The endangered European relict species complex Lathyrus pannonicus shows distinct morphological variation, reflected by the number of subspecies recognized, and complicated patterns of genetic variation. The traditionally recognized subspecies appear to possess different ecological preferences and disjunct distributions, particularly in the western range of the species. In this study, L. pannonicus was investigated by the correlation of distance matrices based on phytosociological, ecological, molecular and morphological data. Ecological characteristics of selected stands of L. pannonicus throughout Europe were assessed using ‘Ellenberg values’ of all the constituent taxa in phytosociological relevés. Genetic distances were calculated using recently developed methods to analyse high degrees of intra‐individual nuclear‐encoded internal transcribed spacer variability. We found that the remarkable genetic (and morphological) diversity in L. pannonicus could not be explained solely by the fragmentation of the distributional range. Instead, patterns of morphological and genetic differentiation were a reflection of the moisture regime in the sampled stands. Two major lineages could be identified: (1) a lineage adapted to dry conditions (Ellenberg indicator F‐value ≤ 3.5) and (2) a lineage preferring moist conditions (F‐value ≥ 4.5). Although both lineages occurred in close proximity in the Pannonian area, they appeared to be reproductively isolated in general. Further data are needed to determine whether these genetically and ecologically defined lineages, or ecospecies, within the L. pannonicus species complex can be formalized as (Linnaean) species or subspecies. © 2011 The Linnean Society of London, Botanical Journal of the Linnean Society, 2011, 165 , 402–421. 相似文献

Excitotoxicity, which is mediated via glutamate receptors, is also a phenomenon of the enteric nervous system. Whether enteric glial cells (EGCs), which resemble astrocytes of the central nervous system, express glutamate receptors and hence are involved in gut excitotoxicity is not yet known. To investigate glutamate receptor subunit expression in EGCs, primary EGC cultures of the myenteric plexus were analyzed by real-time PCR and Western blotting. These studies indeed showed that in EGC cultures, mRNA of the glutamate receptor subunits NR1, NR2A/B, GluR1, GluR3, and GluR5 and the protein bands of the glutamate receptor subunits NR2A/B, GluR1, GluR3, and GluR5 could be detected. Thus, in the enteric nervous system, glutamate receptor subunits are also expressed by EGCs, indicating that these cells might be involved in gut excitotoxicity. 相似文献

Pneumocystis carinii pneumonia is an opportunistic disease usually affecting immunocompromised hosts. Diagnosis is based on the microscopical detection of microrganism on BAL, an invasive sample which requires pt compliance for collection and skilled health care personnel for examination. In order to verify the possible diagnostic utility of noninvasive specimens collected in the upper respiratory tract we examined by the Internal Transcribed Spacers (ITSs) nested PCR, 39 oropharingeal samples (garglings) collected from 20 HIVAb positive pts and from 15 healthy controls. 相似文献

Stylosanthes aff. calcicola is a formally undescribed tetraploid species from the Mexican Yucatán Peninsula, showing morphological similarities to the diploid species S. calcicola , but distinct in a number of characters. We used uni- and biparentally inherited molecular markers to infer the hybrid origin of this species in relation to known diploid species of Stylosanthes . Molecular characterization was based on length and/or DNA sequence variation of nuclear sequence-tagged site (STS) markers, the internal transcribed spacer (ITS) region of nuclear rDNA and the trnL intron of chloroplast DNA (cpDNA). Stylosanthes aff. calcicola contains a distinct cpDNA haplotype and nuclear DNA fragment, with closest relationship to the diploid species S. calcicola . In contrast, the DNA sequences of two nuclear loci reveal a closer relationship to the diploid species S. angustifolia , S. hispida , S. humilis , S. leiocarpa and S. viscosa . The majority of the STS markers showed additivity of PCR fragments in S. aff. calcicola , representing the combination of two genetically different genomes. We postulate that S. aff. calcicola is a distinct species of allotetraploid origin that appears to have originated once from hybridization between two divergent genomes, of which the maternal and paternal parent are closely related to, or derived from, a member of the lineages represented by S. calcicola and S. viscosa , respectively. © 2002 The Linnean Society of London, Botanical Journal of the Linnean Society , 140 , 1–13. 相似文献

1. Provenances of Castanea sativa from populations adapted to different climatic areas of Turkey were grown in a field trial in Italy. Carbon isotope discrimination (Δ) in leaf dry matter and in leaf soluble sugar, were measured, along with photosynthesis, stomatal conductance and mesophyll conductance, to study the variability of primary productivity and its ecological significance in European Chestnut.
2. Genetic variations were found in RuBP carboxylase, chlorophyll, leaf soluble protein and leaf thickness.
3. Carbon isotope discrimination (Δ) in leaf dry matter was greater in drought-adapted than in wet-adapted provenances. A similar variation of Δ was observed in leaf soluble carbohydrates either under watered or drought conditions. Possible environmental effects of variables such as vapour pressure difference, on the relationship between transpiration efficiency and carbon isotope discrimination are discussed, on the basis of short-term and long-term results.
4. Generally low values of Δ encountered among provenances were explained not only by low values of intercellular CO2 partial pressure but also by consistently low values of mesophyll conductance leading to reduced chloroplastic CO2 partial pressure. A decrease in mesophyll conductance was induced by water shortage. Co-ordination was found between stomatal and mesophyll conductance, with the drought-adapted provenances showing much higher mesophyll conductance than the wet-adapted provenances. Variations in mesophyll conductance were related to differences in leaf protein content.
5. Possible ecophysiological adaptive mechanisms are discussed taking into account stomatal sensitivity, modulation of photosynthetic capacity and water-use efficiency under drought conditions. 相似文献

The red squirrel ( Sciurus vulgaris ) is a well-known forest animal distributed all over Europe. Still, we are far from having a firm knowledge of the species' phylogeography. This study investigates the genetic differentiation of S. vulgaris across the species' Eurasian range, using sequence data from the mitochondrial DNA gene (D-loop, 252 base pairs, cytochrome b , 359 base pairs), and eight variable autosomal microsatellite loci genotyped for 236 individuals. The results reveal the presence of two main mitochondrial phylogroups. The first clade comprises the individuals from the region of Calabria in southern Italy, belonging to the subspecies S. v . meridionalis , while the second clade contains the remainder of the studied individuals. Bayesian analysis of microsatellite genotypes resulted in three main clusterings corresponding to the three S. vulgaris subspecies: infuscatus, meridionalis and fuscoater . Geographical distribution of mtDNA haplotypes and mismatch analysis suggest a common refugium for the red squirrel across most of its present range from which expansion happened rather rapidly. The genotype mixing of italicus with northern populations could be a residual of postglacial expansion. The lack of mixing between the Calabrian lineage and the rest of European red squirrel haplotypes can be seen as evidence for distinct histories throughout the Pleistocene. Calabrian mtDNA probably diverged in an ice age contraction and remained isolated from the neighbouring squirrel populations until very recent times. 相似文献
