Microsatellites are continuing to increase in popularity for genetic studies. A major drawback, however, is they often need to be isolated de novo. The traditional method (shotgun cloning) generally results in thousands of recombinant clones, which are then screened by Southern hybridization. Results are typically around 2% positive for mammals, but less than 0.4% for birds. Shotgun cloning is therefore generally not an effective method for isolating microsatellites from avian genomes. Several alternative methods have been developed, but the decision as to which method to use is often arbitrary. Effects of grazing intensity on soil carbon stocks following deforestation of a Hawaiian dry tropical forest 总被引:2,自引:0,他引:2
The effects of forest-to-pasture conversion on soil carbon (C) stocks depend on a combination of climatic and management factors, but factors that relate to grazing intensity are perhaps the least understood. To understand the long-term impact of grazing in converted pastures, methods are needed that accurately measure the impact of grazing on recent plant inputs to soil C in a variety of pasture management and climate settings. Here, we present an analysis from Hawai'i of changes in vegetation structure and soil organic carbon (SOC) along gradients of grazing intensity and elevation in pastures converted from dry tropical forest 100 years ago. We used hyperspectral remote sensing of photosynthetic vegetation, nonphotosynthetic vegetation (NPV) and exposed substrate to understand the effects of grazing on plant litter cover, thus, estimating recent plant inputs to soils (the NPV component). Forest-to-pasture conversion caused a shift from C3 to C4 plant physiology, thus the δ 13 C method was used in soil cores to measure the fraction of SOC accumulated from pasture vegetation sources following land conversion. SOC decreased in pasture by 5–9 kg C m−2 , depending upon grazing intensity. SOC derived from C3 (forest) sources was constant across the grazing gradient, indicating that the observed variation in SOC was attributable to changes in C inputs following deforestation. Soil C stocks were also reduced in pastures relative to forest soils. We found that long-term grazing lowers SOC following Hawaiian forest-to-pasture conversion, and that these changes are larger in magnitude that those occurring with elevation (climate). Further we demonstrate a relationship between remotely sensed measurements of surface litter and field SOC measurements, allowing for regional analysis of pasture condition and C storage where limited field data are available. 相似文献

Radiocarbon signatures (Δ14C) of carbon dioxide (CO2) provide a measure of the age of C being decomposed by microbes or respired by living plants. Over a 2‐year period, we measured Δ14C of soil respiration and soil CO2 in boreal forest sites in Canada, which varied primarily in the amount of time since the last stand‐replacing fire. Comparing bulk respiration Δ14C with Δ14C of CO2 evolved in incubations of heterotrophic (decomposing organic horizons) and autotrophic (root and moss) components allowed us to estimate the relative contributions of O horizon decomposition vs. plant sources. Although soil respiration fluxes did not vary greatly, differences in Δ14C of respired CO2 indicated marked variation in respiration sources in space and time. The 14C signature of respired CO2 respired from O horizon decomposition depended on the age of C substrates. These varied with time since fire, but consistently had Δ14C greater (averaging ~120‰) than autotrophic respiration. The Δ14C of autotrophically respired CO2 in young stands equaled those expected for recent photosynthetic products (70‰ in 2003, 64‰ in 2004). CO2 respired by black spruce roots in stands >40 years old had Δ14C up to 30‰ higher than recent photosynthates, indicating a significant contribution of C stored at least several years in plants. Decomposition of O horizon organic matter made up 20% or less of soil respiration in the younger (<40 years since fire) stands, increasing to ~50% in mature stands. This is a minimum for total heterotrophic contribution, since mineral soil CO2 had Δ14C close to or less than those we have assigned to autotrophic respiration. Decomposition of old organic matter in mineral soils clearly contributed to soil respiration in younger stands in 2003, a very dry year, when Δ14C of soil respiration in younger successional stands dropped below those of the atmospheric CO2. 相似文献

The first scolopocryptopid centipede known from the fossil record is a specimen of the subfamily Scolopocryptopinae in Miocene amber from Chiapas, southern Mexico. It is described here as Scolopocryptops simojovelensis sp. nov. , displaying a distinct combination of morphological characters compared to extant congeners. Anatomical details of the fossil specimen were acquired by non‐invasive 3D synchrotron microtomography using X‐ray phase contrast. The phylogenetic position of the new species is inferred based on a combination of morphological data with sequences for six genes (nuclear 18S and 28S rRNA, nuclear protein‐coding histone H3, and mitochondrial 12S rRNA, 16S rRNA, and protein‐coding cytochrome c oxidase subunit I) for extant Scolopendromorpha. The data set includes eight extant species of Scolopocryptops and Dinocryptops from North America, east Asia, and the Pacific, rooted with novel sequence data for other blind scolopendromorphs. The molecular and combined data sets, analysed in a parsimony/direct optimization framework, identified a stable pattern of two main clades within Scolopocryptopinae. North American and Asian species of Scolopocryptops are united as a clade supported by both morphological and molecular characters. Its sister group is a Neotropical clade in which the type species of Dinocryptops is nested within a paraphyletic assemblage of Scolopocryptops species; Dinocryptops is placed in synonymy with Scolopocryptops. The strength of support for the relationships of extant taxa from the molecular data allow the Chiapas fossil to be assigned with precision, despite ambiguity in the morphological data; the fossil is resolved as sister species to the extant Laurasian clade. © 2012 The Linnean Society of London, Zoological Journal of the Linnean Society, 2012, 166 , 768–786. 相似文献

1. Availability of suitable habitat is a prerequisite for species reintroduction success, and to ensure population persistence, investigations of a species’ habitat utilisation throughout its life history should be conducted as part of a feasibility study. 2. Habitat utilisation models for burbot, Lota lota, developed using data from field studies conducted in France and Germany and information from the literature were used to assess the feasibility of reintroducing burbot into rivers of its former native range in eastern England. 3. Per cent tree roots, aquatic vegetation and flow types were important predictors of adult burbot abundance. Furthermore, the habitat utilisation models were supplemented with information from the literature, which suggested that off‐channel habitat such as wetlands and backwaters is important for spawning and nursery stages. 4. An assessment of the habitat availability in the rivers of the burbot’s former native range using variables related to spawning and nursery and adult life stages showed that although adult habitat was widely distributed, the availability of spawning and nursery habitat was less abundant, potentially limiting successful reestablishment. 5. Potential suitable habitat was concentrated in the central and southern areas of the species’ former English distribution. Overall, rivers of the burbot’s former range potentially afford suitable habitat to sustain a reintroduced population. Abstract Ferns, bryophytes and lichens are the most diverse groups of plants in wet forests in south‐eastern Australia. However, management of this diversity is limited by a lack of ecological knowledge of these groups and the difficulty in identifying species for non‐experts. These problems may be alleviated by the identification and characterization of suitable proxies for this diversity. Epiphytic substrates are potential proxies. To evaluate the significance of some epiphytic substrates, fern and bryophyte assemblages on a common tree‐fern species, Dicksonia antarctica (soft tree‐fern), were compared with those on a rare species, Cyathea cunninghamii (slender tree‐fern), in eastern Tasmania, Australia. A total of 97 fern and bryophyte species were recorded on D. antarctica from 120 trunks at 10 sites, and 64 species on C. cunninghamii from 39 trunks at four of these sites. The trunks of C. cunninghamii generally supported fewer species than D. antarctica, but two mosses (particularly Hymenodon pilifer) and one liverwort showed significant associations with this host. Several other bryophytes and epiphytic ferns showed an affinity for the trunks of D. antarctica. Species assemblages differed significantly between both sites and hosts, and the differences between hosts varied significantly among sites. The exceptionally high epiphytic diversity associated with D. antarctica suggests that it plays an important ecological role in Tasmanian forests. Evidently C. cunninghamii also supports a diverse suite of epiphytes, including at least one specialist species. 相似文献

The search for the genetic architecture of schizophrenia has employed multiple, often converging strategies. One such strategy entails the use of tracing the heritability and neurobiology of endophenotypes. Endophenotypes are quantifiable traits not visible to the eye, which are thought to reflect an intermediate place on the path from genes to disorder. Endophenotype abnormalities in domains such as neurophysiology or neurocognition occur in schizophrenia patients as well as their clinically “unaffected” relatives, and reflect polymorphisms in the DNA of schizophrenia spectrum subjects which create vulnerability to developing schizophrenia. By identifying the single nucleotide polymorphisms (SNPs) associated with endophenotypes in schizophrenia, psychiatric neuroscientists can select new strong inference based molecular targets for the treatment of schizophrenia. 相似文献
