酮清除DPPH自由基、·OH自由基及超氧阴离子的能力相当于同浓度下Vc的97.6%、82.1%、95.5%,黑老虎叶总黄酮是天然抗氧化剂的良好来源。上述结果为黑老虎叶活性成分的提取及开发利用提供了理论基础。 相似文献
